3Comparative functional genomics identifies an iron-limited bottleneck in a Saccharomyces cerevisiae strain with a cytosolic-localized isobutanol pathway显示文摘Metabolic engineering strategies have been successfully implemented to improve the production of isobutanol,a next-generation biofuel,in Saccharomyces cerevisiae.Here,we explore how two of these strategies,pathway re-localization and redox cofactor-balancing,affect the performance and physiology of isobutanol producing strains.We equipped yeast with isobutanol cassettes which had either a mitochondrial or cytosolic localized isobutanol pathway and used either a redox-imbalanced(NADPH-dependent)or redox-balanced(NADH-dependent)ketol-acid reductoisomerase enzyme.We then conducted transcriptomic,proteomic and metabolomic analyses to elucidate molecular differences between the engineered strains.Pathway localization had a large effect on isobutanol production with the strain expressing the mitochondrial-localized enzymes producing 3.8-fold more isobutanol than strains expressing the cytosolic enzymes.Cofactor-balancing did not improve isobutanol titers and instead the strain with the redox-imbalanced pathway produced 1.5-fold more isobutanol than the balanced version,albeit at low overall pathway flux.Functional genomic analyses suggested that the poor performances of the cytosolic pathway strains were in part due to a shortage in cytosolic Fe-S clusters,which are required cofactors for the dihydroxyacid dehydratase enzyme.We then demonstrated that this cofactor limitation may be partially recovered by disrupting iron homeostasis with a fra2 mutation,thereby increasing cellular iron levels.The resulting isobutanol titer of the fra2 null strain harboring a cytosolic-localized isobutanol pathway outperformed the strain with the mitochondrial-localized pathway by 1.3-fold,demonstrating that both localizations can support flux to isobutanol.Francesca V.Gambacorta Ellen R.Wagner Tyler B.Jacobson Mary Tremaine Laura K.Muehlbauer Mick A.McGee Justin J.Baerwald Russell L.Wrobel John F.Wolters Mike Place Joshua J.Dietrich Dan Xie Jose Serate Shabda Gajbhiye Lisa Liu Maikayeng Vang-Smith Joshua J.Coon Yaoping Zhang Audrey P.Gasch Daniel Amador-Noguez Chris Todd Hittinger Trey K.Sato Brian F.Pfleger 2022Synthetic and Systems Biotechnology2022,7,2:1

8On-line Time Differential Perturbed Angular Correlation With Light Probe Nucleus ^(19)F显示文摘1 IntroductionThe time differential perturbed angular correlation(TDPAC) has been known for about 50 years. So far, no on-line TDPAC(TDPACOL) has been performed because of mother nuclei all having a rather long lifetime. In addition, almost all TDPAC experiments have been conducted using probe nuclei with a mass heavier than 40.The present work is motivated to develop a TDPACOL technique with light probe nuclei. The key point for it is to have short-lived mother nuclei with a lifetime of, say, several ten seconds. 10Spectroscopic Measurement of Shock Waves in an Arcjet Plasma Expanding Through a Conical Nozzle显示文摘A diamond-shaped shock wave was created in a helium arcjet plasma. Visi- ble/ultraviolet emission spectroscopy was used to investigate the condition for the formation of stable shocks and to determine characteristics of the plasma. Dependence of the position of the shock front on the gas pressure in the expansion region was investigated. It was found that the shock wave arises from the collision of plasma particles and residual neutral atoms in that region. Continuum and line spectra of neutral helium were measured, from which the electron temper- atures were derived. The electron density was deduced from the Inglis-Teller limit of the He I 2p 3 P-3d 3 D series. 11A HREELS study of the adsorption of formic acid on slightly oxidized Nb(110) surface显示文摘An investigation has been made on the adsorption and decomposition of formic acid on slightly oxidized Nb(110) surface (0/Nb atom ratio = 0.2) using high resolution electron energy loss spectroscopy (HREELS),and a corresponding surface reaction mode is given.At 140 K,formic acid of low exposure on such an Nb(110) surface decomposed to formate,which bonded on Nb in monodentate configuration,simultaneously some formate decomposed to CO,which adsorbed on the surface.Formic acid multilayers formed when the exposure was high.While the temperature was increased to above 190 K,multilayer formic acid desorbed,and the surface was covered with mon-odentate-bonded formate and CO.In the temperature range of 250-300 K,chemisorbed formate changed from monodentate configuration into bridging configuration and CO molecules disappeared.The decomposition of formate at higher temperatures led to the oxidation of Nb.The formate formed in the high exposure case was so stable that it did not decompose even the temperature汪寅生 魏绪明 田志坚 翟润生 T.Ushikubo K.Sato 庄叔贤 1997Science China Chemistry1997,40,1:0
