1Oncogenic AURKA-enhanced N6-methyladenosine modification increases DROSHA mRNA stability to transactivate STC1 in breast cancer stem-like cells显示文摘RNase III DROSHA is upregulated in multiple cancers and contributes to tumor progression by hitherto unclear mechanisms.Here,we demonstrate that DROSHA interacts withβ-Catenin to transactivate STC1 in an RNA cleavage-independent manner,contributing to breast cancer stem-like cell(BCSC)properties.DROSHA mRNA stability is enhanced by N6-methyladenosine(m^(6)A)modification which is activated by AURKA in BCSCs.AURKA stabilizes METTL14 by inhibiting its ubiquitylation and degradation to promote DROSHA mRNA methylation.Moreover,binding of AURKA to DROSHA transcript further strengthens the binding of the m^(6)A reader IGF2BP2 to stabilize m^(6)A-modified DROSHA.In addition,wild-type DROSHA,but not an m^(6)A methylation-deficient mutant,enhances BCSC stemness maintenance,while inhibition of DROSHA m^(6)A modification attenuates BCSC traits.Our study unveils the AURKA-induced oncogenic m^(6)A modification as a key regulator of DROSHA in breast cancer and identifies a novel DROSHA transcriptional function in promoting the BCSC phenotype.Fei Peng Jie Xu Bai Cui Qilan Liang Sai Zeng Bin He Hong Zou Manman Li Huan Zhao Yuting Meng Jin Chen Bing Liu Shasha Lv Peng Chu Fan An Zifeng Wang Junxiu Huang Yajing Zhan Yuwei Liao Jinxin Lu Lingzhi Xu Jin Zhang Zhaolin Su Zhiguang Li Fangjun Wang Eric W-FLam Quentin Liu 2021Cell Research2021,31,3:9
2Monitoring Contact State of Laboratory Fault with Coda Transmission Waves显示文摘We monitored the amplitude changes of coda transmission waves around 500 kHz across the frictional interface of a simulated 1. 5-meter-long fault during normal stress holding test.We find that the amplitude of coda transmission waves increases with the logarithm of stationary contact time. Localized increase amounted to a level ranging from 4% to 16%along the fault is observed during the 1-hour experiment. We discuss that the frictional strength at mesoscopic scale,which is related to the amplitude of coda transmission waves,is responsible for the phenomenon. 6Joint Bandwidth Allocation and Path Selection in WANs with Path Cardinality Constraints显示文摘In this paper,we study the joint bandwidth allocation and path selection problem,which is an extension of the well-known network utility maximization(NUM)problem,via solving a multi-objective minimization problem under path cardinality constraints.Specifically,such a problem formulation captures various types of objectives including proportional fairness,average delay,as well as load balancing.In addition,in order to handle the'unsplittable flows',path cardinality constraints are added,making the resulting optimization problem quite challenging to solve due to intrinsic nonsmoothness and nonconvexity.Almost all existing works deal with such a problem using relaxation techniques to transform it into a convex optimization problem.However,we provide a novel solution framework based on the linearized alternating direction method of multipliers(LADMM)to split the original problem with coupling terms into several subproblems.We then derive that these subproblems,albeit nonconvex nonsmooth,are actually simple to solve and easy to implement,which can be of independent interest.Under some mild assumptions,we prove that any limiting point of the generated sequence of the proposed algorithm is a stationary point.Numerical simulations are performed to demonstrate the advantages of our proposed algorithm compared with various baselines.Jinxin Wang Fan Zhang Zhonglin Xie Zaiwen Wen Gong Zhang 2021Journal of Communications and Information Networks2021,6,3:0
7Characteristics of acoustic response from simulated impulsive lightning current discharge显示文摘Acoustic observation of laboratory simulated lightning discharge can greatly avoid the observation difficulties under natural thunderstorm environment.An acoustic observation experiment based on the impulsive current generation system and a microphone array was carried out and illustrated in this study.A quantitative study on characteristics of response sound pressure initiated from simulated lightning current was performed.The first arrived acoustic N-waves measured at different distances initiated from different discharge amplitudes were compared and analysed.The acoustic amplitude,rise time and duration time defined for N-waves