1Regulation of hepatic blood flow:The hepatic arterial buffer response revisited显示文摘The interest in the liver dates back to ancient times when it was considered to be the seat of life processes. The liver is indeed essential to life,not only due to its complex functions in biosynthesis,metabolism and clearance,but also its dramatic role as the blood volume reservoir. Among parenchymal organs,blood flow to the liver is unique due to the dual supply from the portal vein and the hepatic artery. Knowledge of the mutual communication of both the hepatic artery and the portal vein is essential to understand hepatic physiology and pathophysiology. To distinguish the individual importance of each of these inflows in normal and abnormal states is still a challenging task and the subject of on-going research. A central mechanism that controls and allows constancy of hepatic blood flow is the hepatic arterial buffer response. The current paper reviews the relevance of this intimate hepatic blood flow regulatory system in health and disease. We exclusively focus on the endogenous interrelationship between the hepatic arterial and portal venous inflow circuits in liver resection and transplantation,as well as inflammatory and chronic liver diseases. 2Current biomarkers for hepatocellular carcinoma: Surveillance, diagnosis and prediction of prognosis显示文摘Biomarkers for surveillance, diagnosis and prediction of prognosis in patients with hepatocellular carcinoma(HCC) are currently not ready for introduction into clinical practice because of limited sensitivity and specificity. Especially for the early detection of small HCC novel biomarkers are needed to improve the current effectiveness of screening performed byultrasound. The use of high-throughput technologies in hepatocellular research allows to identify molecules involved in the complex pathways in hepatocarcinogenesis. Several invasive and non-invasive biomarkers have been identified already and have been evaluated in different clinical settings. Gene signatures with prognostic potential have been identified by gene expression profiling from tumor tissue. However, a single 'all-in-one' biomarker that fits all-surveillance, diagnosis, prediction of prognosis-has not been found so far. The future of biomarkers most probably lies in a combination of non-invasive biomarkers, imaging and clinical parameters in a surveillance setting. Molecular profiling of tumorous and non-tumorous liver tissue may allow a prediction of prognosis for the individual patient and hopefully clear the way for individual treatment approaches. 3Plant Surfaces:Structures and Functions for Biomimetic Innovations显示文摘An overview of plant surface structures and their evolution is presented. It combines surface chemistry and architecture with their functions and refers to possible biomimetic applications. Within some 3.5 billion years biological species evolved highly complex multifunctional surfaces for interacting with their environments: some 10 million living prototypes(i.e., estimated number of existing plants and animals) for engineers. The complexity of the hierarchical structures and their functionality in biological organisms surpasses all abiotic natural surfaces: even superhydrophobicity is restricted in nature to living organisms and was probably a key evolutionary step with the invasion of terrestrial habitats some 350–450 million years ago in plants and insects. Special attention should be paid to the fact that global environmental change implies a dramatic loss of species and with it the biological role models. Plants, the dominating group of organisms on our planet, are sessile organisms with large multifunctional surfaces and thus exhibit particular intriguing features.Superhydrophilicity and superhydrophobicity are focal points in this work. We estimate that superhydrophobic plant leaves(e.g., grasses) comprise in total an area of around 250 million km^2, which is about 50% of the total surface of our planet. A survey of structures and functions based on own examinations of almost 20,000 species is provided, for further references we refer to Barthlott et al.(Philos. Trans. R. Soc. A 374: 20160191, 1). A basic difference exists between aquatic nonvascular and land-living vascular plants; the latter exhibit a particular intriguing surface chemistry and architecture. The diversity of features is described in detail according to their hierarchical structural order. The first underlying and essential feature is the polymer cuticle superimposed by epicuticular wax and the curvature of single cells up to complex multicellular structures. A descriptive terminology for this diversity is provided. Simplified, the functions of plant surface characteristics may be grouped into six categories:(1) mechanical properties,(2) influence on reflection and absorption of spectral radiation,(3) reduction of water loss or increase of water uptake, moisture harvesting,(4) adhesion and nonadhesion(lotus effect, insect trapping),(5) drag and turbulence increase, or(6) air retention under water for drag reduction or gas exchange(Salvinia effect). This list is far from complete. A short overview of the history of bionics and the impressive spectrum of existing and anticipated biomimetic applications are provided. 