1Optimization of butanol production from corn straw hydrolysate by Clostridium acetobutylicum using response surface method显示文摘Butanol is a new kind of very potential biofuels.Enzymatic hydrolysis of corn stalk was utilized in this study to produce butanol by Clostridium acetobutylicum CICC 8008.Plackett-Burman (P-B) design and Central Composite Design (CCD) were adopted to screen crucial factors during fermentation as well as the optimization of experimental conditions.The result demonstrated that among the seven factors,namely,Yeast extract,(NH 4) 2 SO 4,KH 2 PO 4,MgSO 4,FeSO 4,CuSO 4 and CaCO 3,only CaCO 3 was selected as the most critical factor.The optimization experiment results for CaCO 3 usage,temperature and reaction time by CCD were determined to be 5.04 g/L,35°C and 70 h,respectively.A corresponding mathematical model was established to predict the fermentation experiment and maximum butanol yield of 6.57 g/L was acquired.The result of verification experiment under the optimum conditions showed that 6.20 g/L was the maximum butanol yield.This demonstrated that statistical method was a powerful tool for the optimization of butanol production from enzymatic hydrolysis of corn stalk.LIN YouSheng WANG Jing WANG XuMing SUN XiaoHong 2011Chinese Science Bulletin2011,56,14:15
2Characterization of Distinct T Cell Receptor Repertoires in Tumor and Distant Non-tumor Tissues from Lung Cancer Patients显示文摘T cells and T cell receptors(TCRs)play pivotal roles in adaptive immune responses against tumors.The development of next-generation sequencing technologies has enabled the analysis of the TCRb repertoire usage.Given the scarce investigations on the TCR repertoire in lung cancer tissues,in this study,we analyzed TCRb repertoires in lung cancer tissues and the matched distant non-tumor lung tissues(normal lung tissues)from 15 lung cancer patients.Based on our results,the general distribution of T cell clones was similar between cancer tissues and normal lung tissues;however,the proportion of highly expanded clones was significantly higher in normal lung tissues than in cancer tissues(0.021%±0.002%vs.0.016%±0.001%,P=0.0054,Wilcoxon signed rank test).In addition,a significantly higher TCR diversity was observed in cancer tissues than in normal lung tissues(431.37±305.96 vs.166.20±101.58,P=0.0075,Mann-Whitney U test).Moreover,younger patients had a significantly higher TCR diversity than older patients(640.7±295.3 vs.291.8±233.6,P=0.036,Mann-Whitney U test),and the higher TCR diversity in tumors was significantly associated with worse cancer outcomes.Thus,we provided a comprehensive comparison of the TCR repertoires between cancer tissues and matched normal lung tissues and demonstrated the presence of distinct T cell immune microenvironments in lung cancer patients.Xiang Wang Botao Zhang Yikun Yang Jiawei Zhu Shujun Cheng Yousheng Mao Lin Feng Ting Xiao 2019Genomics, Proteomics & Bioinformatics2019,17,3:5

6Reducing children’s exposure to di(2-ethylhexyl)phthalate in homes and kindergartens in China:Impact on lifetime cancer risks and burden of disease显示文摘Exposure to di(2-ethylhexyl)phthalate(DEHP)in the indoor environment has been linked with significant health risks for Chinese children.Multi-phase DEHP concentrations in Chinese residences and kindergartens were estimated using a mass balance model based on the current baseline condition and control strategies(i.e.,increasing ventilation rate,reducing area of sources,using mechanical ventilation systems,and using portable air cleaners).The health benefits of each control strategy were quantified as the reduction in lifetime cancer risks(LCR)and burden of disease(BoD).In the current situation,the mean LCR and disability-adjusted life years(DALY)number attributable to indoor DEHP exposure for Chinese children were around 6.0×10^(−6) and 155 thousand,respectively.The mean LCR and DALY might be reduced by 25%-54%and 16%-40%,respectively,by increasing air exchange rates by 100%,reducing the use of source materials by two-thirds or deploying commercial air cleaners in naturally ventilated buildings.Meanwhile,avoidable DALYs could result in a reduction of mean economic losses of 2.2-5.3 billion RMB.Mechanical ventilation systems with filtration units may not be helpful for reducing children’s health risks.House-specific and tailor-made control measures are critical in lowering indoor exposure to DEHP to promote sustainable buildings and children’s health in China.Dongsheng Tao Wen Sun Donghui Mo Yonghui Lin Wei Lv Daniel Mmereki Yousheng Xu Yanghui Ye Yuanjun Tang Chao Ye Cong Dong Zhongming Bu 2024Building Simulation2024,17,3:0
