5A Highly Pathogenic Strain of Porcine Deltacoronavirus Caused Watery Diarrhea in Newborn Piglets显示文摘Porcine deltacoronavirus(PDCoV) is a newly identified virus that causes watery diarrhea in newborn piglets and results in significant economic losses to the pig industry. Since first reported in Hong Kong in 2012, PDCoV has been subsequently detected in USA, South Korea, Thailand, and China's Mainland. Here we isolated a strain of PDCoV, named CHN-GD-2016,from the intestinal content of a diseased newborn piglet with severe diarrhea in a pig farm in Guangdong, China. PDCoV CHN-GD-2016 could be identified by immunofluorescence with PDCoV specific rabbit antisera, and typical crown-shaped particles with spiky surface projections of this PDCoV were observed with electron microscopy. Genomic analysis showed that the PDCoV CHN-GD-2016 was closely related to other Chinese PDCoV strains, with the highest sequence similarity with the strain CHN/Tianjin/2016. Importantly, inoculation of newborn piglets with 1×10~5 TCID_(50) of CHN-GD-2016 by oral feeding successfully reproduced clear clinical symptoms, including vomiting, dehydration, and severe diarrhea in piglets. In addition, the virus RNA in rectal swabs from 1 to 7 days post inoculation was detected, macroscopic and microscopic lesions in small intestine were observed, and viral antigen was also detected in the small intestines with immunohistochemical staining. 6In situ U-Pb dating of titanite by LA-ICPMS显示文摘Titanite is an ideal mineral for U-Pb isotopic dating because of its relatively high U,Th and Pb contents.Here,we developed a technique for U-Pb dating of titanite using the 193 nm ArF laser-ablation system and Agilent 7500a Q-ICP-MS.Standards of titanite (BLR-1 and OLT-1) and zircon (91500 and GJ-1) were dated using single spot and line raster scan analytical methods.To check the matrix effect,titanite (BLR-1) and zircon (91500) standards were analyzed as the external standards.The weighted mean 206 Pb/238 U ages of OLT-1 titanite are 1015±5 Ma (2,n=24) and 1017±6 Ma (2,n=24) by single spot and line raster scan analyses,respectively,using BLR-1 titanite as the external standard.These ages are consistent with its reference age of about 1014 Ma.However,using 91500 zircon as the external standard,the weighted mean 206 Pb/238 U ages are 917±4 Ma (2,n=24) and 927±5 Ma (2,n=24) for BLR-1 titanite,and 891±4 Ma (2,n=24) and 901±5 Ma (2,n=24) for OLT-1 titanite by single spot and line raster scan analyses,respectively.It is evident that these ages are ~12% younger than their reference values.Our results reveal that significant matrix effect does exist between different kinds of minerals during LA-ICPMS U-Pb age determination,whereas it is insignificant between same minerals.Therefore,same mineral must be used as the external standard for fractionation corrections during in situ LA-ICPMS U-Pb age analysis.In addition,we determined U-Pb ages for titanites from the Early Cretaceous Fangshan pluton,which indicates a rapid cooling history of this pluton.SUN JinFeng YANG JinHui wu FuYuan XIE LieWen YANG YueHeng LIU ZhiChao LI XianHua 2012Chinese Science Bulletin2012,57,20:14
7Effect of hydrophilic silica nanoparticles on hydrate formation: Insight from the experimental study显示文摘Invasion of drilling fluid into natural gas hydrate deposits during drilling might damage the reservoir,induce hydrate dissociation and then cause wellbore instability and distortion of the data from well logging. Adding nanoparticles into drilling fluid is an effective method in reducing the invasion of drilling fluid and enhancing borehole stability. However, the addition of nanoparticles might also introduce hydrate formation risk in borehole because they can act as the 'seeds' for hydrate nucleation. This paper presents an experimental study of the effect of hydrophilic silica nanoparticle on gas hydrate formation in a dynamic methane/liquid-water system. In the experiment, the ultrapure water with and without1.0 wt%–6.0 wt% concentrations of silica nanoparticles, grain sizes of 20 and 50 nm, were pressurized by methane gas under varied conditions of temperature and pressure. The induction time, the gas consumption, and the average rate of gas consumption in the system were measured and compared to those in ultrapure water. The results show that a concentration of 4.0 wt% hydrophilic SiO_2 particles with a grain size of 50 nm has a relatively strong inhibition effect on hydrate formation when the initial experimental condition is 5.0 °C and 5.0 MPa. Compared to ultrapure water, the hydrophilic nano-SiO_2 fluid increases the induction time for hydrate formation by 194% and decreases the amount and average rate of hydrate formation by 10% and 17%, respectively. This inhibition effect may be attributed to the hydrophilicity,amount and aggregation of silica nanoparticle according to the results of water activity and zeta potential measurements. 