1Biomimetic polycaprolactone-chitosan nanofibrous substrate influenced cell cycle and ECM secretion affect cellular uptake of nanoclusters显示文摘Biomimetic cell culture substrates are developed as an alternative to the conventional substrates.They provide necessary biochemical and biophysical cues to the cells from their surrounding environment for their optimal growth,behaviour and physiology.Changes in physiology of cells growing on biomimetic substrate can essentially affect results of in vitro biological experiments such as drug cytotoxicity,nanoparticle internalization or signalling pathways.As majority of ECM proteins are fibrous in nature,nanofibrous scaffolds have more biomimicking properties.Therefore,in this study,we developed ECM mimicking polycaprolactone-chitosan nanofiber substrate and evaluated its effect on cell morphology,proliferation,cell cycle and ECM production.Further,cellular uptake of BSA-AuNCs has been assessed on conventional and biomimetic substrate in order to demonstrate the effect of these events on cellular properties.It was observed that the cells that were grown for 15 days on the nanofibers,had majority of cells in the proliferative phase of cell cycle compared to TCPS.Moreover,these cells showed extensive collagen and fibronectin production.Due to these conditions C3H10T1/2 cells displayed higher cell internalization of BSA-AuNCs.Overall,this study indicates that the nano-topographical and biochemical environment could alter the cell proliferative behaviour and ECM production,which affects the cell internalization of BSA-AuNCs.Also,PCL-chitosan nanofibrous substrate could be a better alternative to TCPS for cell culture studies.Dhwani Jhala Hilal Rather Dhaval Kedaria Juhi Shah Sanjay Singh Rajesh Vasita 2019Bioactive Materials2019,4,1:4
2Development of cost effective personnel armour through structural hybridization显示文摘The objective of the present study is to develop cost effective thermoplastic hybrid laminate using Dyneema®HB50 and Tensylon®HSBD 30A through structural hybridization method.Laminates having 20 mm thickness were fabricated and subjected to 7.6239 mm mild steel core projectile with an impact velocity of 730±10 ms1.Parameters such as energy absorption,back face deformation and rate of back face deformation were measured as a function of hybridization ratio.It was observed that hybrid laminate with 50:50 ratio(w/w)of Tensylon®and Dyneema®with Tensylon®as front face showed 200%more energy absorption when compared to 100%Tensylon®laminate and showed equal energy absorption as that of expensive 100%Dyneema®laminate.Moreover,hybrid laminate with TD50:50 ratio showed 40%lower in terms of final back face deformation than Dyneema®laminate.Rate of back face deformation was also found to be slow for hybrid laminate as compared to Dyneema®laminate.Dynamic mechanical analysis showed that,Tensylon®laminate has got higher stiffness and lower damping factor than Dyneema®and hybrid laminates.The interface between Tensylon®and Dyneema®layers was found to be separating during the penetration process due to the poor interfacial bonding.Failure behaviour of laminates for different hybridization ratios were studied by sectioning the impacted laminates.It was observed that,the Tensylon®laminate has undergone shear cutting of fibers as major failure mode whereas the hybrid laminate showed shear cutting followed by tensile stretching,fiber pull out and delamination.These inputs are highly useful for body armour applications to design cost effective armour with enhanced performance.P.Rama Subba Reddy T.Sreekantha Reddy I.Srikanth Juhi Kushwaha V.Madhu 2020Defence Technology(防务技术)2020,16,6:2
3Dengue:epidemiology,prevention and pressing need for vaccine development显示文摘Dengue fever is a mosquito born viral infection,and the complicated form of dengue is dengue hemorrhagic fever(DHF).In the recent decades incidence and distribution of dengue has increased dramatically.Dengue viruses belong to family flaviviridae with four serotypes and are transmitted mainly by mosquito Aedes aegypti.Today almost two-fifth of world’s population (2.5 million) is at risk of dengue and no specific antiviral drug or vaccine is available against it.Uncontrolled population growth in Africa and South East Asia has increased number of susceptible hosts in urban and semi urban areas.About 40%of world population resides in the high risk area for dengue transmission.According to latest estimates by WHO,yearly 50 to 100 million infections occur globally,this includes around 500 000 DHF and 22 000 deaths,mostly among children.Only symptomatic treatment in the form of analgesic,antipyretics and body fluid management is provided to the patient.Prevention strategies mainly focus on two approaches, firsdy on activities to control vector and secondly on activities to protect human from mosquito bite but there is always concerns regarding their sustainably and effectiveness.Theoretically development of an effective dengue vaccine is feasible and production of an effective and affordable vaccine could be a viable option to save humans from this dreadful disease. 4Ipsilateral breast tumor recurrence in early stage breast cancer patients treated with breast conserving surgery and adjuvant radiation therapy: Concordance of biomarkers and tumor location from primary tumor to in-breast tumor recurrence显示文摘BACKGROUND Patients with an in-breast tumor recurrence(IBTR)after breast-conserving therapy have a high risk of distant metastasis and disease-related mortality.Classifying clinical parameters that increase risk for recurrence after IBTR remains a challenge.AIM To describe primary and recurrent tumor characteristics in patients who experience an IBTR and understand the relationship between these characteristics and disease outcomes.METHODS Patients with stage 0-II breast cancer treated with lumpectomy and adjuvant radiation were identified from institutional databases of patients treated from 2003-2017 at our institution.Overall survival(OS),disease-free survival,and local recurrence-free survival(LRFS)were estimated using the Kaplan Meier method.We identified patients who experienced an isolated IBTR.Concordance of hormone receptor status and location of tumor from primary to recurrence was evaluated.The effect of clinical and treatment parameters on disease outcomes was also evaluated.RESULTS We identified 2164 patients who met the eligibility criteria.The median follow-up for all patients was 3.73[interquartile range(IQR)2.27-6.07]years.Five-year OS was 97.7%(95%CI:96.8%-98.6%)with 28 deaths;5-year LRFS was 98.0%(97.2-98.8)with 31 IBTRs.We identified 37 patients with isolated IBTR,19(51.4%)as ductal carcinoma in situ and 18(48.6%)as invasive disease,of whom 83.3%had an in situ component.Median time from initial diagnosis to IBTR was 1.97(IQR:1.03-3.5)years.Radiotherapy information was available for 30 of 37 patients.Median whole-breast dose was 40.5 Gy and 23 patients received a boost to the tumor bed.Twenty-five of thirty-two(78.1%)patients had concordant hormone receptor status,HER-2 receptor status,and estrogen receptor(ER)(P=0.006)and progesterone receptor(PR)(P=0.001)status from primary to IBTR were significantly associated.There were no observed changes in HER-2 status from primary to IBTR.The concordance between quadrant of primary to IBTR was 10/19[(62.2%),P=0.008].Tumor size greater than 1.5 cm(HR=0.44,95%CI:0.22-0.90,P=0.02)and use of endocrine therapy upfront(HR=0.36,95%CI:0.18-0.73,P=0.004)decreased the risk of IBTR.CONCLUSION Among patients with early stage breast cancer who had breast conserving surgery treated with adjuvant RT,ER/PR status and quadrant were highly concordant from primary to IBTR.Tumor size greater than 1.5 cm and use of adjuvant endocrine therapy were significantly associated with decreased risk of IBTR.Juhi M Purswani Fauzia Shaikh S Peter Wu Jennifer Chun Kim Freya Schnabel Nelly Huppert Carmen A Perez Naamit K Gerber 2020World Journal of Clinical Oncology2020,11,1:2
