| 1 | Naringin ameliorates acetic acid induced colitis through modulation of endogenous oxido-nitrosative balance and DNA damage in rats显示文摘The aim of this study was to evaluate the effect of naringin on experimentally induced inflammatory bowel disease in rats.Naringin(20,40 and 80 mg/kg) was given orally for 7 days to Wistar rats before induction of colitis by intrarectal instillation of 2 mL of 4%(v/v) acetic acid solution.The degree of colonic mucosal damage was analyzed by examining mucosal damage,ulcer area,ulcer index and stool consistency.Intrarectal administration of 4%acetic acid resulted in significant modulation of serum alkaline phosphatase,lactate dehydrogenase,superoxide dismutase(SOD),glutathione(GSH),malondialdehyde(MDA) and myeloperoxidase(MPO) content along with colonic nitric oxide(NO),xanthine oxidase(XO) level and protein carbonyl content in the colonic tissue as well as in blood.Naringin(40 and 80 mg/kg) exerted a dose dependent(P < 0.05) ameliorative effect,as it significantly increased hematological parameter as well as colonic SOD and GSH.There was a significant(P < 0.05)and dose dependant inhibition of macroscopical score,ulcer area along with colonic MDA,MPO activity by the7 days of pretreatment of naringin(40 and 80 mg/kg).Biochemical studies revealed a significant(P < 0.05) dose dependant inhibition in serum alkaline phosphatase(ALP) and lactate dehydrogenase(LDH) levels by pretreatment of naringin.Increased levels of colonic NO,XO,protein carbonyl content and DNA damage were also significantly decreased by naringin pretreatment.The findings of the present investigation propose that naringin has an anti-inflammatory,anti-oxidant and anti-apoptotic potential effect at colorectal sites as it modulates the production and expression of oxidative mediators such as MDA,MPO,NO and XO,thus reducing DNA damage. | Venkatashivam Shiva Kumar Anuchandra Ramchandra Rajmane Mohammad Adil Amit Dattatraya Kandhare Pinaki Ghosh Subhash Laxman Bodhankar | 2014 | The Journal of Biomedical Research2014,28,2: | 8 |
| 2 | Spectroscopic analysis on the binding interaction of biologically active pyrimidine derivative with bovine serum albumin显示文摘A biologically active antibacterial reagent, 2–amino-6-hydroxy–4–(4-N, N-dimethylaminophenyl)-pyrimidine-5-carbonitrile(AHDMAPPC), was synthesized. It was employed to investigate the binding interaction with the bovine serum albumin(BSA) in detail using different spectroscopic methods. It exhibited antibacterial activity against Escherichia coli and Staphylococcus aureus which are common food poisoning bacteria. The experimental results showed that the fluorescence quenching of model carrier protein BSA by AHDMAPPC was due to static quenching. The site binding constants and number of binding sites(n E 1) were determined at three different temperatures based on fluorescence quenching results. The thermodynamic parameters, enthalpy change(ΔH), free energy(ΔG) and entropy change(ΔS) for the reaction were calculated to be 15.15 k J/mol, –36.11 k J/mol and 51.26 J/mol K according to van't Hoff equation, respectively. The results indicated that the reaction was an endothermic and spontaneous process, and hydrophobic interactions played a major role in the binding between drug and BSA. The distance between donor and acceptor is 2.79 nm according to F?rster's theory. The alterations of the BSA secondary structure in the presence of AHDMAPPC were confirmed by UV–visible, synchronous fluorescence, circular dichroism(CD) and three-dimensional fluorescence spectra. All these results indicated that AHDMAPPC can bind to BSA and be effectively transported and eliminated in the body. It can be a useful guideline for further drug design. | Vishwas D. | 4 | Effects of Simvastatin on NF-κB-DNA Binding Activity and Monocyte Chemoattractant Protein-1 Expression in a Rabbit Model of Atherosclerosis显示文摘To observe the effects of simvastatin on nuclear factor kappaB (NF-κB)-DNA binding activity and on the expression of monocyte chemoattractant protein-1 (MCP-1) in atherosclerotic plaque in rabbits and to explore the anti-atherosclerotic properties beyond its lipid-lowering effects. Thirty-six New Zealand male rabbits were randomly divided into low-cholesterol group (LC), high-cholesterol group (HC), high-cholesterol+simvastatin group (HC+S) and then were fed for 