| 1 | Establishment of the diploid gynogenetic hybrid clonal line of red crucian carp × common carp显示文摘This study investigated the gynogenetic cytobiological behavior of the third gynogenetic generation (G3), which was generated from the diploid eggs produced by the second gynogenetic generation (G2) of red crucian carp × common carp, and determined the chromosomal numbers of G3, G2×scatter scale carp and G2×allotetraploid hybrids of red crucian carp × common carp. The results showed that the diploid eggs of G2 with 100 chromosomes, activated by UV-irradiated sperm from scatter scale carp and without the treatment for doubling the chromosomes, could develop into G3 with 100 chromosomes. Similar to the first and second gynogenetic generations (G1 and G2), G3 was also diploid (2n=100) and presented the hybrid traits. The triploids (3n=150) and tetraploids (4n=200) were produced by crossing G2 with scatter scale carp and crossing G2 with allotetraploids, respectively. The extrusion of the second polar body in the eggs of G2 ruled out the possibility that the retention of the second polar body led to the formation of the diploid eggs. In addition, we discussed the mechanism of the formation of the diploid eggs generated by G2. The establishment of the diploid gynogenesis clonal line (G1, G2 and G3) provided the evidence that the diploid eggs were able to develop into a new diploid hybrid clonal line by gynogenesis. | 3 | Rapid growth and sterility of growth hormone gene transgenic triploid carp显示文摘Triploid carp(100%)with 150(3n=150)chromosomes were obtained by crossing the females of improved tetraploid hybrids(♀, 4n=200)of red crucian carp(♀)×common carp(♂)with the males of diploid yellow river carp(♂,2n=100).The crosses yielded transgenic triploid carp(positive triploid fish,44.2%of the progeny)and non-transgenic triploid carp(negative triploid fish). | 4 | Formation and biological characterization of three new types of improved crucian carp显示文摘The improved tetraploids (G1×AT) were obtained by distant crossing and gynogenesis and the high-body individuals accounted for 2% among G1×AT. After mating with each other, the high-body in- dividuals produced three kinds of bisexual fertile diploid fishes: high-body red crucian carp, high-body fork-like-tails goldfish and gray common carp. The high-body red crucian carp mating with each other formed three types of improved crucian carp (ICC) including improved red crucian carp (IRCC), im- proved color crucian carp (ICCC) and improved gray crucian carp (IGCC). The phenotypes, chromo- some numbers, gonadal structure and fertility of the three kinds of ICC and their offspring were observed. All the three kinds of ICC possessed some improved phenotypes such as higher body, smaller head and shorter tail. The ratios of the body height to body length of IRCC, ICCC and IGCC were 0.54, 0.51 and 0.54, respectively. All of them were obviously higher than that of red crucian carp 0.41 (P<0.01). Three kinds of ICC had the same chromosome number as red crucian carp with 100 chromosomes. All the ICC possessed normal gonads producing mature eggs or sperm, which was important for the production of an improved diploid population. Compared with red crucian carp, all the ICC had stronger fertility such as higher gametes production, higher fertilization rate and higher hatchery rate. Three types of improved diploid fish population were generated from the three kinds of ICC by self-crossing, respectively. The ICC can serve as ornamental fish and edible fish. They are also ideal parents to produce triploids by mating with tetraploids. | 5 | Genomic variation in the hybrids of white crucian carp and red crucian carp: evidence from ribosomal DNA显示文摘In this study, we conducted a cross of white crucian carp(♀)×red crucian carp(♂)(WR), and characterized the morphology, reproduction and genetics of the progeny. Different from parents, WR with the gray color showed the hybrid morphological traits of both parents. WR possessed normal gonads producing mature eggs or sperm, and exhibited high fertilization rate(90.2%) and high hatchery rate(80.5%), which contributed to produce and enlarge the population. WR with the same DNA content as parents was a diploid fish with 100 chromosomes(2n=100). Amplified ITS of 45 S r DNA, in WR the sequences consisting of 884 bp bases of the entire ITS-1 region, 5.8 S region, and entire