9Numerical simulation of mould filling process for pressure plate and valve handle in LFC显示文摘In lost foam casting (LFC), the distribution of polymer beads during the bead filling process is not uniform, and the collision between polymer beads determines the distribution of two-phase flow of gas and solid. The interaction between the gas and solid phases reveals as coupling effect of the force that gas exerts on particles or vice versa, or that among particles. The gas-solid flow in filling process is nonlinearity, which makes the coupling effect an essential point to carry out a simulation properly. Therefore, information of each particle's motion is important for acquiring the law of filling process. In bead filling process, compressed air is pressed into mold cavity, and discharged from gas vent, creating a pressure difference between outer and inner space near the gas vent. This pressure difference directly changes the spatial distribution and motion trace of gas and solid phases. In this paper, Discrete Element Method (DEM) and Computational Fluid Dynamics (CFD) are employed to simulate the fluid dynamic character based on Newton's Third Law of Motion. The simulation results of some casting products such as pressure plate and valve handle are compared with the result obtained from practical experiment in order to test the feasibility of DEM. 10Hierarchical activation of compartmentalized pools of AMPK depends on severity of nutrient or energy stress显示文摘AMPK, a master regulator of metabolic homeostasis, is activated by both AMP-dependent and AMP-independent mechanisms. The conditions under which these different mechanisms operate, and their biological implications are unclear. Here, we show that, depending on the degree of elevation of cellular AMP, distinct compartmentalized pools of AMPK are activated, phosphorylating different sets of targets. Low glucose activates AMPK exclusively through the AMP-independent, AXIN-based pathway in lysosomes to phosphorylate targets such as ACC1 and SREBP1c, exerting early anti-anabolic and pro-catabolic roles. Moderate increases in AMP expand this to activate cytosolic AMPK also in an AXIN-dependent manner. In contrast, high concentrations of AMP, arising from severe nutrient stress, activate all pools of AMPK independently of AXIN. Surprisingly, mitochondrion-localized AMPK is activated to phosphorylate ACC2 and mitochondrial fission factor (MFF) only during severe nutrient stress. 12KIF18B promotes tumor progression in osteosarcoma by activating β-catenin显示文摘Objective:Osteosarcoma is a common primary highly malignant bone tumor.Kinesin family member 18B(K1F18B)has been identified as a potential oncogene involved in the development and metastasis of several cancer types.While KIF18B overexpression in osteosarcoma tissue is clearly detected,its specific function in the disease process remains to be established.Methods:K IF18B expression was assessed in osteosarcoma tissues and cells.We additionally evaluated the effects of KIF18B on proliferation,migration,and invasion of osteosarcoma cells,both in vitro and in vivo.Results:Our results showed overexpression of KIF18B in osteosarcoma tissues and cells.Knockdown of K IF18B induced G1/S phase arrest and significantly inhibited proliferation,migration,and invasion of osteosarcoma cells,both in vitro and in vivo.K IF18B regulated P-catenin expression at the transcriptional level by controlling nuclear aggregation of ATF2 and at the post-transcriptional level by interacting with the adenomatous polyposis coli(APC)tumor suppressor gene in osteosarcoma cells.Conclusions:KIF18B plays a carcinogenic role in osteosarcoma by regulating expression ofβ-catenin transcriptionally via decreasing nuclear aggregation of ATF2 or post-transcriptionally through interactions with APC.Our collective findings support the potential utility of KIF18B as a novel prognostic biomarker for osteosarcoma.Tian Gao Ling Yu Zhiwei Fang Jiayong Liu Chujie Bai Shu Li Ruifeng Xue Lu Zhang Zhichao Tan Zhengfu Fan 2020Cancer Biology & Medicine2020,17,2:7
13Effect of preparation method on the bimetallic NiCu/SAPO-11 catalysts for the hydroisomerization of n-octane显示文摘The bimetallic NiCu/SAPO-11 catalysts were prepared by co-impregnation, sequential impregnation, coprecipitation, and mechanical mixing methods. Powder X-ray diffraction, nitrogen adsorption-desorption,temperature-programmed desorption of ammonia, transmission electron microscopy, temperatureprogrammed reduction of hydrogen, and X-ray photoelectron spectroscopy were used to characterize the physicochemical properties of the catalysts. The catalytic performance of the catalysts was assessed by the hydroisomerization of n-octane. Results indicated that the conversion of n-octane and selectivity to n-octane isomers were related to the preparation methods of the catalysts. The catalysts with Ni-Cu alloy effectively restrained the hydrogenolysis reaction that decreases the selectivity of isomerization. The catalyst prepared by the mechanical mixing of NiO and CuO hardly formed Ni-Cu alloy, showing obvious hydrogenolysis and low selectivity to n-octane isomers. The unbalance between the metal and acid sites resulted in the low conversion of n-octane and selectivity to n-octane isomers. 