1Microstructure Characteristics of Inconel 625 Superalloy Manufactured by Selective Laser Melting显示文摘Selective laser melting(SLM), an additive manufacturing process, is capable of manufacturing metallic parts with complex shapes directly from computer-aided design(CAD) models. SLM parts are created on a layer-by-layer manner, making it more flexible than traditional material processing techniques. In this paper, Inconel 625 alloy, a widely used material in the aerospace industry, were chosen as the build material. Scanning electron microscopy(SEM), electron back scattering diffraction(EBSD) and X-ray diffraction(XRD) analysis techniques were employed to analyze its microstructure. It was observed that the molten pool was composed of elongated columnar crystal. Due to the rapid cooling speed, the primary dendrite arm space was approximately 0.5 μm and the hardness of SLM state was very high(343 HV). The inverse pole figure(IPF) indicated that the growing orientation of the most grains was<001> due to the epitaxial growth and heat conduction. The XRD results revealed that the austenite structure with large lattice distortion was fully formed. No carbides or precipitated phases were found.After heat treatment the grains grew into two microstructures with distinct morphological characters,namely, rectangular grains and limited in the molten pool, and equiaxed grains along the molten boundaries. Upon experiencing the heat treatment, MC carbides with triangular shapes gradually precipitated. The results also identified that a large number of zigzag grain boundaries were formed. 2Development of genome-wide insertion/deletion markers in rice based on graphic pipeline platform显示文摘DNA markers play important roles in plant breeding and genetics.The Insertion/Deletion(InDel) marker is one kind of co-dominant DNA markers widely used due to its low cost and high precision.However,the canonical way of searching for InDel markers is time-consuming and laborintensive.We developed an end-to-end computational solution(InDel Markers Development Platform,IMDP) to identify genome-wide InDel markers under a graphic pipeline environment.IMDP constitutes assembled genome sequences alignment pipeline(AGA-pipe) and next-generation resequencing data mapping pipeline(NGS-pipe).With AGA-pipe we are able to identify 12,944 markers between the genome of rice cultivars Nipponbare and 93-11.Using NGS-pipe,we reported 34,794 InDels from re-sequencing data of rice cultivars Wu-Yun-Geng7 and Guang-Lu-Ai4.Combining AGApipe and NGS-pipe,we developed 205,659 InDels in eight japonica and nine indica cultivars and 2,681 InDels showed a subgroup-specific pattern.Polymerase chain reaction(PCR)analysis of subgroup-specific markers indicated that the precision reached 90%(86 of 95).Finally,to make them available to the public,we have integrated the InDels/markers information into a website(Rice InDel Marker Database,RIMD,http://gffzz71b3756633b74a6ehvu6bxbcwv0vw6vxb.ffgz.tsg.suse.edu.cn/).The application of IMDP in rice will facilitate efficiency for development of genome-wide InDel markers,in addition it can be used in other species with reference genome sequences and NGS data.Yang L Xiao Cui Rui Li Piaopiao Huang Jie Zong Danqing Yao Gang Li Dabing Zhang Zheng Yuan 2015Journal of Integrative Plant Biology2015,57,11:9
3Synthesis and biological evaluation of 12-N-p-chlorobenzyl sophoridinol derivatives as a novel family of anticancer agents显示文摘Taking 12-N-p-chlorobenzyl sophoridinol 2 as a lead, a series of novel sophoridinic derivatives with various 30-substituents at the 11–side chain were synthesized and evaluated for their anticancer activity from sophoridine(1), a natural antitumor medicine. Among them, the sophoridinic ketones 5a–b, alkenes 7a–b and sophoridinic amines 14a–b displayed reasonable antiproliferative activity with IC50 values ranging from 3.8 to 5.4 μmol/L. Especially, compounds 5a and 7b exhibited an equipotency in both adriamycin(AMD)-susceptible and resistant MCF-7 breast carcinoma cells,indicating a different mechanism from AMD. The primary mechanism of action of 5a was to arrest the cell cycle at the G0/G1 phase, consistent with that of parent compound 1. 