Distribution of T-DNA carrying a Ds element on rice chromosomes显示文摘Over 3000 rice plants with T-DNA carrying a Ds element were constructed by Agro-bacterium tumefaciens mediation. Using inverse PCR methodology, 590 unique right flanking sequences of T-DNA (Ds) were retrieved from independent transformants and classified into six main types on the basis of the origin of filler DNA between the right border of T-DNA and flanking sequence of rice genome. Type I sequences were the most common and showed canonical in-tegration that T-DNA right border was followed by rice genome sequence with or without filler DNA of no more than 50 bp, while type II sequences displayed a vector-genome combination that T-DNA right border was followed by a vector fragment and then connected with rice genome sequence. The location and distribution of 340 type I and II flanking sequences on the rice chromosome were determined using BLAST analysis. The 340 Ds insertions at an average in-terval of 0.8 megabase (Mb) constructed a basic framework of Ds starter points on whole rice chromosomes. The frequency of T-DNA (Ds) inserted into the exons of predicted genes on chromosome one was 21%. Knowledge of T-DNA (Ds) locations on chromosomes will prove to be a useful resource for isolating rice genes by Ds transposon tagging as these Ds insertions can be used as starting lines for further mutagenesis. | WANG Jiang LI Lin WAN Xinshan AN Linsheng ZHANG Jingliu | 2004 | Science China(Life Sciences)2004,47,4: | 5
The Effect of Pore Solution on the Hysteretic Curve of Expansive Soil under Cyclic Loading显示文摘A dynamic triaxial instrument was used to study the effects of different concentrations of sodium chloride and stress amplitudes on the dynamic properties of an expansive soil under cyclic loading. In particular, four parameters were considered in such a parametric investigation, namely, hysteresis curve morphology characteristic non-closure degreeεp,the ratio of the short and long axisα,the slope of the long axis k and the enclosed area S.The results show that with an increase in the sodium chloride concentration, the soil particle double electric layer becomes thinner, the distance between soil particles decreases, and the whole sample becomes denser.Theεp-N,α-N and S–N relation curves all show a decreasing trend. The ratio of plastic deformation to total deformation grows with increasing the dynamic stress amplitude, and the curves show an upwards trend. The k-N relationship curve displays an increasing trend with the concentration and a general downwards trend as the dynamic stress amplitude is made higher. This also indicates that sodium chloride solutions can improve the engineering properties of expansive soil to a certain extent. With an increase in the vibration times N,the shape of the hysteretic curve becomes narrower, and the whole soil exhibits a cyclic strain hardening. With the help of an exponential function,a model is introduced to predict the relationship between the concentration and the hysteretic curve. | Xinshan Zhuang Wu Wen Rong Zhou Gaoliang Tao Wentao Li | 2023 | Fluid Dynamics & Materials Processing2023,19,7: | 0
A targeted QTL analysis for fiber length using a genetic population between two introgressed backcrossed inbred lines in upland cotton(Gossypium hirsutum)显示文摘Cotton fiber is the most important natural raw material for the textile industry, and fiber length(FL) is one of the most important traits in cotton.Quantitative trait locus(QTL)mapping based on high-density genetic maps is an efficient approach to identify genetic regions for FL.In our study, two backcrossed inbred lines(BILs) were chosen as parents to construct a high-density genetic map in F2 which was used to fine map FL QTL in F2:3 population. The genetic map had a total size of 3462.8 cM, containing 9182 singlenucleotide polymorphisms(SNPs) based on genotyping-by-sequencing.Two FL related stable QTL were identified on two chromosomes(qFL-A08–1 on A08 and qFL-D03–1 on D03),and qFL-A08–1 was confirmed by a meta-analysis.Utilizing previously obtained RNA-seq data for the two BILs and qRT-PCR analysis, two candidate genes annotated as cytochrome b5(CB5, Gh_A08 G1729) and microtubule end-binding 1 C(EB1C, Gh_D03 G0232) that may regulate FL during the fiber elongation stage were identified. In addition, nine recombination hotspots in this population were found. The results of this study will provide an important foundation for further studies on the molecular and genetic regulation of fiber elongation. | Guoyuan Liua Wenfeng Pei Dan Li Jianjiang Ma Yupeng Cui Nuohan Wang Jikun Song Man Wu Libei Li Xinshan Zang Shuxun Yu Jinfa Zhang Jiwen Yu | 2019 | The Crop Journal2019,7,3: | 0