14A general alternate loading technique and its applications in the inverse designs of centrifugal and mixed-flow pump impellers显示文摘For the inverse designs of centrifugal and mixed-flow pump impellers,clarifying the generation process of secondary flows and putting forward corresponding suppression measures is an important approach to improve the impeller performance.In this paper,to provide a better qualitative insight into the generation mechanism of secondary flows in the impeller,a simple kinematic equation is derived based on the ideal assumptions,which indicates that the potential rothalpy gradient(PRG)is the most important dynamic source that actively induces secondary vortical flows.Induced by the natural adverse PRG on the S1 and S2 stream surfaces,two typical secondary flows,H-S and P-S secondary flows,are clearly presented.To specially suppress these typical secondary flows,a general alternate loading technique(GALT)is proposed,aiming to adjust the real blade loadingδp to control the PRG features.At the blade fore part,theδp on the hub streamline should be slowly increased to avoid breakneck growth of the potential rothalpy to reduce adverse streamwise PRG on the S2 streamsurface.At the blade middle part,theδp should be moderately decreased to reduce adverse streamwise PRG on the S1 streamsurface.At the blade aft part,the difference in theδp between the shroud and hub streamlines should be decreased faster to control the exit uniformity.By applying the GALT to the impeller designs of three typical pump types in hydraulic engineering,the organizational effect of the PRG on fundamental flow structures is proven.The GALT can effectively control the PRG distributions and suppress the secondary flows,thereby widening the pump’s high-efficiency zone,improving flow uniformity and suppressing pressure fluctuations.Compared with the current Z-G method and the ALT,the GALT can meet the requirements of'de-experience'better,thereby enabling the designers to obtain good products explicitly and quickly.WANG ChaoYue WANG FuJun AN DongSen YAO ZhiFeng XIAO RuoFu LU Li HE ChengLian ZOU ZhiChao 2021Science China(Technological Sciences)2021,64,4:7
15Estimation method for a skip-stop operation strategy for urban rail transit in China显示文摘The skip-stop operation strategy(SOS) is rarely applied to Chinese urban rail transit networks because it is a simple scheme and a less universally popular transportation service. However, the SOS has performance advantages, in that the total trip time can be reduced depending on the number of skipped stations, crowds of passengers can be rapidly evacuated at congested stations in peak periods, and the cost to transit companies is reduced. There is a contradiction between reducing the trip time under the SOS and increasing the passengers' waiting times under an all-stop scheme. Given this situation, the three objectives of our study were to minimize the waiting and trip times of all passengers and the travel times of trains. A comprehensive estimation model is presented for the SOS. The mechanism through which the trip time for all passengers is affected by the SOS is analyzed in detail. 16Effects of Shuanghuanglian oral liquids on patients with COVID-19:a randomized,open-label,parallel-controlled,multicenter clinical trial显示文摘We conducted a randomized,open-label,parallel-controlled,multicenter trial on the use of Shuanghuanglian(SHL),a traditional Chinese patent medicine,in treating cases of COVID-19.A total of 176 patients received SHL by three doses(56 in low dose,61 in middle dose,and 59 in high dose)in addition to standard care.The control group was composed of 59 patients who received standard therapy alone.Treatment with SHL was not associated with a difference from standard care in the time to disease recovery.Patients with 14-day SHL treatment had significantly higher rate in negative conversion of SARS-CoV-2 in nucleic acid swab tests than the patients from the control group(93.4%vs.73.9%,P=0.006).Analysis of chest computed tomography images showed that treatment with high-dose SHL significantly promoted absorption of inflammatory focus of pneumonia,which was evaluated by density reduction of inflammatory focus from baseline,at day 7(mean difference(95%CI),−46.39(−86.83 to−5.94)HU;P=0.025)and day 14(mean difference(95%CI),−74.21(−133.35 to−15.08)HU;P=0.014).No serious adverse events occurred in the SHL groups.This study illustrated that SHL in combination with standard care was safe and partially effective for the treatment of COVID-19.Li Ni Zheng Wen Xiaowen Hu Wei Tang Haisheng Wang Ling Zhou Lujin Wu Hong Wang Chang Xu Xizhen Xu Zhichao Xiao Zongzhe Li Chene Li Yujian Liu Jialin Duan Chen Chen Dan Li Runhua Zhang Jinliang Li Yongxiang Yi Wei Huang Yanyan Chen Jianping Zhao Jianping Zuo Jianping Weng Hualiang Jiang Dao Wen Wang 2021Frontiers of Medicine2021,15,5:7

19A hidden fault prediction model based on the belief rule base with power set and considering attribute reliability显示文摘Hidden faults are important characteristics of complex systems that cannot be observed directly.The hidden behavior of a system, such as the health state and safety level, has a direct correlation with these hidden faults. After the predicted hidden behavior reaches a fault boundary, certain measures must be taken to avoid fault occurrences. Thus, hidden faults can be predicted by the hidden behavior of a system.The belief rule base(BRB) has been used to predict hidden behaviors. However, two problems remain to be solved in engineering practice. First, when the observed information is absent, ignorance may exist in the output. If only global ignorance is considered, it may be unreasonable in certain cases, which can influence the prediction model. Second, the effects of disturbance factors such as noise and sensor quality may cause the reliability of the gathered information to decline, which indirectly leads to unreliability of the hidden behavior. Thus, to address the global ignorance and unreliable hidden behavior, a new hidden BRB model with a power set and considering attribute reliability(PHBRB-r) is proposed for hidden fault prediction. In the PHBRB-r model, the effects of disturbance factors on hidden behavior are considered using attribute reliability, and the discernment frame is a power set.
