1Classification of the Gut Microbiota of Patients in Intensive Care Units During Development of Sepsis and Septic Shock显示文摘The gut microbiota of intensive care unit(ICU)patients displays extreme dysbiosis associated with increased susceptibility to organ failure,sepsis,and septic shock.However,such dysbiosis is difficult to characterize owing to the high dimensional complexity of the gut microbiota.We tested whether the concept of enterotype can be applied to the gut microbiota of ICU patients to describe the dysbiosis.We collected 131 fecal samples from 64 ICU patients diagnosed with sepsis or septic shock and performed 16S rRNA gene sequencing to dissect their gut microbiota compositions.During the development of sepsis or septic shock and during various medical treatments,the ICU patients always exhibited two dysbiotic microbiota patterns,or ICU-enterotypes,which could not be explained by host properties such as age,sex,and body mass index,or external stressors such as infection site and antibiotic use.ICU-enterotype I(ICU E1)comprised predominantly Bacteroides and an unclassified genus of Enterobacteriaceae,while ICU-enterotype II(ICU E2)comprised predominantly Enterococcus.Among more critically ill patients with Acute Physiology and Chronic Health Evaluation II(APACHE II)scores>18,septic shock was more likely to occur with ICU E1(P=0.041).Additionally,ICU E1 was correlated with high serum lactate levels(P=0.007).Therefore,different patterns of dysbiosis were correlated with different clinical outcomes,suggesting that ICU-enterotypes should be diagnosed as independent clinical indices.Thus,the microbial-based human index classifier we propose is precise and effective for timely monitoring of ICU-enterotypes of individual patients.This work is a first step toward precision medicine for septic patients based on their gut microbiota profiles.Wanglin Liu Mingyue Cheng Jinman Li Peng Zhang Hang Fan Qinghe Hu Maozhen Han Longxiang Su Huaiwu He Yigang Tong Kang Ning Yun Long 2020Genomics, Proteomics & Bioinformatics2020,18,6:7

3Engineering self-healing adhesive hydrogels with antioxidant properties for intrauterine adhesion prevention显示文摘Intrauterine adhesion (IUA) is the fibrosis within the uterine cavity. It is the second most common cause of female infertility, significantly affecting women’s physical and mental health. Current treatment strategies fail to provide a satisfactory therapeutic outcome for IUA patients, leaving an enormous challenge for reproductive science. A self-healing adhesive hydrogel with antioxidant properties will be highly helpful in IUA prevention. In this work, we prepare a series of self-healing hydrogels (P10G15, P10G20, and P10G25) with antioxidant and adhesive properties. Those hydrogels exhibit good self-healing properties and can adapt themselves to different structures. They possess good injectability and fit the shape of the human uterus. Moreover, the hydrogels exhibit good tissue adhesiveness, which is desirable for stable retention and therapeutic efficacy. The in vitro experiments using P10G20 show that the adhesive effectively scavenges ABTS+, DPPH, and hydroxyl radicals, rescuing cells from oxidative stress. In addition, P10G20 offers good hemocompatibility and in vitro and in vivo biocompatibility. Furthermore, P10G20 lowers down the in vivo oxidative stress and prevents IUA with less fibrotic tissue and better endometrial regeneration in the animal model. It can effectively downregulate fibrosis-related transforming growth factor beta 1 (TGF-β1) and vascular endothelial growth factor (VEGF). 7Absence of FHIT expression is associated with apoptosis inhibition in colorectal cancer显示文摘Objective: To investigate the expression of fragile histidine triad (FHIT) protein in normal colorectal tissue, colorectal adenoma and colorectal cancer (CRC), and to study the relationships between the expression of FHIT protein and the clinical pathology, the apoptosis-associated protein (Bcl-2, Bax, Survivin), apoptosis in colorectal cancer. Methods: Tissue microarray (TMA) and immunohistochemistry SP were used to detect the expression of FHIT gene, Bcl-2, Bax and Survivin in 16 cases of the normal colorectal tissue, 16 cases of colorectal adenoma and 80 cases of the colorectal cancer. TUNEL was used to detect the apoptosis index (AI) in 80 cases of the colorectal cancer. Results: (1) The positive rates of FHIT gene expression in normal colorectal tissue, colorectal adenoma and adenocancer were 93.75%, 68.75% and 46.25% respectively. There were no significant differences in the relationships between the FHIT gene expression and histological types, the gender as well as the age (P>0.05). There were significant relationships between FHIT gene expression and lymph node metastasis, histological grades, Duke’s system as well as the 5-year survival rate after operation. (2) The positive rates of Bax, Bcl-2 and Survivin in colorectal adenocancer were 72.50%, 51.25%, 77.50% respectively. The expression of FHIT gene was positively correlated with that of Bcl-2, Bax and Survivin. (3) The mean AI in FHIT negative tumors was significantly lower than that in FHIT positive tumors (P<0.01). Conclusion: FHIT gene may play a role in the oncogenesis and progression of colorectal cancer. 10Accumulation conditions and exploration directions of Ordovician lower assemblage natural gas, Ordos Basin, NW China显示文摘Based on drilling cores, well logging and seismic data, source rocks and reservoirs are evaluated;and the natural gas genesis is identified through the analysis of natural gas isotopes, components and fluid inclusions, to study the gas accumulation conditions of the gypsum salt rock related strata of the Ordovician lower assemblage in Ordos Basin.(1) The natural gas from Ordovician lower assemblage is high thermal evolution dry gas from marine source rock, characterized by relatively light δ^(13)C value of methane and heavy δ^(13)C value of ethane. The natural gas is identified as gas cracking from crude oil according to component analysis. Thermochemical sulfate reduction(TSR) reaction has happened between the hydrocarbon fluid and sulfate as sulfur crystals are found in the cores, hydrogen sulfide is found in the natural gas, and hydrocarbon and hydrogen sulfide fluid inclusions are widespread in secondary minerals.