5施马伦贝格病毒核衣壳蛋白的原核表达、纯化及多克隆抗体的制备显示文摘本研究旨在表达施马伦贝格病毒(Schmallenberg virus,SBV)的核衣壳蛋白(N),进而制备其多克隆抗体。以SBV(BH80株)的RNA为模板,RT-PCR扩增N基因序列,并将其克隆至原核表达载体pET-28a-c(+)中,获得pET-28a-SBV-N重组质粒。经酶切和测序鉴定后,将重组质粒转化E.coli BL21(DE3),IPTG诱导表达HisSBV-N融合蛋白。在非变性条件下用Ni-NTA柱纯化His-SBV-N,并以此为抗原免疫新西兰大白兔制备多抗。经免疫亲和层析纯化后,利用Western blot和间接免疫荧光对多抗进行鉴定。结果表明:(1)His-SBV-N融合蛋白在E.coli BL21(DE3)中同时以可溶性和包涵体2种形式表达,相对分子质量约为30ku;(2)制备的多抗效价高达1∶64 000。该多抗不仅能与His-SBV-N融合蛋白发生特异性反应,而且还能识别SBV的不同毒株,但与布尼亚病毒科内亲缘关系最近的沙门达病毒(Shamonda virus)、道格拉斯病毒(Douglas virus)和赤羽病病毒(Akabane virus)的核衣壳蛋白存在交叉反应。His-SBV-N融合蛋白及其多抗的成功制备,为施马伦贝格病血清学检测方法的建立奠定了物质基础。张永宁 吴绍强 吕继洲 冯春燕 王彩霞 袁向芬 邓俊花 林祥梅 Wernike Kerstin 2013畜牧兽医学报2013,44,10:11

7Yangtze Three Gorges Reservoir, China: A holistic assessment of organic pollution, mutagenic effects of sediments and genotoxic impacts on fish显示文摘Besides obvious benefits,the Three Gorges Dam's construction resulted in new pollution scenarios with the potentials to threaten the Three Gorges Reservoir(TGR) ecosystem.In order to record organic contamination,to find links to ecotoxicological impacts and to serve as reference for ensuing monitoring,several sites in the TGR area were screened applying the triad approach with additional lines-of-evidence as a holistic assessment method.Sediments and the benthic fish species Pelteobagrus vachellii were sampled in 2011 and 2012 to determine organic pollution levels,mutagenic potentials and genotoxic impacts.Two regional hot-spots near the cities of Chongqing and Kaixian were identified and further investigated in 2013.Only polycyclic aromatic hydrocarbons(PAHs) could be detected in sediments in 2011(165–1653 ng/g),emphasizing their roles as key pollutants of the area.Their ubiquity was confirmed at Chongqing(150–433 ng/g) and Kaixian(127–590 ng/g) in2013.Concentrations were comparable to other major Chinese and German rivers.However,the immense sediment influx suggested a deposition of 216–636 kg PAH/day(0.2–0.6 mg PAH/(m2·day)),indicating an ecotoxicological risk.PAH source analysis highlighted primary impacts of combustion sources on the more industrialized upper TGR section,whereas petrogenic sources dominated the mid-low section.Furthermore,sediment extracts from several sites exhibited significant activities of frameshift promutagens in the Ames fluctuation assay.Additionally,significant genotoxic impairments in erythrocytes of P.vachellii were detected(Chongqing/Kaixian),demonstrating the relevance of genotoxicity as animportant mode of action in the TGR's fish.PAHs,their derivatives and non-target compounds are considered as main causative agents.Tilman Floehr Bj?rn Scholz-Starke Hongxia Xiao Josef Koch Lingling Wu Junli Hou Anja Wolf Axel Bergmann Kerstin Bluhm Xingzhong Yuan Martina Ro?-Nickoll Andreas Sch?ffer Henner Hollert 2015Journal of Environmental Sciences2015,27,12:9

11Gender-specific associations between coronary heart disease and other chronic diseases: cross-sectional evaluation of national survey data from adult residents of Germany显示文摘Background Combinations of coronary heart disease(CHD) and other chronic conditions complicate clinical management and increase healthcare costs. The aim of this study was to evaluate gender-specific relationships between CHD and other comorbidities. Methods We analyzed data from the German Health Interview and Examination Survey(DEGS1), a national survey of 8152 adults aged 18-79 years. Female and male participants with self-reported CHD were compared for 23 chronic medical conditions. Regression models were applied to determine potential associations between CHD and these 23 conditions. Results The prevalence of CHD was 9%(547 participants): 34%(185) were female CHD participants and 66%(362) male. In women, CHD was associated with hypertension(OR = 3.28(1.81-5.9)), lipid disorders(OR = 2.40(1.50-3.83)), diabetes mellitus(OR = 2.08(1.24-3.50)), kidney disease(OR = 2.66(1.101-6.99)), thyroid disease(OR = 1.81(1.18-2.79)), gout/high uric acid levels(OR = 2.08(1.22-3.56)) and osteoporosis(OR = 1.69(1.01-2.84)). In men, CHD patients were more likely to have hypertension(OR = 2.80(1.94-4.04)), diabetes mellitus(OR = 1.87(1.29-2.71)), lipid disorder(OR = 1.82(1.34-2.47)), and chronic kidney disease(OR = 