Substation rotational object detection based on multi-scale feature fusion and refinement显示文摘In modern energy systems, substations are the core of electricity transmission and distribution. However, similar appearance and small size pose significant challenges for automatic identification of electrical devices. To address these issues,we collect and annotate the substation rotated device dataset(SRDD).Further, feature fusion and feature refinement network(F3RNet)are constructed based on the classic structure pattern of backbone-neck-head.Considering the similar appearance of electrical devices, the deconvolution fusion module(DFM)is designed to enhance the expression of feature information. The balanced feature pyramid(BFP)is embedded to aggregate the global feature. The feature refinement is constructed to adjust the original feature maps by considering the feature alignment between the anchors and devices. It can generate more accurate feature vectors. To address the problem of sample imbalance between electrical devices, the gradient harmonized mechanism(GHM)loss is utilized to adjust the weight of each sample. The ablation experiments are conducted on the SRDD dataset.F3RNet achieves the best detection performance compared with classical object detection networks. Also,it is verified that the features from global feature maps can effectively recognize the similar and small devices. | Bin Li Yalin Li Xinshan Zhu Luyao Qu Shuai Wang Yangyang Tian Dan Xu | 2023 | Energy and AI2023,14,4: | 0
A comparative analysis of small RNAs between two Upland cotton backcross inbred lines with different fiber length: Expression and distribution显示文摘The cotton fiber is the most important raw material for the textile industry and an ideal model system for studying cell elongation. However, the genetic variation of fiber elongation in relation to miRNA is poorly understood. A high-throughput comparative RNA-seq of two lines differing in fiber length(FL) from a backcross inbred line(BIL)population of G. hirsutum × G. barbadense revealed differentially expressed(DE) miRNAs and their targets in rapidly elongating fibers. A real-time quantitative PCR analysis was further performed to validate the results. A total of 463(including 47 DE) miRNAs were identified, and seven DE miRNAs were co-localized with seven FL quantitative trait loci(QTL)identified in the G. hirsutum × G. barbadense population. Of 82(including 21 DE) targets identified, nine(including one DE) were also co-localized with the seven FL QTL. The relationship between the allopolyploid and its diploid ancestral species with respect to miRNAs and their targets was also characterized. These results will facilitate the understanding of the molecular genetic mechanism of fiber elongation with regards to miRNAs in cotton. | Guoyuan Liu Man Wu Wenfeng Pei Xihua Li Nuohan Wang Jianjiang Ma Xinshan Zang Shuxun Yu Jinfa Zhang Jiwen Yu | 2019 | The Crop Journal2019,7,2: | 0
Gas expansion caused by formation uplifting and its effects on tight gas accumulation:A case study of Sulige gas field in Ordos Basin,NW China显示文摘Gas expansion caused by significant exhumation in the Sulige gas field in the Ordos Basin since Late Cretaceous and its effects on hydrocarbon accumulation have been investigated systematically based on comprehensive analysis of geochemical, fluid inclusion and production data. The results indicate that gas volume expansion since the Late Cretaceous was the driving force for adjustment and secondary charging of tight sandstone gas reservoirs in the Sulige gas field of the Ordos Basin. The gas retained in the source rocks expanded in volume, resulting in gas re-expulsion,migration and secondary charging into reservoirs, while the gas volume expansion in the tight reservoirs caused the increase of gas saturation,gas-bearing area and gas column height, which worked together to increase the gas content of the reservoir and bring about large-scale gas accumulation events. The Sulige gas field had experienced a two-stage accumulation process, burial before the end of Early Cretaceous and uplifting since the Late Cretaceous. In the burial stage, natural gas was driven by hydrocarbon generation overpressure to migrate and accumulate, while in the uplifting stage, the gas volume expansion drove internal adjustment inside gas reservoirs and secondary charging to form new reservoirs. On the whole, the gas reservoir adjustment and secondary charging during uplifting stage is more significant in the eastern gas field than that in the west, which is favorable for forming gas-rich area. | LI Jun ZHAO Jingzhou WEI Xinshan SHANG Xiaoqing WU Weitao WU Heyuan CHEN Mengna | 2022 | Petroleum Exploration and Development2022,49,6: | 0