4Synthesis and biological evaluation of 8-substituted berberine derivatives as novel anti-mycobacterial agents显示文摘Tuberculosis(TB)is a disease which kills two million people every year and infects over one-third of the world's population.Eighteen new 8-substituted berberine derivatives were synthesized and evaluated for their anti-mycobacterial activities against M ycobacterium tuberculosis(M.tuberculosis)strain H_(37)Rv.Among these compounds,compound 6i was the most effective antitubercular agent with an MIC of 1.0μg/mL.Most importantly,compound 6i also exhibited a potent effect against clinically isolated rifampicin-and isoniazid-resistant M.tuberculosis strains,suggesting a different mode of action from the current drugs.Therefore,it shows potential for the development of new anti-TB agents.Yanxiang Wang Haigen Fu Yinghong Li Jiandong Jiang Danqing Song 2012Acta Pharmaceutica Sinica B2012,2,6:4
5Synthesis and biological evaluation of sophocarpinic acid derivatives as anti-HCV agents显示文摘Chronic hepatitis C virus(HCV)infection has become a major public health burden worldwide.Twenty-two sophocarpinic acid or matrine derivatives were synthesized and their anti-HCV activities were evaluated in vitro.The structure-activity analysis revealed that(i)sophocarpinic acids with a D-seco 3-ring structure scaffold were more favorable than matrines with a 4-ring scaffold;(ii)the introduction of an electron-withdrawing group on the phenyl ring in 12-N-benzenesulfonylΔβγsophocarpinic acids was beneficial for the antiviral activity against HCV.Among them,compounds 9h and 9j exhibited the most potent inhibitory activities on HCV replication with selectivity indies of 70.3 and 30.9,respectively.Therefore,both were selected as antiviral candidates for further investigation.Yinghong Li Zonggen Peng Limei Gao Danqing Song 2014Acta Pharmaceutica Sinica B2014,4,4:4
6Settlement and metamorphosis of Styela canopus Savigny larvae in response to some neurotransmitters and thyroxin显示文摘The larvae of ascidian Styela canopus Savigny were treated with epinephrine, norepinephrine, L-DOPA, GABA and thyroxin to test the ability of these compounds to induce or inhibit larval settlement and metamorphosis. The results showed that epinephrine, norepinephrine and L-DOPA at the concentration of 1μmol/dm 3 induced larval settlement and metamorphosis in S. canopus, with short exposure (1 h) to 1 μmol/dm 3 of L-DOPA inducing rapid settlement. In contrast, GABA at the concentrations of 0.1 ~100.0 μmol/dm 3 significantly inhibited the settlement and metamorphosis of S. canopus larvae. In addition, thyroxin at 1~50 μg/dm 3 had no effect on larval settlement and metamorphosis in S. canopus. 7Synthesis and biological evaluation of new 13-n-nonylprotoberberine derivatives as antitubercular agents显示文摘Tuberculosis is a serious threat to public health throughout the world.A series of new l3-n-nonylprotoberberine derivatives was designed,synthesized and evaluated for anti-mycobacterial activity against Mycobacterium tuberculosis strain H_(37)Rv.Several compounds(2,11a-c,11g,13d,15c)exhibited excellent anti-tubercular activity with an MIC below 1.0μg/mL.Notably,compound 13d showed potential antibacterial effect against both drug-susceptible and multidrug-resistant(MDR)M.tuberculosis with MIC ranges of 0.0625-1.0μg/mL.These results suggest a mode of action different from that of the current anti-mycobacterial drugs rifampicin and isoniazid.Hence,compound 13d is an attractive lead compound for the development of new antitubercular agents.Yinghong Li Yanxin Liu Yanxiang Wang Sheng Tang Danqing Song 2013Acta Pharmaceutica Sinica B2013,3,1:3