(2) Around the gypsum-salt lows, argillaceous rocks are extensive in the Ordovician lower assemblage, reaching a cumulative thickness of 20–80 m. The effective source rocks include argillaceous rock rich in organic laminae, algal clump and algal dolomite. Analysis shows that the source rocks have a dominant TOC of 0.1%–0.5%, 0.31% on average and 3.24% at maximum. The source rocks have an average TOC of 0.58% after recovered through organic acid salt method, indicating the source rocks have high hydrocarbon supply potential.(3) In the sedimentary period, the palaeo-uplift controlled the distribution of reservoirs. The inherited secondary palaeo-uplift in Wushenqi–Jingbian east of the central palaeo-uplift and the low uplift formed by thick salt rocks near Shenmu–Zizhou area controlled the distribution of penecontemporaneous grain shoal dolomite reservoirs. The salinization sedimentary environment of gypsum salt rock can promote the development of reservoir. There are three types of dolomite reservoirs, the one with intercrystalline pore, with dissolution pore, and with fracture;intercrystalline and dissolution pores are main reservoir spaces.(4) There are two types of cap rocks, namely tight carbonate rock and gypsum-salt rock, constituting two types of source-reservoir-cap assemblages respectively. The general accumulation model is characterized by marine source rock supplying hydrocarbon, beach facies limy dolomite reservoir, small fractures acting as migration pathways, and structural-lithologic traps as accumulation zones.(5) The third and fourth members of Majiagou Formation are major target layers in the lower assemblage. The Wushengqi–Jingbian secondary paleo-uplift area and Shenmu–Zizhou low uplift are dolomite and limestone transition zone, there develops tight limestone to the east of the uplift zone, which is conducive to the formation of gas reservoir sealed by lithology in the updip. 11Absence of FHIT expression is associated with apoptosis inhibition in colorectal cancer显示文摘The fragile histidine triad(FHIT)gene,a candidate tumor suppressor gene located at 3p14.2,has been shown to be involved in the carcinogenesis of many human tissues,including digestive tract tissues.However,the expression and the role of the FHIT in the initiation and the development of the colorectal cancer(CRC)are poorly understood.We have shown that the FHIT gene exhibits significantly decreased expression in human CRC compared to colorectal adenoma and normal colorectal tissue by tissue microarray(TMA).The positive rate of FHIT gene expression in normal colorectal tissue,adenoma and adenocarcinoma were 93.75%,68.75%and 46.25%,respectively.We show this decreased expression to be significantly correlated with the progression of colorectal carcinoma(P<0.05)as well as with differentiation and lymph node metastasis(P<0.05).We detected two somatic alterations in the FHIT gene in human CRC.The presence of this mutation correlated significantly with decreased FHIT expression in the human CRC.In our present study we tested the hypothesis that the decreased FHIT expression resulted in apoptosis inhibition associated with abnormal expression of apoptosis related proteins.To test this hypothesis we did a series of experiments.In the first test,we assessed apoptosis status using a standard TUNEL(terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling)assay by comparing FHIT-positive CRC vs.FHIT-negative CRC.In the second experiment,the protein expression of the FHIT and other apoptosis related proteins(Bax,Bcl-2 and Survivin)were measured in human CRC by TMA.Our combined results demonstrate the mutation in the FHIT gene significantly reduced FHIT expression in human CRC.Both TUNEL and TMA experiments demonstrated significantly inhibited apoptosis by down-regulation of Bax and the up-regulation of Survivin and Bcl-2.Collectively,these studies identify the mechanism by which an important tumor suppressor gene,FHIT is inactivated specifically in human CRC contributing to our understanding of the mechanism of colorectal carcinogenesis.CAO Jie CHEN Xiaoping LI Wanglin XIA Jie DU Hong TANG Weibiao CHEN Shanming WANG Hui CHEN Xiwen XIAO Huanqing LI Yuyuan 2007Frontiers of Medicine2007,1,2:0
12Round empty core infiltration and anti-filtration recharge wells and laboratory recharge tests显示文摘Anti-filtration recharge wells are commonly installed in river channels and irrigation canals on the Shandong Peninsula,China,and can be used as an important recharge facility for underground reservoirs.However,during recharge,as the running time increases,the recharge capacity gradually decreases.This study was undertaken to develop a new recharge well and mitigate this deficiency.A round empty core infiltration and antifiltration recharge well and laboratory recharge test equipment were developed.Subsequently,1:25 models of the recharge pond,a round empty core recharge wellhead,and an artificial recharge well were prepared.Using equal recharge levels and ambient groundwater levels,laboratory steady-flow recharge tests on the existing anti-filtration recharge well,the new round empty core infiltration and anti-filtration recharge well,and an artificial recharge well were carried out.Experimental data on the measured groundwater table and single-well recharge quantity were also collected and analyzed.The results showed that compared with the existing anti-filtration recharge well,the new round empty core infiltration and anti-filtration recharge well had stronger anti-deposition and anti-scouring properties,and the single-well recharge quantity increased by 403%.With an increase in the number of recharge tests,the single-well recharge quantity gradually decreased and tended to stabilize as a whole.Yuxi LI Wanglin LI Jiapeng HE Chunhui ZHANG 2020Frontiers of Earth Science2020,14,2:0
