1Screening beneficial bacteriostatic lactic acid bacteria in the intestine and studies of bacteriostatic substances显示文摘Lactic acid bacteria(LAB)are a representative probiotic.As the dominant flora in the human intestinal tract,LAB can regulate the balance of human intestinal flora and improve host health.The purpose of this study was to isolate and screen LAB that are well suited to the intestinal characteristics of the Chinese population,with excellent probiotics and high antibacterial activity.After 16S ribosomal RNA(rRNA)homology and phylogenetic tree analysis,potential probiotics were tested for their antibacterial activity,resistance to artificial gastrointestinal fluid and drugs,surface hydrophobicity,and safety.Three strains of LAB with acid resistance,bile salt resistance,epithelial cell adhesion,and no multidrug resistance were selected:Lactobacillus salivarius,Leuconostoc lactis,and Lactobacillus paracasei.Analysis of the antibacterial active substances in the three strains and their fermentation broths revealed that the main antibacterial substances of L.lactis were organic acids,whereas those of L.salivarius and L.paracasei were organic acids and bacteriocins with broad-spectrum antibacterial activity.These three strains of probiotic LAB with high antibacterial activity were identified as bacterial resources that could potentially be used to develop probiotic preparations for the prevention and treatment of intestinal diseases caused by intestinal pathogens.Zhijing LIU Cong XU Ran TIAN Wan WANG Jiage MA Liya GU Fei LIU Zhanmei JIANG Juncai HOU 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,7:10

11Relatively rapid evolution rates of SARS-CoV-2 spike gene at the primary stage of massive vaccination显示文摘A series of stringent non-pharmacological and pharmacological interventions were implemented to contain the pandemic but the pandemic continues.Moreover,vaccination breakthrough infection and reinfection in convalescent coronavirus disease 2019(COVID-19)cases have been reported.Further,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)variants emerged with mutations in spike(S)gene,the target of most current vaccines.Importantly,the mutations exhibit a trend of immune escape from the vaccination.Herein the scientific question that if the vaccination drives genetic or antigenic drifts of SARS-CoV-2 remains elusive.We performed correlation analyses to uncover the impacts of wide vaccination on epidemiological characteristics of COVID-19.In addition,we investigated the evolutionary dynamics and genetic diversity of SARS-CoV-2 under immune pressure by utilizing the Bayesian phylodynamic inferences and the lineage entropy calculation respectively.We found that vaccination coverage was negatively related to the infections,severe cases,and deaths of COVID-19 respectively.With the increasing vaccination coverage,the lineage diversity of SARS-CoV-2 dampened,but the rapid mutation rates of the S gene were identified,and the vaccination could be one of the explanations for driving mutations in S gene.Moreover,new epidemics resurged in several countries with high vaccination coverage,questioning their current pandemic control strategies.Hence,integrated vaccination and non-pharmacological interventions are critical to control the pandemic.Furthermore,novel vaccine preparation should enhance its capabilities to curb both disease severity and infection possibility.Jing Yang Min Han Liang Wang Likui Wang Tianrui Xu Linhuan Wu Juncai Ma Gary Wong Wenjun Liu George F.Gao Yuhai Bi 2022Biosafety and Health2022,4,4:0
12Functions of Nitrogen,Phosphorus and Potassium in Energy Status and Their Influences on Rice Growth and Development显示文摘Nitrogen(N),phosphorus(P)and potassium(K)are important essential nutrients for plant growth and development,but their functions in energy status remains unclear.Here,we grew Nipponbare rice seedlings in a growth chamber for 20 d at 30℃/24℃day/night)under natural sunlight conditions with different nutrient regimes.The results showed that N had the strongest influence on the plant growth and development,followed by P and K.The highest nonstructural carbohydrate content,dry matter weight,net photosynthetic rate(Pn),ATP content,as well as NADH dehydrogenase,cytochrome oxidase and ATPase activities were found in the plants that received sufficient N,P and K.The lowest values of these parameters were detected in the N-deficient plants.Higher dry matter accumulation was observed in the K-deficient than in the P-deficient treatments,but there was no significant difference in the ratio of respiration rate to Pn between these two treatments,suggesting that differences in energy production efficiency may have accounted for this result.This hypothesis was confirmed by higher ATP contents and activities of NADH dehydrogenase,cytochrome oxidase and ATPase in the K-deficient plants than in the P-deficient plants.We therefore inferred different abilities in energy production efficiency among N,P and K in rice seedlings,which determined rice plant growth and development.MA Jiaying CHEN Tingting LIN Jie FU Weimeng FENG Baohua LI Guangyan LI Hubo LI Juncai WU Zhihai TAO Longxing FU Guanfu 2022Rice science2022,29,2:0
13Three-dimensional reconstruction and comparison of vacuolar membranes in response to viral infection显示文摘The vacuole is a unique plant organelle that plays an important role in maintaining cellular homeostasis under various environmental stress conditions. However, the effects of biotic stress on vacuole structure has not been examined using three-dimensional(3D) visualization. Here, we performed 3D electron tomography to compare the ultrastructural changes in the vacuole during infection with different viruses. The 3D models revealed that vacuoles are remodeled in cells infected with cucumber mosaic virus(CMV) or tobacco necrosis virus A Chinese isolate(TNV-AC), resulting in the formation of spherules at the periphery of the vacuole. These spherules contain neck-like channels that connect their interior with the cytosol. Confocal microscopy of CMV replication proteins 1 a and 2 a and TNV-AC auxiliary replication protein p23 showed that all of these proteins localize to the tonoplast.Electron microscopy revealed that the expression of these replication proteins alone is sufficient to induce spherule formation on the tonoplast, suggesting that these proteins play prominent roles in inducing vacuolar membrane remodeling. This is the first report of the 3D structures of viral replication factories built on the tonoplasts.