8Synthesis and biological evaluation of novel tricyclic matrinic derivatives as potential anti-filovirus agents显示文摘Twenty-six novel tricyclic sophoridinic and matrinic derivatives containing a common chlorinated benzene fragment were designed, synthesized and evaluated for their anti-ebolavirus(EBOV)activities. Structure–activity relationship analysis indicated:(i) 12 N-dichlorobenzyl motif was beneficial for the activity;(ii) the chiral configuration at C5 atom might not affect the activity much. Among the target compounds, compound 7d exhibited the most potent potency against EBOV with an IC_(50) value of 5.29 μmol/L and an SI value of over 37.8. Further in vivo anti-EBOV assay of 7d identified its high effectiveness, and in vivo anti-MARV assay of 7d suggested its inspiring broad-spectrum anti-filovirus activity. 9Power allocation scheme for maximizing spectrum efficiency and energy efficiency tradeoff for multi-cluster NOMA system显示文摘In this paper,a power allocation to maximize tradeoff between spectrum efficiency(SE)and energy efficiency(EE)is considered for the downlink non-orthogonal multiple access(NOMA)system with arbitrarily clusters and arbitrarily users,where the subcarriers of clusters are mutually orthogonal to each other.Specifically,an optimization problem of maximizing SE-EE tradeoff is formulated by optimizing power allocation among users under the constraints of user rate requirements.Then,the optimization problem is decomposed into a group of sub-problems with the aim of maximizing SE-EE tradeoff for each cluster,which is solved by using bisection method and monotonicity of function.Finally,the power allocation optimization problem among users is transformed into that between clusters,and a two steps inter-cluster power allocation algorithm is developed to solve this problem.Simulation results show that SE-EE tradeoff of the proposed scheme is better than that of the existing schemes.Li Ya Zhang Danqing Tian Xinji 2020The Journal of China Universities of Posts and Telecommunications2020,27,3:2
10Discovery and identification of EIF2AK2 as a direct key target of berberine for anti-inflammatory effects显示文摘Using chemoproteomic techniques,we first identified EIF2AK2,eEF1A1,PRDX3 and VPS4B as direct targets of berberine(BBR)for its synergistically anti-inflammatory effects.Of them,BBR has the strongest affinity with EIF2AK2 via two ionic bonds,and regulates several key inflammatory pathways through EIF2AK2,indicating the dominant role of EIF2AK2.Also,BBR could subtly inhibit the dimerization of EIF2AK2,rather than its enzyme activity,to selectively modulate its downstream pathways including JNK,NF-κB,AKT and NLRP3,with an advantage of good safety profile.In EIF2AK2 gene knockdown mice,the inhibitory IL-1β,IL-6,IL-18 and TNF-a secretion of BBR was obviously attenuated,confirming an EIF2AK2-dependent anti-inflammatory efficacy.The results highlight the BBR's network mechanism on anti-inflammatory effects in which EIF2AK2 is a key target,and inhibition of EIF2AK2 dimerization has a potential to be a therapeutic strategy against inflammationrelated disorders.Wei Wei Qingxuan Zeng Yan Wang Xixi Guo Tianyun Fan Yinghong Li Hongbin Deng Liping Zhao Xintong Zhang Yonghua Liu Yulong Shi Jingyang Zhu Xican Ma Yanxiang Wang Jiandong Jiang Danqing Song 2023Acta Pharmaceutica Sinica B2023,13,5:2
11Optimization of Extraction Process and Antioxidant Activity of Polysaccharide in Embelia parviflora Wall.by RSM显示文摘[Objectives]To optimize the extraction process of polysaccharide in Embelia parviflora Wall.by response surface methodology,and to study the antioxidant activity in vitro of polysaccharide in E.parviflora,so as to provide a basis for the further development and utilization of E.parviflora.[Methods]The solid-to-liquid ratio,extraction temperature,extraction time and extraction times were used as single factors to investigate the effects.On this basis,the response surface methodology(RSM)was used to optimize the extraction process of polysaccharide in E.parviflora.DPPH and ABTS free radicals were used to investigate the antioxidant capacity of polysaccharide in E.parviflora.[Results]The optimal solid-to-liquid ratio for extraction of polysaccharide from E.parviflora was 32∶1(mL/g),extraction temperature was 77℃,and extraction time was 36 min.The IC50 values of DPPH and ABTS were 0.04 and 0.03 mg/mL,respectively.[Conclusions]The optimized extraction process of polysaccharide in E.parviflora is stable,the extraction rate is high,and has strong antioxidant activity.It is expected to provide a reference for the industrial extraction of polysaccharide from E.parviflora.Wenli LI Xianxian LIU Xiaolian LIANG Yong CHEN Danqing PANG Zhouyan HUANG Qian HAN Wenjun LIU Liangni CHEN 2020Medicinal Plant2020,11,3:2