12 weeks. At the end of the experiment, standard enzymatic assays, electrophoretic mobility shift assay (EMSA), immunohistochemical staining, and morphometry were performed to observe serum lipids, NF-κB-DNA binding activity, MCP-1 protein expression, intima thickness and plaque area of aorta respectively in all three groups. Our results showed that the serum lipids, NF-κB-DNA binding activity, expression of MCP-1 protein, intima thickness, and plaque area of aorta in the LC and HC+S groups were significantly lower than those in the HC group (P<0.05). There was no significant difference in the serum lipids between the LC and HC+S groups (P>0.05), but the NF-κB-DNA binding activity, the expression of MCP-1 protein and the intima thickness and plaque area of aorta in the HC+S group were significantly decreased as compared to the LC group (P<0.05). | 6 | 基因分型指导华法林预防和治疗血栓栓塞性疾病有效性与安全性的系统评价显示文摘目的:本文旨在应用Meta分析的方法评价基因分型指导华法林预防和治疗血栓栓塞性疾病的有效性与安全性。方法:计算机系统检索Pub Med、Cochrane图书馆、万方、维普(VIP)、中国知网(CNKI)等数据库有关基因分型指导华法林预防和治疗血栓栓塞性疾病的文献,检索日期由建库截止2014年1月1日。由2位作者独立进行数据提取和文献质量方法学评价。应用Rev Man5.2进行统计分析。结果:共有11项研究最终被纳入系统评价,最终对10项随机对照研究(2 905名受试者)进行定量分析。Meta分析结果显示:基因分型组与对照组比较,在国际标准化比值(international normalized ratio,INR)处于治疗范围的时间百分比(SMD=0.11,95%CI=-0.06~0.29,P=0.21),稳定剂量达标率[相对危险度(relative risk,RR)=1.13;95%CI=1.00~1.28,P=0.05],抗凝过度(RR=0.95,95%CI=0.83~1.08,P=0.44),出血事件(RR=0.89,95%CI=0.78~1.01,P=0.08),栓塞事件(RR=0.61,95%CI=0.30~1.23,P=0.17)等方面,2组之间的差异均无统计学意义;对INR处于治疗范围的时间百分比和稳定剂量达标率2方面进行亚组分析,结果提示2组之间仍未见统计学差异。对结局指标进行敏感性分析显示结果稳定可靠。结论:基因分型指导华法林预防和治疗血栓栓塞性疾病较对照方案在有效性与安全性方面的优势性证据仍不足。 | 王建灵 Gyawali Laxman 李丹 熊斌 苏立 | 2014 | 重庆医科大学学报2014,39,10: | 2 |
| 7 | Applications of snake venoms in treatment of cancer显示文摘Snake venoms are folk medicines used since ages. The components of snake venoms have high specific affinity and actions on cells and cell components. Also snake venoms are largely cytotoxic to tumor cells than normal cells. In addition to these, they have several therapeutic actions that make them an attractive option in the management of cancer. The advent of modern technologies has greatly helped in extracting and identifying new components of therapeutic interests in short time. The article highlights the importance of snake venoms in the management of cancer, so as to motivate curious researchers to devote their skills in this fascinating area. | 10 | Eosinophilic enteritis presenting as a rare cause for ileo-ileal intussusception显示文摘Eosinophilic enteritis,a relatively rare entity,usually involves gastric antrum or proximal small bowel. Our case is rarer in its involvement of the distal small bowel and presents unusually as intussusception. The disease if diagnosed in the initial stages responds well to medical treatment but if associated with complications or misdiagnosed,surgical modality is the treatment of choice. In our case,the patient presented with acute intestinal obstruction due to intussusception and emergency laparotomy with ileoileal anastomosis was done. Histopathology confirmed the diagnosis as eosinophilic enteritis. | 19 | Corrosion behavior of ultrafine-grained AA2024 aluminum alloy produced by cryorolling显示文摘The objectives of this study were to produce ultrafine-grained(UFG) AA2024 aluminum alloy by cryorolling followed by aging and to evaluate its corrosion behavior. Solutionized samples were cryorolled to ~85% reduction in thickness. Subsequent aging resulted in a UFG structure with finer precipitates of Al_2CuMg in the cryorolled alloy. The(1) solutionized and(2) solutionized and cryorolled samples were uniformly aged at 160°C/24 h and were designated as CGPA and CRPA, respectively; these samples were subsequently subjected to corrosion studies. Potentiodynamic polarization studies in 3.5 wt% NaCl solution indicated an increase in corrosion potential and a decrease in corrosion current density for CRPA compared to CGPA. In the case of CRPA, electrochemical impedance spectroscopic studies indicated the presence of two complex passive oxide layers with a higher charge transfer resistance and lower mass loss during intergranular corrosion tests. The improved corrosion resistance of CRPA was mainly attributed to its UFG structure, uniform distribution of fine precipitates, and absence of coarse grain-boundary precipitation and associated precipitate-free zones as compared with the CGPA alloy. | P.
