| 1 | Discovery of mitochondrial chimeric-gene associated with cytoplasmic male sterility of HL-rice显示文摘The mitochondrial genome libraries of HL-type sterile line(A) and maintainer line(B) have been constructed. Mitochondrial gene, atp6, was used to screen libraries, due to the different Southern and Northern blot results between sterile and maintainer line. Sequencing analysis of positive clones proved that there were two copies of atp6 gene in sterile line and only one in maintainer line. One copy of atpt6 in sterile line was same to that in maintainer line; the other showed different flanking sequence from the 49th nucleotide downstream of the termination codon of atp6 gene. A new chimeric gene, orfH79, was found in the region. | 6 | Effects of partial portal vein arterialization on the hilar bile duct in a rat model显示文摘BACKGROUND: Liver revascularization is frequently required during the enlarged radical operation for hilar cholangio carcinoma involving the hepatic artery. Researchers have carried out a number of experiments applying partial porta vein arterialization (PVA) in clinical practice. In this study we aimed to establish a theoretical basis for clinical application o partial PVA and to investigate the effects of partial PVA on ra hilar bile duct and hepatic functions. METHODS: Thirty rats were randomly and equally assigned into 3 groups: control (group A), hepatic artery ligation+bile duct recanalization (group B), and partial PVA+bile duc recanalization (group C). Proliferation and apoptosis o rat hilar bile duct epithelial cells, arteriolar counts of the peribiliary plexus (PBP) of the bile duct wall, changes in serum biochemistry, and pathologic changes in the bile duc were assessed 1 month after operation. RESULTS: The proliferation of hilar bile duct epithelial cells in group B was greater than in groups A and C (P<0.01). No apoptotic hilar bile duct epithelial cells were detected in any of the groups. The PBP arteriolar counts of the hilar bile duc wall were similar in groups A and C (P>0.05), but the coun was lower in group B than in group A (P<0.01). No statistically significant differences in alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase and albumin were found in the 3 groups. The gamma-glutamyltransferase value was higher in group B than in groups A and C (P<0.01) The hepatic tissues of groups A and C showed no significan abnormality. Chronic inflammatory changes in the hilar bile duct walls were observed only in group B. | 7 | ^(14)C measurement of forest soils in Dinghushan Biosphere Reserve显示文摘Organic carbon in forest soils of Qingyunsi and Wukesong profiles can be divided into fast and slow components. Δ14C values of these profiles decrease with increasing of depth. The Δ14C values in 30—40 cm depth interval of Wukesong profile are decreasing sharply until a very low value,showing that a strong geological environment change occurred about 1 560 years ago. The 14C apparent ages of Wukesong profile show that the coniferous and broad-leaf mixed forests around Wukesong profile have been developing since 425 a BP, which is consistent with historical documents. | 8 | Expression of co-stimulator 4-1BB molecule in hepatocellular carcinoma and adjacent non-tumor liver tissue,and its possible role in tumor immunity显示文摘AIM:To investigate the expression of 4-1BB molecule in hepatocellular carcinoma (HCC) and its adjacent tissues.METHODS:Reverse transcription-polymerase chain reaction (RT-PCR) was used to determine the gene expression of 4-1BB in hepatocarcinoma and its adjacent tissues,and peripheral blood mononuclear cells (PBMCs) from both HCC and health control groups.Flow cytometry was used to analyse the phenotypes of T cell subsets from the blood of HCC patients and healthy volunteers,and further to determine whether 4-1BB molecules were also expressed on the surface of CD4+ and CD8+ T cells. The localization of 4-1BB proteins on tumor infiltrating T cells was determined by direct immunofluorescence cytochemical staining and detected by confocal microscopy.RESULTS:4-1BB mRNA, which was not detectable in normal liver,was found in 19 