3Biomarker and genomic analyses reveal molecular signatures of non-cardioembolic ischemic stroke显示文摘Acute ischemic stroke(AIS)is a major cause of disability and mortality worldwide.Non-cardioembolic ischemic stroke(NCIS),which constitutes the majority of AIS cases,is highly heterogeneous,thus requiring precision medicine treatments.This study aimed to investigate the molecular mechanisms underlying NCIS heterogeneity.We integrated data from the Third China National Stroke Registry,including clinical phenotypes,biomarkers,and whole-genome sequencing data for 7695 patients with NCIS.We identified 30 molecular clusters based on 63 biomarkers and explored the comprehensive landscape of biological heterogeneity and subpopulations in NCIS.Dimensionality reduction revealed fine-scale subpopulation structures associated with specific biomarkers.The subpopulations with biomarkers for inflammation,abnormal liver and kidney function,homocysteine metabolism,lipid metabolism,and gut microbiota metabolism were associated with a high risk of unfavorable clinical outcomes,including stroke recurrence,disability,and mortality.Several genes encoding potential drug targets were identified as putative causal genes that drive the clusters,such as CDK10,ERCC3,and CHEK2.We comprehensively characterized the genetic architecture of these subpopulations,identified their molecular signatures,and revealed the potential of the polybiomarkers and polygenic prediction for assessing clinical outcomes.Our study demonstrates the power of large-scale molecular biomarkers and genomics to understand the underlying biological mechanisms of and advance precision medicine for NCIS.Lingling Ding Yu Liu Xia Meng Yong Jiang Jinxi Lin Si Cheng Zhe Xu Xingquan Zhao Hao Li Yongjun Wang Zixiao Li 2023Signal Transduction and Targeted Therapy2023,8,6:0
4Gauze:enabling communication-friendly block synchronization with cuckoo filter显示文摘Block synchronization is an essential component of blockchain systems.Traditionally,blockchain systems tend to send all the transactions from one node to another for synchronization.However,such a method may lead to an extremely high network bandwidth overhead and significant transmission latency.It is crucial to speed up such a block synchronization process and save bandwidth consumption.A feasible solution is to reduce the amount of data transmission in the block synchronization process between any pair of peers.However,existing methods based on the Bloom filter or its variants still suffer from multiple roundtrips of communications and significant synchronization delay.In this paper,we propose a novel protocol named Gauze for fast block synchronization.It utilizes the Cuckoo filter(CF)to discern the transactions in the receiver’s mempool and the block to verify,providing an efficient solution to the problem of set reconciliation in the P2P(Peer-to-Peer Network)network.By up to two rounds of exchanging and querying the CFs,the sending node can acknowledge whether the transactions in a block are contained by the receiver’s mempool or not.Based on this message,the sender only needs to transfer the missed transactions to the receiver,which speeds up the block synchronization and saves precious bandwidth resources.The evaluation results show that Gauze outperforms existing methods in terms of the average processing latency(about lower than Graphene)and the total synchronization space cost(about lower than Compact Blocks)in different scenarios.Xiaoqiang DING Liushun ZHAO Lailong LUO Junjie XIE Deke GUO Jinxi LI 2023Frontiers of Computer Science2023,17,3:0
5Analysis on the Safety of Expressway Interchange显示文摘Currently, expressways encourage the use of interchange design methods in the layout mode to improve the safety of expressway operation in order to relieve road transportation pressure and strengthen the prevention of traffic safety accidents. The advantages and characteristics of the interchange method are more obvious and have important application value when compared to other interchange methods.