3.28(1.81-5.9)). Conclusion Our analysis revealed two sets of chronic conditions associated with CHD. The first set occurred in both women and men, and comprised known risk factors: hypertension, lipid disorders, kidney disease, and diabetes mellitus. The second set appeared unique to women: thyroid disease, osteoporosis, and gout/high uric acid. 12Early life exposure to per-and polyfluoroalkyl substances(PFASs):A critical review显示文摘Due to the dynamic developmental processes during pregnancy,infancy,childhood and adolescence,exposure to PFASs is hypothesized to have the most pronounced negative effects during this period.In this review we critically evaluate the current state of the science regarding human early life exposure processes(until 18 years of age)to per-and polyfluoroalkyl substances(PFASs).Efficient placental transfer of perfluoroalkyl acids(PFAAs)results in relatively high prenatal exposure compared with many neutral organic contaminants.The few biomonitoring studies that specifically target infants,toddlers and other children suggest relatively high serum concentrations of perfluorooctane sulfonic acid(PFOS)and perfluorooctanoic acid(PFOA)in early life with peak concentrations occurring sometime before the child reaches 20 months.This peak in serum concentrations is most likely explained by exposure via breastfeeding,ingestion of house dust and/or specific contact events with consumer products leading to high body weight normalized estimated daily intakes(EDIs).Although children have higher EDIs of PFASs than adults,these are not always reflected by higher serum levels of PFASs in children in cross-sectional biomonitoring studies due to the confounding effect of age and birth cohort,and different exposure histories due to production changes.Longitudinal exposure studies measuring internal and external exposure(for multiple pathways and PFASs)at several time points during early life are strongly encouraged to understand temporal changes in exposure of individual children.A better quantitative understanding of early life exposure processes would help to improve the validity of epidemiological studies and allow informed decisions regarding setting of regulatory thresholds and appropriate mitigation actions.Kerstin Winkens Robin Vestergren Urs Berger Ian TCousins 2017Emerging Contaminants2017,3,2:6

15Spatial distributions of tropical tree species in northern Vietnam under environmentally variable site conditions显示文摘Ecological interactions of species and thus their spatial patterns may differ between homogeneous and heterogeneous forests. To account for this, techniques of point pattern analysis were implemented on mapped locations of tree individuals from two 1-ha tropicalforest plots in Vietnam. We analyzed the effect of environmental heterogeneity on tree distributions; spatial distribution patterns of dominant species; inter-specific associations; and conspecific associations between life stages. Our analyses showed that: environmental conditions were homogeneous at plot 1 but heterogeneous at plot 2; in both plots, all six dominant species were aggregated at various scales up to 30 m, and tree species were aggregated at larger scales in the homogeneous site than in the heterogeneous site; attraction between pairs of species was remarkably higher at the homogeneous site while negative associations were more frequent in the heterogeneous site; some species, H. kurzii, T. ilicifolia(homogeneous plot) and D. sylvatica, S. wightianum(heterogeneous plot),showed a lack of early life-stage individuals near conspecific adults. Moreover, additional clustering of young individuals was independent from conspecific adults, except D. sylvatica in both sites. These findings are consistent with the Janzen-Connell hypothesis. 16Coexpression of receptor-tyrosine-kinases in gastric adenocarcinoma-a rationale for a molecular targeting strategy?显示文摘AIM: To define the (co-)expression pattern of target receptor-tyrosine-kinases (RTK) in human gastric adenocarcinoma. METHODS: The (co-)expression pattern of VEGFR1-3,PDGFRα/b and EGFR1 was analyzed by RT-PCR in 51 human gastric adenocarcinomas. In addition,IHC staining was applied for confirmation of expression and analysis of RTK localisation. RESULTS: The majority of samples revealed a VEGFR1 (98%),VEGFR2 (80%),VEGFR3 (67%),PDGFRα (82%) and PDGFRβ(82%) expression,whereas only 62% exhibited an EGFR1 expression. 78% of cancers expressed at least four out of six RTKs. While VEGFR1-3 and PDGFRα revealed a predominantly cytoplasmatic staining in tumor cells,accompanied by an additional nuclear staining for VEGFR3 ,EGFR1 was almost exclusively detected on the membrane of tumor cells. 