ITS-2 region. The sequences showed high similarity between WR and its parents and leaned towards male inheritance. In WR, NTS of 5S r DNA consisted of three length types with total 654 bp bases. From sequence analysis of NTS, WR shared 94.2% and 95.1% similarities with their female and male parent, respectively. Sequence analysis of ITS and NTS revealed that there existed recombination and variation in the hybrid progeny, which was the genetic base for adaptation and speciation. In conclusion, we obtained WR from hybridization and it exhibited hybrid traits in morphology and variation in genetic composition showing essential difference with its parents. However, for a long time, systematic theories and technologies related to hybridization in fish have been lacking. In this study, through long-term systematic research, we investigated and obtained the main rules regarding inheritance and reproduction related to fish distant hybridization. Furthermore, we established one-step and multistep breeding technologies that were suitable for interspecific hybridization and intraspecific hybridization. Simultaneously, we used these two breeding technologies to produce a batch of diploid fish lineages and tetraploid fish lineages and improved fishes. In addition, we widely discuss the methods, technologies and results of hybridization breeding, referring to the domestic and foreign literature on fish hybridization. | 7 | A new type of triploid crucian crap—red crucian carp (♀)×allotetraploid (♂)显示文摘A new type of triploid hybrids (TC) was produced by crossing red crucian carp (♀) with allotetraploid hybrids (♂). This type of triploids with no barbel was spindle-shaped and gray in color. Compared with their parents, some data of the morphological traits in the triploid hybrids were intermediate to those of their parents, and some were beyond their parents with heterozygous traits. Under the same culture conditions, the growth rate of the new type triploids was faster than their maternal red crucian carp (RC). The gonadal development of triploids was obviously slower than that of diploids, and the germ cells in them were degenerated. Compared with another type of triploids produced by Japanese crucian carp (♀) with allotetraploid hybrids (♂), the new type triploids not only kept faster growth rate and sterility, but also had no barbel that was similar to the common carp. | 8 | Effects of CO_(2)atmosphere on slow pyrolysis of high-ash lignite显示文摘Slow pyrolysis of a typical high-ash lignite in China was carried out in atmospheres of N_(2),20%CO_(2)/N_(2),40%CO_(2)/N_(2),and 60%CO_(2)/N_(2)in a fixed bed reactor.The evolution of char,tar,and gases in yield and characteristics,and the physico-chemical characteristics of chars in different atmospheres were compared.Results revealed that CO_(2)almost behaves as an inert gas in the evolution of char and CO before 600℃.The decrease in char and CO_(2)yields and the increase in CO yield at temperatures higher than 600℃are ascribed to the occurrence of char-CO_(2)gasification.The higher the CO_(2)level present in the atmosphere,the higher the severity of the gasification becomes.Introducing CO_(2)into the atmosphere raises tar and water yields but cuts down H_(2),CH_(4),and C_(2)H_(6)yields.The promotion of tar yield results from the higher phenols and aliphatics yields in the presence of CO_(2).The reduction of H_(2)yield is associated with the increase in water yield,while the decline of light hydrocarbon gases is connected with the suppression effects of CO_(2)on methyl decomposition.The role of inherent minerals was also investigated by comparing product yields from raw coal and demineralized coal.The comparison indicated that the char-CO_(2)gasification rate is sharply reduced to nearly zero without the catalytic effects of calcium,iron,and magnesium minerals.The evolution of tar and light hydrocarbon gases in the CO_(2)-containing atmosphere greatly depends on inherent minerals.Without minerals,tar yield is inhibited in the presence of CO_(2).In the absence of minerals,CO_(2)hardly has influences on CH_(4)and C_(2)H_(6)yields. | Qinhui Wang Kaikun Li Zhihang Guo Mengxiang Fang Zhongyang Luo Kefa Cen | 2018 | Carbon Resources Conversion2018,1,1: | 4 |

