1Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment.Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen 2018Cancer Communications2018,38,1:20

3Inhibitory effect of blue honeysuckle extract on high-fat-diet-induced fatty liver in mice显示文摘Blue honeysuckle is rich in polyphenols, and recently receiving attention because of its potential antioxidant and anti-inflammatory properties. Nonalcoholic fatty liver disease(NAFLD) is the leading cause of chronic liver disease that develops hepatic inflammation and metabolic syndrome. The present study aims to study the effect of blue honeysuckle extract(BHE) on fat deposition and hepatic lipid peroxidation in a high-fat-diet(HFD)-induced mouse model. Mice were fed a normal diet(ND) or a HFD containing 0.5% or 1% of BHE or not for 45 d. Liver sections were stained by hematoxylin-eosin staining.Serum lipids were measured by a clinical analyzer. Insulin was examined by ELISA, and hepatic proteins were detected by Western blotting. Dietary supplementation of BHE dose-dependently suppressed HFD-induced obesity and hepatic fat deposition. 4Dietary protocatechuic acid ameliorates inflammation and up-regulates intestinal tight junction proteins by modulating gut microbiota in LPS-challenged piglets显示文摘Background: Weaning is one of the major factors that cause stress and intestinal disease in piglets. Protocatechuic acid(PCA) is an active plant phenolic acid which exists in Chinese herb, Duzhong(Eucommia ulmoides Oliver), and is also considered as the main bioactive metabolite of polyphenol against oxidative stress and inflammation. This study aimed to investigate the effect of PCA on growth performance, intestinal barrier function, and gut microbiota in a weaned piglet model challenged with lipopolysaccharide(LPS).Methods: Thirty-six piglets(Pig Improvement Company line 337 × C48, 28 d of age, 8.87 kg ± 0.11 kg BW) were randomly allocated into 3 treatments and fed with a basal diet(CTL), a diet added 50 mg/kg of aureomycin(AUR), or a diet supplemented with 4000 mg/kg of PCA, respectively. The piglets were challenged with LPS(10 μg/kg BW) on d 14 and d 21 by intraperitoneal injection during the 21-d experiment. Animals(n = 6 from each group) were sacrificed after being anesthetized by sodium pentobarbital at 2 h after the last injection of LPS. The serum was collected for antioxidant indices and inflammatory cytokines analysis, the ileum was harvested for detecting mRNA and protein levels of tight junction proteins by PCR and immunohistochemical staining, and the cecum chyme was collected for intestinal flora analysis using 16 S rRNA gene sequencing.Results: Dietary supplementation of PCA or AUR significantly increased the expression of tight junction proteins including ZO-1 and claudin-1 in intestinal mucosa, and decreased the serum levels of thiobarbituric acid reactive substances(TBARS) and IL-6, as compared with CTL group. In addition, PCA also decreased the serum levels of IL-2 and TNF-α(P < 0.05). Analysis of gut microbiota indicated that PCA increased the Firmicutes/Bacteroidetes ratio(P < 0.05). 6ζ-Carotene Isomerase Suppresses Tillering in Rice through the Coordinated Biosynthesis of Strigolactone and Abscisic Acid显示文摘Rice tillering is an important agronomic trait affecting grain yield.Here,we identified a high-tillering mutant tillering20(t20),which could be restored to the wild type by treatment with the strigolactone(SL)analog rac-GR24.T20 encodes a chloroplast ζ-carotene isomerase(Z-ISO),which is involved in the biosynthesis of ca-rotenoids and their metabolites,SL and abscisic acid(ABA).The t20 mutant has reduced SL and ABA,raising the question of how SL and ABA biosynthesis is coordinated,and whether they have overlapping functions in tillering.We discovered that rac-GR24 stimulated T20 expression and enhanced all-trans-p-carotene biosynthesis.Importantly,rac-GR24 also stimulated expression of Oryza sativa 9-CIS-EPOXY-CAROTENOID DIOXYGENASE 1(OsNCED1)through induction of Oryza sativa HOMEOBOX12(0sHOX12),promoting ABA biosynthesis in shoot base.On the other hand,ABA treatment significantly repressed SL biosynthesis and the ABA biosynthetic mutants displayed elevated SL biosynthesis.ABA treatment reduced the number of basal tillers in both t20 and wild-type plants.Furthermore,while ABA-deficient mu-tants aba1 and aba2 had the same number of basal tillers as wild type,they had more unproductive upper tillers at maturity.This work demonstrates complex interactions in the biosynthesis of carotenoid,SLs and ABA,and reveals a role for ABA in the regulation of rice tillering.Xue Liu Qingliang Hu Jijun Yan Kai Sun Yan Liang Meiru Jia Xiangbing Meng Shuang Fang Yiqin Wang Yanhui Jing Guifu Liu Dianxing Wu Chengcai Chu Steven M.Smith Jinfang Chu Yonghong Wang Jiayang Li Bing Wang 2020Molecular Plant2020,13,12:11
