1In vitro Inhibitory Activity of Shisandra chinensis and Polygonatum sibiricum against Vibrio harveyi and Its Biofilms显示文摘[Objective] This study aimed to analyze the in vitro inhibitory activity of Shisandra chinensis and Polygonatum sibiricum against Vibrio harveyi and its biofilms. [Result] By agar diffusion test,in vitro inhibitory activity of S.chinensis and P. sibiricum against V. harveyi was investigated. The minimal inhibitory concentration(MIC) a nd minimal bactericidal concentration(MBC) of S.chinensis and P.sibiricum against V.harveyi were determined by doubling dilution method. The inhibitory activity of S. chinensis and P.sibiricum on the formation of V.harveyi biofilms was evaluated by modified MTT assay. [Result]Both S.chinensis and P.sibiricum had inhibitory activity against V.harveyi. The inhibition zone diameter of S.chinensis against V.harveyi was 17.95 mm; MIC and MBC of S.chinensis were both 3.125 mg / ml. The inhibition zone diameter of P. sibiricum against V.harveyi was 12.22 mm; MIC and MBC of P.sibiricum were 3.125 and 6. 250 mg / ml,respectively. When the concentration was higher than 6. 25 mg / ml,S.chinensis decoction had extremely significant inhibitory activity against V.harveyi( P < 0. 01); when the concentration was higher than 3. 125 mg / ml,P. sibiricum had extremely significant inhibitory activity against V. harveyi(P <0. 01). 3Seamless and early gap healing of osteochondral defects by autologous mosaicplasty combined with bioactive supramolecular nanofiber-enabled gelatin methacryloyl(BSN-GelMA)hydrogel显示文摘Autologous mosaicplasty is a common approach used to treat osteochondral defects in clinical practice.Gap integration between host and transplanted plugs requires bone tissue reservation and hyaline cartilage regeneration without uneven surface,graft necrosis and sclerosis.However,poor gap integration is a serious concern,which eventually leads to deterioration of joint function.To deal with such complications,this study has developed a strategy to effectively enhance integration of the gap region following mosaicplasty by applying injectable bioactive supramolecular nanofiber-enabled gelatin methacryloyl(GelMA)hydrogel(BSN-GelMA).A rabbit osteochondral defect model demonstrated that BSN-GelMA achieved seamless osteochondral healing in the gap region between plugs of osteochondral defects following mosaicplasty,as early as six weeks.Moreover,the International Cartilage Repair Society score,histology score,glycosaminoglycan content,subchondral bone volume,and collagen II expression were observed to be the highest in the gap region of BSN-GelMA treated group.This improved outcome was due to bio-interactive materials,which acted as tissue fillers to bridge the gap,prevent cartilage degeneration,and promote graft survival and migration of bone marrow mesenchymal stem cells by releasing bioactive supramolecular nanofibers from the GelMA hydrogel.This study provides a powerful and applicable approach to improve gap integration after autologous mosaicplasty.It is also a promising off-the-shelf bioactive material for cell-free in situ tissue regeneration.Hongwei Wu Yuna Shang Wei Sun Xinyi Ouyang Wenyan Zhou Jieji Lu Shuhui Yang Wei Wei Xudong Yao g Xiaozhao Wang Xianzhu Zhang Yishan Chen Qiulin He Zhimou Yang Hongwei Ouyang 2023Bioactive Materials2023,,1:1
4Molecular Cloning and Bioinformatics Analysis of kdpE Gene from Vibrio alginolyticus显示文摘The two-component regulatory system plays an important role in the growth and virulence of bacteria.In this study,the phosphate transcriptional regulatory protein (KdpE) gene of Vibrio alginolyticus strain HY9901 was cloned.Sequence analysis revealed that the length of kdp E gene is 732 bp and encodes a putative protein of 243 amino acids.The predicted molecular weight (MW) of Kdp E was 27.66 k D with an estimated p I of 5.01.Using SignalP 4.0 and TMHMM Server 2.0 software,it was predicted that the Kdp E protein was located in cytoplasm.It did not contain a signal peptide or a transmembranous region.A phylogenetic tree was constructed by MEGA 5.0 software,indicating that the Kdp E of V.alginolyticus had high genetic relationship with Vibrio campbellii and Vibrio parahaemolyticus.Using SWISS-MODEL work-space,the three-dimensional structures of conserved domain REC in the Kdp E was determined.These results can provide a basis for further studies on the two-component regulatory systems of V.alginolyticus.Hong Shunhua Yang Fan Pang Huanying Zhang Yanfei Song Dawei Wang Zhiwen Lu Yishan Jian Jichang 2016Animal Husbandry and Feed Science2016,8,5:0
5Characterization and Expression Analysis of B Cell Receptor Accessory Molecule CD79 Gene in Humphead Snapper(Lutjanus sanguineus)显示文摘CD79,a key component of the B cell antigen receptor complex,is composed of CD79α(Igα) and CD79β(Igβ) encoded by mb-1 and B29 respectively,and plays an important role in B cell signaling.In this study,we isolated and characterized mb-1 and B29 from humphead snapper(Lutjanus sanguineus).Their tissue distribution and expression profiles after stimulations in vitro and in vivo were also investigated.The humphead snapper mb-1 and B29 contain open reading frames of 684 bp and 606 bp,encoding 227 amino acids and 201 amino acids,respectively.Both CD79α and CD79β possess signal peptide,extracellular Ig domain,transmembrane region and immunoreceptor tyrosine kinase activation motif(ITAM).Mb-1 is highly expressed in lymphoid organs(thymus,posterior kidney and spleen) and mucosal-associated lymphoid tissues(gill and intestine),while B29 is mainly detected in posterior kidney,spleen,gill and skin.Furthermore,transcription of mb-1 and B29 in head kidney leucocytes was up-regulated following lipopolysaccharide(LPS),pokeweed mitogen(PWM),and polyinosinic-polycytidylic acid(Poly I:C) stimulation,respectively,and their expression level in anterior kidney and spleen was also increased after challenged with formalin-inactived Vibrio harveyi.These results indicated that humphead snapper CD79 molecule might play an important role in immune response to pathogen infection.HUANG Yucong YANXiuying CAI Shuanghu CAI Jia JIAN Jichang LU Yishan 3,4TANG Jufen WU Zaohe 2016Journal of Ocean University of China2016,15,2:0