17Distribution and effects of tree leaf litter on vegetation composition and biomass in a forest–grassland ecotone显示文摘Aims After abandonment of grasslands,secondary succession leads to the invasion by woody species.This process begins with the accumulation of tree litter in the forest–grassland ecotone.Our objectives were to determine the relationships between litter amounts and vegetation composition and cover along natural forest–grassland ecotones and to experimentally study the initial effects of tree litter accumulation on grassland vegetation and on microsite conditions.Methods We established 11 transects varying from 12 to 15 m in length in different forest–grassland ecotones in the Lahn-Dill highlands,Germany,and measured the mass and cover of tree litter and the cover and composition of vegetation at five sequential positions along each transect by using 1 m2 plots with five replications.In a field experiment,we established plots subjected to different litter amounts(0,200 and 600g m^(−2))and evaluated changes in grassland vegetation,soil temperature and soil nutrient availability below the litter layer.Important Findings Tree litter amounts decrease from 650 to 65g m^(−2) across the forest–grassland ecotone.Vegetation changed from shrubs and annual species(adapted to more stressful conditions)in the forests edge to grasses,rosettes and hemirosette species(with higher competitive abilities)in the grassland.These anthropogenic forest–grassland ecotones showed abrupt edges,and the two adjacent ecosystems were characterized by different species pools and functional groups.In the field experiment,the presence of a litter layer reduced vegetation biomass and cover;the species richness was only reduced in the treatment with high litter(600g m^(−2)).Additionally,adding litter on top of vegetation also reduced thermal amplitude and the number of frost days,while increasing the availability of some nutrients,such as nitrogen and aluminium,the latter being an indicator of soil acidification.Adding a tree litter layer of 600g m^(−2) in grassland areas had strong effects on the composition and diversity of grassland vegetation by reducing the cover of several key grassland species.In,or near,forest edges,litter accumulation rapidly changes established vegetation,microsite conditions and soil nutrients.Alejandro Loydi Kerstin Lohse Annette Otte Tobias W.Donath R.Lutz Eckstein 2014Journal of Plant Ecology2014,7,3:3

20Prognostic value of serum microRNA-122 in hepatocellular carcinoma is dependent on coexisting clinical and laboratory factors显示文摘BACKGROUND There is ongoing search for new noninvasive biomarkers to improve management of patients with hepatocellular carcinoma(HCC).Studies,mostly from the Asian-Pacific region,demonstrated differential expression of liverspecific microRNA-122(miR-122)in tissue as well as in sera of patients with hepatitis B virus-and hepatitis C virus-induced HCC.AIM To evaluate prognostic value of miR-122 in patients with HCC in a European population and determine potential factors related to alteration of miR-122 in sera.METHODS Patients with confirmed HCC(n=91)were included in the study over a two-year period.Patients were characterized according to Child-Pugh score,Barcelona clinic liver cancer(BCLC)staging system,etiology of liver disease,laboratory parameters and overall survival.MiR-122 was measured in sera using TaqMan assay normalized to spiked-in cel-miR-39.RESULTS Serum miR-122 quantity was independent of the Child-Pugh score,the BCLC stage or the underlying etiology.Significant positive correlation was found between miR-122 and alanine aminotransferase(P<0.0001),aspartate aminotransferase(P=0.0001),alpha-fetoprotein(AFP)(P=0.0034)and hemoglobin concentration(P=0.076).Negative correlation was observed between miR-122 level and creatinine concentration(P=0.0028).AFP,Child-Pugh score and BCLC staging system were associated with survival differences.In overall cohort low miR-122 in sera was only associated with a trend for a better overall survival without reaching statistical significance.Subgroup analysis revealed that low miR-122 was significantly associated with better prognosis in patients with advanced cirrhosis(Child-Pugh class B/C),advanced tumor stage(BCLC B/C/D)and normal AFP(<7 ng/mL).CONCLUSION Our results strongly support the value of miR-122 as potential biomarker of liver injury and probably prognosis.Nevertheless,the value of miR-122 in prediction of prognosis of HCC patients was limited to certain patients’subgroups.Since circulating miR-122 may be influenced by impaired renal function,AFP and hemoglobin concentration,those factors need to be considered while interpreting miR-122 level.Martin Franck Kerstin Schütte Peter Malfertheiner Alexander Link 2020World Journal of Gastroenterology2020,26,1:3
