1Calreticulin and cancer显示文摘Calreticulin(CALR)is an endoplasmic reticulum(ER)-resident protein involved in a spectrum of cellular processes.In healthy cells,CALR operates as a chaperone and Ca^(2+)buffer to assist correct protein folding within the ER.Besides favoring the maintenance of cellular proteostasis,these cell-iintrinsic CALR functions support Ca^(2+)-dependent processes,such as adhesion and integrin signaling,and ensure normal antigen presentation on MHC Class I molecules.Moreover,cancer cells succumbing to immunogenic cell death(ICD)expose CALR on their surface,which promotes the uptake of cell corpses by professional phagocytes and ultimately supports the initiation of anticancer immunity.Thus,loss-of-function CALR mutations promote oncogenesis not only as they impair cellular homeostasis in healthy cells,but also as they compromise natural and therapy-driven immunosurveillanee.However,the prognostic impact of total or membrane-exposed CALR levels appears to vary considerably with cancer type.For instance,while genetic CALR defects promote pre-neoplastic myeloproliferation,patients with myeloproliferative neoplasms bearing CALR mutations often experience improved overall survival as compared to patients bearing wild-type CALR.Here,we discuss the context-dependent impact of CALR on malignant transformation,tumor progression and response to cancer therapy.Jitka Fucikova Radek Spisek Guido Kroemerc Lorenzo Galluzzi 2021Cell Research2021,31,1:12

3Helicobacter pylori infection and other bacteria in pancreatic cancer and autoimmune pancreatitis显示文摘Helicobacter pylori(H.pylori)is an infectious agent influencing as much as 50%of the world’s population.It is the causative agent for several diseases,most especially gastric and duodenal peptic ulcer,gastric adenocarcinoma and mucosaassociated lymphoid tissue lymphoma of the stomach.A number of other,extragastric manifestations also are associated with H.pylori infection.These include neurological disorders,such as Alzheimer’s disease,demyelinating multiple sclerosis and Parkinson’s disease.There is also evidence for a relationship between H.pylori infection and such dermatological diseases as psoriasis and rosacea as well as a connection with infection and open-angle glaucoma.Generally little is known about the relationship between H.pylori infection and diseases of the pancreas.Most evidence about H.pylori and its potential role in the development of pancreatic diseases concerns pancreatic adenocarcinoma and autoimmune forms of chronic pancreatitis.There is data(albeit not fully consistent)indicating modestly increased pancreatic cancer risk in H.pylori-positive patients.The pathogenetic mechanism of this increase is not yet fully elucidated,but several theories have been proposed.Reduction of antral Dcells in H.pylori-positive patients causes a suppression of somatostatin secretion that,in turn,stimulates increased secretin secretion.That stimulates pancreatic growth and thus increases the risk of carcinogenesis.Alternatively,H.pylori,as a part of microbiome dysbiosis and the so-called oncobiome,is proven to be associated with pancreatic adenocarcinoma development via the promotion of cellular proliferation.The role of H.pylori in the inflammation characteristic of autoimmune pancreatitis seems to be explained by a mechanism of molecular mimicry among several proteins(mostly enzymes)of H.pylori and pancreatic tissue.Patients with autoimmune pancreatitis often show positivity for antibodies against H.pylori proteins.H.pylori,as a part of microbiome dysbiosis,also is viewed as a potential trigger of autoimmune inflammation of the pancreas.It is precisely these relationships(and associated equivocal conclusions)that constitute a center of attention among pancreatologists,immunologists and pathologists.In order to obtain clear and valid results,more studies on sufficiently large cohorts of patients are needed.The topic is itself sufficiently significant to draw the interest of clinicians and inspire further systematic research.Next-generation sequencing could play an important role in investigating the microbiome as a potential diagnostic and prognostic biomarker for pancreatic cancer.Lumir Kunovsky Petr Dite Petr Jabandziev Jiri Dolina Jitka Vaculova Martin Blaho Martina Bojkova Jana Dvorackova Magdalena Uvirova Zdenek Kala Jan Trna 2021World Journal of Gastrointestinal Oncology2021,13,8:5