| 10 | miR-199-5p regulates spermiogenesis at the posttranscriptional level via targeting Tekt1 in allotriploid crucian carp显示文摘Background:Sperm abnormalities are one of the primary factors leading to male sterility,but their pathogenesis is still unclear.Although miRNAs are suggested to exert important roles in the regulation of spermatogenesis at both transcriptional and posttranscriptional levels,little is currently known regarding the regulation of sperm flagella assembly by microRNAs(miRNAs).The role of miRNAs in the development of sperm abnormalities in sterile triploid fish has not been studied.Results:In this study,we found that miR-199-5p was widely expressed in all detected tissues of different-ploidy crucian carp.As one of the testis-specific candidate markers,Tekt1 was predominantly expressed in the testis.Quantitative real-time PCR(qRT-PCR)analyses showed that the expression trend of miR-199-5p was exactly opposite to that of Tekt1.Through bioinformatics analysis,we identified a putative miR-199-5p binding site in the Tekt1 mRNA.We further identified Tekt1 as a target of miR-199-5p using luciferase reporter assay.Finally,we confirmed that miR-199-5p was necessary for sperm flagellar assembly and spermatogenesis in vivo via intraperitoneal injection of miR-199-5p antagomir or agomir in diploid red crucian carp.Moreover,miR-199-5p gainof-function could lead to spermatids apoptosis and abnormal spermatozoa structure,which is similar to that of allotriploid crucian carp.Conclusions:Our studies suggested that abnormally elevated miR-199-5p inhibited the sperm flagella formation in spermiogenesis by negatively regulating the expression of Tekt1,thereby causing sperm abnormalities of male allotriploid crucian carp. | Shengnan Li Qiubei Wang Lu Huang Siyu Fan Ting Li Yuqing Shu Chun Zhang Yi Zhou Qingfeng Liu Kaikun Luo Min Tao Shaojun Liu | 2022 | Journal of Animal Science and Biotechnology2022,13,5: | 1 |

| 12 | Embryonic development and organogenesis of Chinese giant salamander,Andrias davidianus显示文摘The morphology and organogenesis of Chinese giant salamander, Andrias davidianus, in its different developmental periods and stages are described in detail, which provides an intact criterion for distinguishing different stages of its developmental process. Based on the external morphological and internal histological features, six periods including 20 stages of organogenesis of Chinese giant salamander are established, which are cleavage period, blastula period, gastrula period, neurula period, organogenesis stage and hatching stage. Generally, the embryonic development of Chinese giant salamander is consistent with those of Eastern newt, Cynops orientalis, and Black spots frog, R. nigromaculata. However, they have some differences in the early cleavage process and the development of digestive system. The cleavage of Chinese giant salamander, A. davidianus is not a discoidal division type, which is different from other species reported. | 13 | An improved hybrid bream derived from a hybrid lineage of Megalobrama amblycephala(♀)×Culter alburnus(♂)显示文摘Distant hybridization is an important technique in fish genetic breeding.In this study,based on the establishment of an allodiploid fish lineage(BT,2n=48,F_(1)–F_(6))derived from distant hybridization between female Megalobrama amblycephala(BSB,2n=48)and male Culter alburnus(TC,2n=48),and the backcross progeny(BTB,2n=48)derived by backcrossing female F_(1) of BT to male BSB,an improved hybrid bream(BTBB,2n=48)was obtained by backcrossing BTB(♀)to BSB(♂).Moreover,the morphological and genetic characteristics of BTBB individuals were investigated;BTBB was similar to BSB in appearance but had a higher body height than BSB.The study results regarding chromosome numbers and DNA content indicated that BTBB is a diploid hybrid fish.The 5S rDNA and Hox gene of BTBB were inherited from the original parents.Gonadal development in BTBB was normal.On the other hand,BTBB had a faster growth rate,higher muscle protein level,and lower muscle carbohydrate level than BSB.Hence,bisexual fertile BTBB is promoted and can be applied as a high-quality fish,and it can also be used as a new fish germplasm resource to develop high-quality fish further.Thus,this study is of great significance for fish genetic breeding. | Dingbin Gong Min Tao Lihui Xu Fangzhou Hu Zehong Wei Shi Wang Yude Wang Qingfeng Liu Chang Wu Kaikun Luo Chenchen Tang Rong Zhou Chun Zhang Yuequn Wang Shaojun Liu | 2022 | Science China(Life Sciences)2022,65,6: | 0 |
