| 1 | Activation of extrasynaptic GABA_A receptors inhibits cyclothiazideinduced epileptiform activity in hippocampal CA1 neurons显示文摘Extrasynaptic GABA_A receptors(GABA_ARs)-mediated tonic inhibition is reported to involve in the pathogenesis of epilepsy.In this study,we used cyclothiazide(CTZ)-induced in vitro brain slice seizure model to explore the effect of selective activation of extrasynaptic GABA_ARs by 4,5,6,7-tetrahydroisoxazolo[5,4-c]pyridine-3-ol(THIP)on the CTZ-induced epileptiform activity in hippocampal neurons.Perfusion with CTZ dose-dependently induced multiple epileptiform peaks of evoked population spikes(PSs)in CA1 pyramidal neurons,and treatment with THIP(5 umol/L)significantly reduced the multiple PS peaks induced by CTZ stimulation.Western blot showed that the 5-subunit of the GABA_AR,an extrasynaptic specific GABA_AR subunit,was also significantly down-regulated in the cell membrane 2h after CTZ treatment.Our results suggest that the CTZ-induced epileptiform activity in hippocampal CA1neurons is suppressed by the activation of extrasynaptic GABA_ARs,and further support the hypothesis that tonic inhibition mediated by extrasynaptic GABA_ARs plays a prominent role in seizure generation. | Li Wan Xu Liu Zheng Wu Wanting Ren Shuzhen Kong Raya Abou Dargham Longzhen Cheng Yun Wang | 2014 | Neuroscience Bulletin2014,30,5: | 3 |
| 2 | Solid-state cold spraying of FeCoCrNiMn high-entropy alloy:an insight into microstructure evolution and oxidation behavior at 700-900℃显示文摘About 3 mm thick five-element equimolar high-entropy alloy(HEA) FeCoCrNiMn was successfully deposited by solid-state cold spraying(CS).The high-temperature oxidation behavior of the CSed HEA was investigated at 700-900℃.Heat treatment was performed on the CSed HEA before oxidation to heal the incomplete interfaces between the deposited particles.Results show that the microstructure of the CSed HEA is characterized by grain refinement and abundant interparticle incomplete interfaces.Post-spray heat treatment promotes recrystallization and grain growth in the CSed HEA.After oxidation testing,the oxide scales are composed of multi-layers:a Mn_(2)O_(3)(or Mn_(3)O_(4)) outer layer,a Mn-Cr spinel intermediate layer and a Cr_(2)O_(3) inner layer.The CSed HEA exhibits higher parabolic rate constants and more favorable internal oxidation than the bulk HEAs that have similar compositions in the literature.Such a discrepancy becomes pronounced at higher temperatures.The grain refinement and numerous particle boundaries are responsible for such a distinctive performance of the CSed HEA. | Yaxin Xu Weny Li Longzhen Qu Xiawei Yang Bo Song Rocco Lupoi Shuo Yin | 2021 | Journal of Materials Science & Technology2021,,9: | 2 |

| 7 | Clinical role of ^(18)F-FDG PET/CT-based simultaneous modulated accelerated radiotherapy treatment planning for locally advanced nasopharyngeal carcinoma显示文摘Objective The aim of this study was to compare the long-term local control, overall survival, and late toxicities of positron emission tomography/computed tomography(PET/CT)-guided dose escalation radiotherapy versus conventional radiotherapy in the concurrent chemoradiotherapy treatment of locally advanced nasopharyngeal carcinoma(NPC).Methods A total of 48 patients with stage III–IVa NPC were recruited and randomly administered PET/CTguided dose escalation chemoradiotherapy(group A) or conventional chemoradiotherapy(group B). The dose-escalation radiotherapy was performed using the simultaneous modulated accelerated radiotherapy technique at prescribed doses of 77 gray(Gy) in 32 fractions(f) to the gross target volume(GTV): planning target volume(PTV) 1 received 64 Gy/32 f, while PTV2 received 54.4 Gy/32 f. Patients in group B received uniform-dose intensity-modulated radiotherapy, PTV1 received 70 Gy/35 f and PTV2 received 58 Gy/29 f. Concurrent chemotherapy consisted of cisplatin [20 mg/m2 intravenous(IV) on days 1–4] and docetaxel(75 mg/m2 IV on days 1 and 8) administered during treatment weeks 1 and 4. All patients received 2–4 cycles of adjuvant chemotherapy of the same dose and drug regimen.Results The use of fluorine-18-fluorodeoxyglucose(^(18)F-FDG) PET/CT significantly reduced the treatment volume delineation of the GTV in 83.3%(20/24) of patients. The 5-year local recurrence-free survival rates of the two groups were 100% and 79.2%, respectively(P = 0.019). The 5-year disease free survival(DFS) rates were 95.8% and 75.0%, respectively(P = 0.018). The 5-year local progression-free survival and DFS rates were significantly different. The 5-year overall survival(OS) rates were 95.8% and 79.2%, respectively. Differences in OS improvement were insignificant(P = 0.079). Late toxicities were similar in the two groups. The most common late toxicities of the two arms were grade 1–2 skin dystrophy, xerostomia, subcutaneous fibrosis, and hearing loss.