5Antibodies Against Annexin V and Prothrombin,Their Correlation with Other Anti-phospholipid Antibodies in Recurrent Pregnancy Loss显示文摘Objective To study the findings of serum antibodies against annexin V, prothrombin,ph-inositol, ph-acid, ph-ethanolamine, ph-serine, ph-glycerol, cardiolipin, and beta2-glycoprotein I and analyze the trophoblast annexin V receptorsMethods Sera from 156 patients aged 26-41 years with recurrent pregnancy loss (3-7 times) were investigated. Eighty-four fertile healthy women aged 24-38 yearswere included in a control group. ELISA methods were used for detecting a panel of seraanti-phospholipid antibodies. Immunolocalization of annexin V receptors in 143trophoblast specimens of 156 patients was investigated by the immunofluorescencetechnique using Annexin V-FITC, Apoptosis and AnnexinV-CY3 commercial kits.Results Positivity for anti-phospholipid antibodies mainly against ph-serine, ph-ethanolamine, and ph-inositol was found together in 80.8% (126 out of 156 patients),anti-prothrombin antibodies in 12% (18), and anti-annexin V antibodies in 13. 5%(21) women. No significant levels of anti-phospholipid antibodies were found in 6controls. Placenta immunohistopathology also exhibited some changes manifested bythe presence of apoptotic and necrotic cells in trophoblast, and very fewmicrothrombotization in some intervillous spaces.Conclusion Our detailed study demonstrated the prevalence of majority of anti-phospholipid antibodies as a high risk factor for repeated reproductive failure. 6SARS-CoV-2 spike protein binds to bacterial lipopolysaccharide and boosts proinflammatory activity显示文摘There is a link between high lipopolysaccharide(LPS)levels in the blood and the metabolic syndrome,and metabolic syndrome predisposes patients to severe COVID-19.Here,we define an interaction between SARS-CoV-2 spike(S)protein and LPS,leading to aggravated inflammation in vitro and in vivo.Native gel electrophoresis demonstrated that SARS-CoV-2 S protein binds to LPS.Microscale thermophoresis yielded a of〜47 nM for the interaction.Computational modeling and all-atom molecular dynamics simulations further substantiated the experimental results,identifying a main LPS-binding site in SARS-CoV-2 S protein.S protein,when combined with low levels of LPS,boosted nuclear factor-kappa B(NF-k B)activation in monocytic THP-1 cells and cytokine responses in human blood and peripheral blood mononuclear cells,respectively.The in vitro inflammatory response was further validated by employing NF-kB reporter mice and in vivo bioimaging.Dynamic light scattering,transmission electron microscopy,and LPS-FITC analyses demonstrated that S protein modulated the aggregation state of LPS,providing a molecular explanation for the observed boosting effect.Taken together,our results provide an interesting molecular link between excessive inflammation during infection with SARS-CoV-2 and comorbidities involving increased levels of bacterial endotoxins.Ganna Petruk Manoj Puthia Jitka Petrlova Firdaus Samsudin Ann-Charlotte Stromdahl Samuel Cerps Lena Uller Sven Kjellstrom Peter J.Bond Artur Schmidtchen 2020Journal of Molecular Cell Biology2020,12,12:3
7Monitoring dioxins and PCBs in eggs as sensitive indicators for environmental pollution and global contaminated sites and recommendations for reducing and controlling releases and exposure显示文摘This review compiles information on PCDD/F-and PCB-contaminated eggs from 20 years of global egg monitoring around emission sources in four continents conducted by the International Pollutants Elimination Network(IPEN)and Arnika as well as a compilation of data from scientific literature.IPEN monitored 127 pooled egg samples including samples from 113 chicken flocks at potential PCDD/F-and PCB-contaminated sites around priority sources listed in the Stockholm Convention(e.g.waste incinerators,metal industries,cement plants,and open burning).99(88%)of pooled egg samples were above the EU maximum limits for PCDD/Fs(2.5 pg PCDD/F-TEQ/g fat)or the sum of PCDD/Fs and dioxinlike PCBs(5 pg PCDD/F-PCB-TEQ/g fat).Children consuming such eggs exceed the tolerable weekly intake(TWI).This demonstrates that close to 90%of these areas were not safe for the production of free-range eggs.Sixteen out of the 113 egg samples(14%)were contaminated above 50 pg TEQ/g fat and exceeded the EU maximum limit more than 10 times.From the 26 pooled egg samples around incinerators 24(92%)exceeded