| 1 | SARS-CoV-2 infection aggravates chronic comorbidities of cardiovascular diseases and diabetes in mice显示文摘Background:Cardiovascular diseases(CVDs)and diabetes mellitus(DM)are top two chronic comorbidities that increase the severity and mortality of COVID-19.However,how SARS-CoV-2 alters the progression of chronic diseases remain unclear.Methods:We used adenovirus to deliver h-ACE2 to lung to enable SARS-CoV-2 infection in mice.SARS-CoV-2’s impacts on pathogenesis of chronic diseases were studied through histopathological,virologic and molecular biology analysis.Results:Pre-existing CVDs resulted in viral invasion,ROS elevation and activation of apoptosis pathways contribute myocardial injury during SARS-CoV-2 infection.Viral infection increased fasting blood glucose and reduced insulin response in DM model.Bone mineral density decreased shortly after infection,which associated with impaired PI3K/AKT/mTOR signaling.Conclusion:We established mouse models mimicked the complex pathological symptoms of COVID-19 patients with chronic diseases.Pre-existing diseases could impair the inflammatory responses to SARS-CoV-2 infection,which further aggravated the pre-existing diseases.This work provided valuable information to better understand the interplay between the primary diseases and SARS-CoV-2 infection. | Yuanwu Ma Dan Lu Linlin Bao Yajin Qu Jiangning Liu Xiaolong Qi Lei Yu Xu Zhang Feifei Qi Qi Lv Yunpeng Liu Xudong Shi Caixian Sun Jing Li Jie Wang Yunlin Han Kai Gao Wei Dong Ning Liu Shan Gao Jing Xue Qiang Wei Sidan Pan Hong Gao Lianfeng Zhang Chuan Qin | 2021 | Animal Models and Experimental Medicine2021,4,1: | 3 |

| 3 | The protection of CoronaVac against the infection of wild-type SARS-CoV-2(WH-09)or Omicron variant in nude-hACE2 mice显示文摘Background:Immunocompromised individuals have an increased risk of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection and severe outcomes,but we pay less attention to these people.Athymic nude mice are a murine strain with a spontaneous deficiency of the Foxn1 gene,which can result in thymic degeneration or its absence,leading to immunosuppression and a decrease in the number of T cells,and are widely used in preclinical evaluations of disease in immunocompromised populations.Methods:We investigated the protection of the coronavirus disease 2019(COVID-19)inactivated vaccine(CoronaVac)against the infection of wild-type SARS-CoV-2(WH-09)or Omicron variant utilizing a hybrid-type nude-hACE2 mouse model.Results:Compared with nude-hACE2/W mice,the viral load in the brain and lung tissue of nude-hACE2 mice(nude-hACE2/WV)infected with WH-09 after vaccination significantly decreased,and the histopathological changes were also reduced.The viral load in the brain and lung tissue of nude-hACE2 mice(nude-hACE2/OV)infected with the Omicron variant after vaccination was lower than that in nude-hACE2/O,but histopathological symptoms did not improve significantly.Conclusion:CoronaVac provides some protection against infection of both WH-09 and the Omicron variant in the nude-hACE2 mice.Our findings aimed to provide a reference for vaccination against SARS-CoV-2 in immunocompromised populations. | Kaili Lin Meixuan Liu Lu Sun Hanjun Fu Hongwei Qiao Shunyi Wang Sidan Pan Hong Gao | 2023 | Animal Models and Experimental Medicine2023,6,4: | 0 |

| 5 | Polydiacetylene-based poly-ion complex enabling aggregation-induced emission and photodynamic therapy dual turn-on for on-demand pathogenic bacteria elimination显示文摘Poly-ion complex(PIC)integrating non-antibiotic theranostics holds great promise in the combat against drug-resistant bacteria.Photosensitizers with aggregation-induced emission(AIE)characteristic are particularly intriguing theranostic agents,but incorporating them into antibacterial PIC to enable both fluorescence and reactive oxygen species(ROS)generation turn-on is deemed a great challenge.Here we report the development of a PIC that can dually boost the fluorescence and ROS generation in the presence of pathogen bacteria.The PIC is constructed based on an anionic polydiacetylene poly(deca-4,6-diynedioic acid)(PDDA),which completely degrades in the presence of ROS.A cationic polymer quaternized poly(2-(dimethylamino)ethyl methacrylate)(PQDMA)that can disrupt bacterial membrane is co-loaded together with a highly efficient AIE photosensitizer TPCI in the PIC.PIC is nonfluorescent initially in that PDDA can quench the AIE of TPCI in PIC.When pathogenic bacteria are present,they can disturb the assembly of PIC to release TPCI,whose fluorescence turns on sensitively to indicate the existence of bacteria.The on-demand irradiation can be subsequently applied to excite TPCI,which generates ROS to degrade PDDA and deform the PIC.As a result,TPCI and PQDMA are completely released to eliminate bacteria through a synergy of turned-on photodynamic therapy(PDT)and membrane disruption.The highly efficient detection and inhibition against both Gramnegative and Gram-positive bacteria have validated this polydiacetylene-based PIC system as an effective non-antibiotic antibacterial theranostic platform as well as a new strategy to enable“turn-on”fluorescence sensing and imaging of AIE fluorophores. | Sidan Tian Yuan Lu Zhenyan He Qiang Yue Zhiyong Zhuang Yingzhou Wang Fanling Meng Liang Luo | 2022 | Science China Chemistry2022,65,9: | 0 |