7Reservoir Characteristics of Lacustrine Shale and Marine Shale:Examples from the Songliao Basin,Bohai Bay Basin and Qiannan Depression显示文摘Lacustrine shale from the Qingshankou Formatin of Songliao basin and the Shahejie Formation of Bohai Bay basin, and marine shale from the lower Cambrian Jiumenchong Formation of Qiannan depression were analysed by using rock pyrolysis, TOC(total organic carbon), XRD(X-ray diffraction), SEM(scanning electron microscope), FE-SEM(field emission scanning electron microscope), high pressure mercury intrusion, and low pressure N_2 and CO_2 gas adsorption experiments, in aim to reveal their reservoir features. The results show that:(1) the width of micro-pores of all the studied samples mainly ranges from 0.45 to 0.7 nm indicated by CO_2 isotherms, and the width of meso-pores is less than 10 nm, with type IV adsorption isotherms and type H2 hysteresis loop, indicative of 'ink-bottle'-shaped pores. Good correlations exist among pore volume, surface area and averaged pore diameter, and a good positive correlation exists between micro-pore volume and TOC content; however, there is no obvious correlation between meso-pore volume and TOC content;(2) interparticle pores, pores among the edge of mineral grains and organic matter pores were all identified in marine and lacustrine shale, among which the interparticle pores may be influence by dissolution effect. Not all bitumen develops organic matter pore, and only high to over mature bitumen present pores. Now the description methods of micrometer scale pores developed in shale are very lack. 8Terrace dating as an archive of the run-through of the Sanmen Gorges显示文摘Analysis of morphological properties in the Sanmen Gorges region of the Yellow River shows that four river terraces have been developed at the entrance to the gorges. Commonly, thick aeolian loess-paleosol sequence was deposited on the terraces, which makes it easy to date the terraces. The dating of paleomagnetic, loess-paleosol sequence matching and thermoluminescence show that terraces T2, T3 and T4 were formed 0. 129 Ma, 0. 625 Ma and 0. 865 Ma ago, respectively. The formation of these terraces in this region indicates that the Yellow River has experienced four intensive downcutting events during the last 0.9 Ma. 10Microbial reduction of uranium(Ⅵ)by Bacillus sp.Dwc-2:A macroscopic and spectroscopic study显示文摘The microbial reduction of U(VI) by Bacillus sp. dwc-2, isolated from soil in Southwest China,was explored using transmission electron microscopy(TEM), X-ray photoelectron spectroscopy(XPS) and X-ray absorption near edge spectroscopy(XANES). Our studies indicated that approximately 16.0% of U(VI) at an initial concentration of 100 mg/L uranium nitrate could be reduced by Bacillus sp. dwc-2 at pH 8.2 under anaerobic conditions at room temperature.Additionally, natural organic matter(NOM) played an important role in enhancing the bioreduction of U(VI) by Bacillus sp. dwc-2. XPS results demonstrated that the uranium presented mixed valence states(U(VI) and U(IV)) after bioreduction, which was subsequently confirmed by XANES. 11Dating and genesis of the upper Weihe River terraces in Longxi basin,China显示文摘Seven terraces along the Weihe River in Longxi basin have been investigated. These terraces all consist of archetypal duality in structure, namely the floodplain deposits and the layer of gravels overlaid by varying thickness of the loess. By resorting to various dating approaches such as palaeomagnetic, optically stimulated luminescenece (OSL), 14C and loess-paleosol sequence, we provide preliminary timing of these seven terraces along upper Weihe River. Analysis on sedimentation characteristics and dating of these terraces showed that seven terraces may be jointly generated by tectonic uplifts and climatic changes. Tectonic uplifts may accommodate initial river incision, and climatic change may be responsible for processes of subsequent channel widening and aggradation. Aggradation normally occurs during glacial periods, in contrast to down-cuttings during transitions from glacial to interglacial period. Moreover, on the basis of the timings and heights of these terraces, we infer that the long-term rate of river incision was determined to be 0.2 m/kyr during the last 870 kyr, which differs from other river incision rates. 13Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 14Bioaccumulation characterization of uranium by a novel Streptomyces sporoverrucosus dwc-3显示文摘The biosorption mechanisms of uranium on an aerobic bacterial strain Streptomyces sporoverrucosus dwc-3,isolated from a potential disposal site for(ultra-)low uraniferous radioactive waste in Southwest China,were evaluated by using transmission electron microscopy(TEM),energy dispersive X-ray(EDX)analysis,Fourier transform infrared spectroscopy(FT-IR),X-ray photoelectron spectroscopy(XPS),proton induced X-ray emission(PIXE)and enhanced proton backscattering spectrometry(EPBS).Approximately60% of total uranium at an initial concentration of 10 mg/L uranium nitrate solution could be absorbed on 100 mg S.sporoverrucosus dwc-3 with an adsorption capacity of more than3.0 mg/g(wet weight)after 12 hr at room temperature at p H 3.0.The dynamic biosorption process of S.sporoverrucosus dwc-3 for uranyl ions was well described by a pseudo second-order model.S.sporoverrucosus dwc-3 could accumulate uranium on cell walls and within the cell,as revealed by SEM and TEM analysis as well as EDX spectra.XPS and FT-IR analysis further suggested that the absorbed uranium was bound to amino,phosphate and carboxyl groups of the cells.Additionally,PIXE and EPBS results confirmed that ion exchange also contributed to the adsorption process of uranium.Xiaolong Li Congcong Ding Jiali Liao Liang Du Qun Sun Jijun Yang Yuanyou Yang Dong Zhang Jun Tang Ning Liu 2016Journal of Environmental Sciences2016,28,3:5

18Molecular characterization of RAPD and SCAR marker linked to the frog-eye leaf spot resistance gene in soybean显示文摘Two fragments SCS362o and SCS3580 of the co-dominant marker OPS0362o&580 that were linked to the resistance gene of soybean frog-eye leaf spot have been completely sequenced. A significant insertion of 30 bp is the main reason of the polymorphism between the two fragments. The results of Southern hybridization indicate that SCS362o derives from a single- or low-copy sequence and can be used as an RFLP probe. A co-dominant SCAR marker SCSSeao&sao has been developed based on the sequences. The segregation of SCS362o&58o is similar to that of RAPD marker OPS03620&580. Significant polymorphism has been shown between resistant and susceptible genotypes when 62 soybean genotypes were surveyed for the SCAR marker. 19Quantum conductance of armchair carbon nanocoils: roles of geometry effects显示文摘Armchair carbon nanocoils (CNCs) with different geometric parameters are constructed and optimized using a tight-binding (TB) total energy model. The quantum conductance of these nanocoils is simulated employing a π-orbital TB model incorporated with the non-equilibrium Green's function theory. Compared with the perfect armchair carbon nanotubes (CNTs) and armchair CNTs with only Stone-Wales (SW) defects, the quantum conductance spectra of the armchair CNCs present distinct gaps around the Fermi level, which are mainly originated from the existence of sp3 carbon in the three-dimensional spiral structures. 20A fast,edge-preserving,distance-regularized model with bilateral filtering for oil spill segmentation of SAR images显示文摘Marine oil spills are among the most significant sources of marine pollution.Synthetic aperture radar(SAR)has been used to improve oil spill observations because of its advantages in oil spill detection and identification.However,speckle noise,weak boundaries,and intensity inhomogeneity often exist in the oil spill regions of SAR imagery,which will seriously aff ect the accurate identification of oil spills.To enhance marine oil spill segmentation of SAR images,a fast,edge-preserving framework based on the distance-regularized level set evolution(DRLSE)model was proposed.Specifically,a bilateral filter penalty term is designed and incorporated into the DRLSE energy function(BF-DRLSE)to preserve the edges of oil spills,and an adaptive initial box boundary was selected for the DRLSE model to reduce the operation time complexity.Two sets of RadarSat-2 SAR data were used to test the proposed method.The experimental results indicate that the bilateral filtering scheme incorporated into the energy function during level set evolution improved the stability of level set evolution.Compared with other methods,the proposed improved BF-DRLSE algorithm displayed a higher overall segmentation accuracy(97.83%).In addition,using an appropriate initial box boundary for the DRLSE method accelerated the global search process,improved the accuracy of oil spill segmentation,and reduced computational time.Therefore,the results suggest that the proposed framework is eff ective and applicable for marine oil spill segmentation.Wandi WANG Hui SHENG Yanlong CHEN Shanwei LIU Jijun MAO Zhe ZENG Jianhua WAN 2021Journal of Oceanology and Limnology2021,39,4:3