liver tissues adjacent to tumor edge(<1.0cm).Low expression of 4-1BB mRNA was shown in 8 tumor tissues and 6 liver tissues located within 1 to 5cm away from tumor edge. In PBMCs, 4-1BB mRNA was almost not detected. Percentage of CD4^+, CD8^+ and CD3^+/CD25^+ T cells, as well as ratio of CD4 to CD8 revealed no difference between groups (P>0.05,respectively),while a significant lower percentage of CD3^+ T cell was found in HCC group as compared to healthy control group (P<0.05).However, 4-1BB molecules were almost not found on the surface of CD4+ and CD8+ T cells in HCC and healthy control group.Double-staining of 4-1BB^+/CD4^+ and 4-1BB^+/CD8^+ immunofluorescence on tumor infiltrating T cells was detected in 13 liver tissues adjacent to tumor edge (<1.0cm) by confocal microscopy.CONCLUSION: Although HCC may escape from immune attack by weak immunogenicity or downregulated expression of MHC-1 molecules on the tumor cell surface,tumor infiltrating T cells can be activated via other costimulatory signal pathways to exert a limited antitumor effect on local microenvironment. | 11 | Impact of Low Frequency Electro-acupuncture on Glucose and Lipid Metabolism in Unmarried PCOS Women:A Randomized Controlled Trial显示文摘Objective:To explore the effect of electro-acupuncture(EA)on glucose and lipid metabolism in unmarried patients with polycystic ovary syndrome(PCOS).Methods:Fifty-four PCOS patients were equally randomized into true acupuncture group and sham acupuncture group(control)for totally 16 weeks of treatment by random method with a computerized randomization program.Patients in true acupuncture group accepted traditional acupuncture methods with EA and two sets of acupoint groups were used alternatively.The first set consisted of Zhongji(CV 3),Qihai(CV 6),Guilai(ST 29),Sanyinjiao(SP 6),Yinlingquan(SP 9),Hegu(LI4)and Baihui(GV 20),and the second set consisted of Tianshu(ST 25),ST 29,CV 3,CV 6,SP 6,Taichong(LR 3),Neiguan(PC)6 and GV 20.Patients in the sham acupuncture group accepted shallow acupuncture methods through EA without electricity at 4 non-meridian points in each shoulder and upper arm.Outcome measures included body mass index(BMI),waist-hip-ratio(WHR),oral glucose tolerance test(OGTT),insulin release test,glucose and lipid metabolism indicators such as total cholesterol(TC),triglycerides,high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol,adiponectin,leptin,visfatin,resistin,and interleukin(IL-6).Results:Twenty-six subjects in the true acupuncture group and 20 subjects in the sham group completed the clinical trial.After 16 weeks of treatment,no significant difference in the outcome measures were observed between the two groups(P>0.05).However,as compared with baseline data,a reduction in weight,BMI,hipline,WHR,fasting glucose,homeostatic model assessment of insulin sensitivity,visfatin and HDL-C,and an increase in resistin and IL-6 were observed in the true acupuncture group(P<0.05).In addition,a reduction in visfatin and an increase in TC were also observed in the sham group(P<0.05).Conclusions:Acupuncture may have a beneficial effect in the treatment of PCOS by improving glucose and lipid metabolism.Moreover,the sham acupuncture may be not completely ineffective.Sham acupuncture may improve some of the aspects of the glucose and lipid metabolism of PCOS patients through a placebo effect.(Registration Nos.ChiCTR-TRC-12002529 and NCT01812161) | DONG Hao-xu WANG Qing WANG Zhi WU Xiao-ke CHENG Ling ZHOU Zhong-ming YANG Li Yl Ping HUANG Dong-mei | 2021 | Chinese Journal of Integrative Medicine2021,27,10: | 4 |

There may be some relationship between RNA editing and cytoplasmic male sterility (CMS), a kind of phenomenon that is attributed to mitochondrial genome mutations. The research materials used are the gametophytic male sterility line (A), maintainer line (B) and F1 hybrid (F1) of HL-type CMS rice. cDNAs and DNAs of atp6 and coxII have been obtained from A, B and F1 by PCR and RT-PCR. Comparing sequences of cDNAs and DNAs, 18 and 15 editing sites were found respectively in the transcripts of atp6 and coxll. A, B and F1 shared the same editing sites.