the limit with a mean of 43.1 pg TEQ/g fat(2.6e234 pg TEQ/g).All 21 egg samples around metal industries(4.4e112.6 pg TEQ/g fat)were above limits with mean concentration of 26.0 pg TEQ/g fat.Also all 7 egg samples measured at e-waste recycling sites were above limits(mean 308 pg TEQ/g fat).In 58(51%)pooled egg samples the PCB-TEQ was above 5 pg TEQ/g fat exceeding the EU maximum limit with dioxin-like PCBs alone.This highlights the role of commercial PCBs for global contamination with dioxin-like compounds.It was discovered that around metal industries,shredder plants,open burning sites of e-waste and dump sites,a high share of contamination was caused by dl-PCBs.This clearly shows severe PCB release from the end-of-life management of PCB-containing equipment in developing countries.Also highly contaminated eggs were found at many sites where plastic was incinerated.The highest contaminated egg sample ever measured came from an e-waste site in Ghana and had 856 pg TEQ/g fat plus 300 pg TEQ from brominated dioxins(PBDD/Fs).Other extreme PCDD/F contaminations of eggs were found at a chlor-alkali site(514 pg TEQ/g fat),Agent Orange contaminated areas in Vietnam(490,249 and 246 pg TEQ/g fat)and e-waste sites(568 and 520 pg TEQ/g fat).Where DR CALUX®bioassay revealed higher TEQ compared to measured PCDD/F-PCB-TEQ in IPEN studies,polybrominated PBDD/F were also measured and detected up to 300 pg TEQ/g fat at e-waste sites.One positive outcome from the IPEN studies is that all 10 pooled supermarket eggs in developing countries were below regulatory limit.Policy recommendations are made including:a systematic assessment of areas around PCDD/Fs and PCBs sources;measures for reduction of exposures of populations;urgent control of emission sources including PCB equipment,the open burning of plastic,and the use of plastic as fuel in boilers/incinerators in developing countries without air pollution control.Furthermore,soil limits need to be re-assessed and lowered for free-range poultry.Jindrich Petrlik Lee Bell Joe DiGangi Serge Molly Allo'o Allo'o Gilbert Kuepouo Griffins Ochieng Ochola Valeriya Grechko Nikola Jelinek Jitka Strakova Martin Skalsky Yuyun Ismawati Drwiega Jonathan N.Hogarh Eric Akortia Sam Adu-Kumi Akarapon Teebthaisong Maria Carcamo Bjorn Beeler Peter Behnisch Claudia Baitinger Christine Herold Roland Weber 2022Emerging Contaminants2022,8,1:2
8Large-scale permafrost degradation as a primary factor in Larix sibirica forest dieback in the Khentii massif,northern Mongolia显示文摘The objective of this study is to investigate the potential causes of widespread Larix sibirica Ledeb.mortality observed in the Khentii massif of northern Mongolia.The ratio of deadwood to living trees in affected stands in the Goricho region,the southernmost study site situated close to the Gobi Desert,was as high as 3.6:1.Moisture fluctuations monitored over 2 years using electrical impedance spectrometry revealed that the Goricho study site had higher soil moisture levels than the two less affected sites Barun Bayan and Dzun Bayan.High soil moisture was recorded in an area characterized by highly skeletal soils,ones with more than 35%by volume of rock fragments,and comparatively shallow soil horizons,from valley to mountains.The layer of permafrost influencing hydrogeological processes is much deeper in the Goricho region compared to the undisturbed study sites.Redundancy analysis confirmed a significant number of dead L.sibirica on sites with developed soils.Live forest stands,however damaged,grow in this region on well-drained scree slopes or on rocky bastions.The mass mortality observed for L.sibirica may be directly linked to accelerated permafrost thaw in the area bordered by the Tuul and the Terelj Rivers.Our assumption is that L.sibirica root system necrosis occurred as a result of long-term waterlogging of developed soils with high spatial heterogeneity,normally able to absorb high quantities of groundwater.The areas unaffected were scree fields and rocky bastions characterized by adequate drainage.All of our findings support the primary stages of large-scale permafrost thaw,i.e.,correlating increases in soil moisture with increasing permafrost active layer thickness.David Juricka Jitka Novotna Jakub Houska Jana Parilkova Jan Hladky Vaclav Pecina Hana Cihlarova Marcela Burnog Jakub Elbl Zdena Rosicka Martin Brtnicky Jindrich Kynicky 2020Journal of Forestry Research2020,31,1:2