7CHCHD2 maintains mitochondrial contact site and cristae organizing system stability and protects against mitochondrial dysfunction in an experimental model of Parkinson’s disease显示文摘Background:Parkinson’s disease(PD)is the second most common neurodegenerative disease after Alzheimer’s dementia.Mitochondrial dysfunction is involved in the pathology of PD.Coiled-coil-helix-coiled-coil-helix domain-containing 2(CHCHD2)was identified as associated with autosomal dominant PD.However,the mechanism of CHCHD2 in PD remains unclear.Methods:Short hairpin RNA(ShRNA)-mediated CHCHD2 knockdown or lentivirus-mediated CHCHD2 overexpression was performed to investigate the impact of CHCHD2 on mitochondrial morphology and function in neuronal tumor cell lines represented with human neuroblastoma(SHSY5Y)and HeLa cells.Blue-native polyacrylamide gel electrophoresis(PAGE)and two-dimensional sodium dodecyl sulfate-PAGE analysis were used to illustrate the role of CHCHD2 in mitochondrial contact site and cristae organizing system(MICOS).Co-immunoprecipitation and immunoblotting were used to address the interaction between CHCHD2 and Mic10.Serotype injection of adeno-associated vector-mediated CHCHD2 and 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)administration were used to examine the influence of CHCHD2 in vivo.Results:We found that the overexpression of CHCHD2 can protect against methyl-4-phenylpyridinium(MPP+)-induced mitochondrial dysfunction and inhibit the loss of dopaminergic neurons in the MPTP-induced mouse model.Furthermore,we identified that CHCHD2 interacted with Mic10,and overexpression of CHCHD2 can protect against MPP+-induced MICOS impairment,while knockdown of CHCHD2 impaired the stability of MICOS.Conclusion:This study indicated that CHCHD2 could interact with Mic10 and maintain the stability of the MICOS complex,which contributes to protecting mitochondrial function in PD.Lin Lu Hengxu Mao Miaomiao Zhou Yuwan Lin Wei Dai Jiewen Qiu Yousheng Xiao Mingshu Mo Xiaoqin Zhu Zhuohua Wu Zhong Pei Wenyuan Guo Pingyi Xu Xiang Chen 2022Chinese Medical Journal2022,,13:0
8Multi-omics analysis reveals RNA splicing alterations and their biological and clinical implications in lung adenocarcinoma显示文摘Alternative RNA splicing is one of the most important mechanisms of posttranscriptional gene regulation,which contributes to protein diversity in eukaryotes.It is well known that RNA splicing dysregulation is a critical mechanism in tumor pathogenesis and the rationale for the promising splice-switching therapeutics for cancer treatment.Although we have a comprehensive understanding of DNA mutations,abnormal gene expression profiles,epigenomics,and proteomics in lung adenocarcinoma(LUAD),little is known about its aberrant alternative splicing profles.Here,based on the multi-omics data generated from over 1000 samples,we systematically studied the RNA splicing alterations in LUAD and revealed their biological and clinical implications.We identifed 3688 aberrant alternative splicing events(AASEs)in LUAD,most of which were alternative promoter and exon skip.The specific regulatory roles of RNA binding proteins,somatic mutations,and DNA methylations on AASEs were comprehensively interrogated.We dissected the functional implications of AASEs and concluded that AASEs mainly affected biological processes related to tumor proliferation and metastasis.We also found that one subtype of LUAD with a particular AASEs pattern was immunogenic and had a better prognosis and response rate to immunotherapy.These findings revealed novel events related to tumorigenesis and tumor immune microenvironment and laid the foundation for the development of splice-switching therapies forLUAD.Quanyou Wu Lin Feng Yaru Wang Yousheng Mao Xuebing Di Kaitai Zhang Shujun Cheng Ting Xiao 2022Signal Transduction and Targeted Therapy2022,7,9:0