and peak frequencies were found to have an obvious correlation with the discharging amplitudes of simulated lightning currents and the observing distances.The linear change of acoustic amplitude is more obvious than that of the other parameters with the changed discharging amplitude or observing distance.A certain degree of shape change of the simulated lightning current except for the amplitude may not significantly affect the response acoustic characteristics when comparing the first acoustic N-waves from 8/20μs lightning current and another kind of impulsive current which has a wider shape and two subsequent peaks.The subsequent current peaks in short time delays were proved to successively generate acoustic N-waves independently without overlapping,which meanwhile,indicated a consistent linear relationship of amplitude and time delay between electrical and acoustic pulses.Jianguo Wang Jinxin Cao Li Cai Yadong Fan Mi Zhou Quanxin Li 2019High Voltage2019,4,3:0

9Characteristics of the AE Distribution and Triggering Mechanism of a Simulated Meter-Scale Fault after Stick-Slip Events显示文摘With the more complete acoustic emission(AE)catalog improved by the multi-channel AE matched-filter technique(MFT),we study the spatiotemporal evolution of the AE activities after laboratory stick-slip events incorporate with the slip data recorded by displacement transducers on an^1.5 m granite fault.The results show that the number of the AE events identified by MFT is about 9 times larger than that of the traditional method.A logarithmic expansion of early AE events along the fault strike is observed as a function of time,whereas the fault does not slip in the same manner.Thus,we related the expansion of the early AE events along the fault to the stress transfer caused by the adjacent AE events.Moreover,there is a good correlation between the cumulative number of the later AE events and the amount of fault slip.It suggests that the stress change caused by the continuous slip of the simulated fault after the stick-slip events response for the later AE events near or on the simulated fault.HOU Jinxin XIE Fan REN Yaqiong WANG Baoshan 2020Earthquake Research in China2020,34,3:0
10NLRP3 inflammasome signal pathway involves in Vibrio harveyi-induced inflammatory response in murine peritoneal macrophages in vitro显示文摘Vibrio harveyi,an important zoonotic pathogen,can infect wounds and cause inflammatory response.Understanding the inflammatory response pathways could facilitate the exploration of molecular mechanisms for treating V.harveyi infection.NLR family pyrin domain-containing 3(NLRP3)inflammasome is involved in the interaction between hosts&pathogenic microorganisms&could be sensed by various pathogen-associated molecular patterns(PAMPs)or damage-associated molecular patterns(DAMPs).Nonetheless,the function of NLRP3 inflammasome in V.harveyi infection remains unclear.In the present study,we established a V.harveyi infection model using murine peritoneal macrophages(PMs).Various techniques,including western blot analysis,enzyme-linked immunosorbent assay(ELISA),RT-qPCR,immunofluorescence,&inhibition assays,were used to explore the molecular mechanism of V.harveyi-induced inflammation.The results showed that many inflammatory cytokines participated in V.harveyi infection,with interleukin(IL)-1βbeing the most abundant.Pan-caspase inhibitor pretreatment significantly decreased the secretion of IL-1βin murine PMs.Moreover,the identification of V.harveyi involved a large number of NLR molecules,especially the NLRP3 receptor,&further studies revealed that NLPR3 inflammasome was activated by V.harveyi infection,as evidenced by puncta-like NLRP3 surrounding cell nuclear,ASC specks in the nucleus&cytoplasm,&ASC oligomerization.Inhibition of NLRP3 inflammasome impaired the release of mature IL-1βin V.harveyi-infected murine PMs.Furthermore,blocking the secretion of mature IL-1βcould markedly decrease the release of other proinflammatory cytokines,including IL-6,IL-12,&tumor necrosis factor-α.Overall,these data indicated that NLRP3 inflammasome was activated in response to V.harveyi infection and enhanced inflammatory response by promoting IL-1βsecretion in murine PMs.Guili Yu Jinxin Wang Wei Zhang Qiankun Yang Gang Liu Lei Wang Babatunde Kazeem Bello Xiao Zhang Tianmeng Zhang Hui Fan Panpan Zhao Wei Liang Jingquan Dong 2021Acta Biochimica et Biophysica Sinica2021,53,12:0
