1Homogentisic acid γ-lactone suppresses the virulence factors of Pseudomonas aeruginosa by quenching its quorum sensing signal molecules显示文摘Bacterial quorum sensing(QS) molecules are involved in the coordination of certain behaviors such as biofilm formation, virulence and antibiotic resistance. QS molecules(autoinducers) and their corresponding receptors have been recognized as important therapeutic targets for drug-resistant infections and biofilm-associated infections(BAI). This study assessed the multiple biological effects of homogentisic acid y-lactone(HgAL), a furanone derivative. The anti-QS and anti-biofilm effects of HgAL against PAO1 strain were evaluated using CLSM, SEM, HPLC and other biochemical methods The results showed that HgAL could effectively inhibit the production of pyocyanin and extracellular matrix, as well as reduce the adherence ability and biofilm formation of Pseudomonas aeruginosa. Inhibition of virulence is attributed to the suppressive effect of HgAL on biosynthesis of 3-oxo-C_(12)-HSLand C4-HSL(two kinds of QS signaling molecules in P. aeruginosa). Our results support HgAL as a potential agent for prevention of BAI in the healthcare settings.Shuihong Li Polly H.M. Leung Xinya Xu Chunhui Wu 2018Chinese Chemical Letters2018,29,2:2
2Discovery of ferrocene-carborane derivatives as novel chemical antimicrobial agents against multidrug-resistant bacteria显示文摘Antimicrobial resistance has now become a very serious global public health problem. New drug discovery and development are urgently needed to combat the growing threat of multidrug-resistant (MDR) bacteria. The aim of this study was to explore the potential application of three ferrocene-carborane derivatives as new promising agents to confront the problem of increasing antibiotic resistance. The results of agar diffusion bioassay, minimal inhibitory concentrations (MIC) testing and time-kill assay illustrate their broad-spectrum antimicrobial activities to both American Type Culture Collection (ATCC) control strains and MDR clinical isolates. It is evident that the relevant antimicrobial properties are all in a dose-dependent manner and gradually transform into a bactericidal effect from a bacteriostatic effect with the increasing of the drug concentration. Furthermore, these ferrocene-carborane derivatives have no/little toxic effect on normal cells like HELF cells and lead to little hemolysis at their MICs. This raises the possibility to develop novel antimicrobial drugs using these new ferrocene carborane derivants.LI ShuiHong WU ChangYu LV XiaYi TANG Xiao ZHAO XinQing YAN Hong JIANG Hui WANG XueMei 2012Science China Chemistry2012,55,11:2
3Graphene-supported biomimetic catalysts with synergistic effect of adsorption and degradation for efficient dye capture and removal显示文摘Design and development of iron porphyrin-based artificial enzymes system have been attracting a lot of attention.Herein, without any toxic reductant and harsh processing,we present a facile one-pot method to fabricate bifunctional catalytic nanocomposites consisting of graphene and hemin by using vitamin C as a mild reduction reagent. The presence of graphene helps the formation of a high degree of highly active and stable hemin on the graphene surface in a monomeric form through theirπ-πstacking interaction. As a result, such nanocomposites possess a superior adsorption capacity and intrinsic peroxidase-like catalytic activity. Moreover,by the combination of their dye adsorption ability,RGOhemin nanocomposites can serve as a suitable candidate for efficient capture and removal of dyes via a synergistic effect. Our findings may pave the way to apply graphene-supported artificial enzymes in a variety of fields, such as environmental chemistry, bionics, medicine, and biotechnology.Qin Li Xiaopei Wang Xueqing Xiong Shuihong Zhu Zhaohui Meng Yongying Hong Changxu Lin Xiangyang Liu Youhui Lin 2020Chinese Chemical Letters2020,31,1:1

5New strategy for reversing biofilm-associated antibiotic resistance through ferrocene-substituted carborane ruthenium(Ⅱ)-arene complex显示文摘Bacterial biofilms are inherently resistant to antimicrobial agents and are difficult to eradicate with conventional antimicrobial agents, resulting in many persistent and chronic bacterial infections. In this contribution, a new strategy for reversing the biofilm-associated antibiotic resistance has been explored by induction of a carborane ruthenium(II)-arene complex (FcRuSB). Our results demonstrate that the FcRuSB could be utilized as an inducer to efficiently reverse the biofilm-associated antibiotic resistance of multidrug-resistant (MDR) clinical isolates of Staphylococcus aureus and Pseudomonas aeruginosa. The induced effect of FcRuSB is correlated with a considerable decrease in the expression of extracellular matrix proteins (EMP) of the two strains. The considerable decrease of the EMP of induced cells, resulting in the reduction of adherence and biofilm formation ability of the two types of MDR pathogens, and then can cause significantly enhanced sensitivity of them to antibiotics.LI ShuiHong WU ChangYu TANG Xiao GAO ShengPing ZHAO XinQing YAN Hong WANG XueMei 2013Science China Chemistry2013,56,5:0
