| 1 | Mefatinib as first-line treatment of patients with advanced EGFR-mutant non-small-cell lung cancer:a phase Ib/II efficacy and biomarker study显示文摘EGFR inhibitors have revolutionized the treatment of advanced non-small-cell lung cancer(NSCLC).Mefatinib is a novel,bioavailable,second-generation,irreversible pan-EGFR inhibitor.This phase Ib/II open-label,single-arm,multi-center study investigated the efficacy,safety,biomarker,and resistance mechanisms of mefatinib in the first-line treatment of patients with advanced EGFR-mutant NSCLC.This study included 106 patients with EGFR-mutant stage IIIB-IV NSCLC who received first-line mefatinib at a daily dose of either 60 mg(n=51)or 80 mg(n=55).The primary endpoint was progression-free survival(PFS).Secondary endpoints were overall response rate(ORR),disease control rate(DCR),overall survival(OS),and safety.The cohort achieved an ORR of 84.9%and DCR of 97.2%.The median PFS was 15.4 months and the median OS was 31.6 months.Brain metastasis was detected in 29%of patients(n=31)at diagnosis and demonstrated an ORR of 87.1%,PFS of 12.8 months,and OS of 25.2 months.Adverse events primarily involved skin and gastrointestinal toxicities,which were well-tolerated and manageable.Analyses of mutation profiles were performed using targeted sequencing of plasma samples at baseline,first follow-up 6 weeks from starting mefatinib therapy(F1),and at progression.Patients with concurrent TP53 mutations had comparable PFS as wild-type TP53(14.0 vs 15.4 months;p=0.315).Furthermore,circulating tumor DNA clearance was associated with longer PFS(p=0.040)and OS(p=0.002).EGFR T790M was the predominant molecular mechanism of mefatinib resistance(42.1%,16/38).First-line mefatinib provides durable PFS and an acceptable toxicity profile in patients with advanced EGFR-mutant NSCLC. | Pingli Wang Yuping Li Dongqing Lv Lingge Yang Liren Ding Jianya Zhou Wei Hong Youfei Chen Dongqing Zhang Susu He Jianying Zhou Kai Wang | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 1 |
| 2 | Compounding effects of human activities and climatic changes on coexistence of oasis-desert ecosystems:Prioritizing resilient decision-making for a riskier world显示文摘Water-salt balance is critical for the stable coexistence of salt-affected and groundwater-fed oasis-desert ecosystems. Yet, a comprehensive investigation of how soil salinization and groundwater degradation threaten the coexistence of oasis-desert ecosystems is still scarce, especially under the compounding effects of human activities and climatic changes. Here, we assessed the impacts of irrigated agriculture on hydrological regimes in oasisdesert systems, investigated the spatio-temporal variations of soil salinization in irrigated cropland, and evaluated the implications of the interplays of soil salinization and groundwater degradation on the coexistence of oasis-desert ecosystems in northwestern China, based on meaningful modelling approaches and comprehensive measurements over 1995–2020. The results showed that the irrigation return flow coefficient decreased sharply from 0.21 ± 0.09 in the traditional irrigation period to 0.09 ± 0.01 in the water-saving irrigation period. The continuous drop in groundwater tables and significant degradation of groundwater quality are occurring throughout this watershed. The eco-environmental flows are reaching to their limit with watershed closures(i.e.,the drainage from the oasis region into the desert region is being weakened or even eliminated), although these progressions were largely hidden by regional precipitation and streamflow variability. The process of salt migration and accumulation across different landscapes in oasis-desert system is being reshaped, and soil salinization in water-saving agricultural irrigated lands is accelerating with a regional average annual growth rate of18%. The vegetation in this watershed is degrading, and anthropogenic disturbance accelerates this trend. Our results highlight that environmental stress adaptation strategies must account for resilience maintenance to avoid accelerating catastrophic transitions in oasis-desert ecosystems. | 4 | Integrated bioinformatics analysis reveals correlations of high TRIM59 expression with worse prognosis and immune infiltrates in lung adenocarcinoma显示文摘Objective:Most patients with advanced lung cancer have a poor prognosis.Recent studies have identified TRIM59 as a novel molecule that serves as a prognostic factor for the progression of non-small cell lung cancer.In the present study,we investigated the role of TRIM59 in predicting the prognosis of lung adenocarcinoma(LUAD)as well as the correlation between TRIM59 expression and immune infiltrates.Methods:We analyzed TRIM59 expression in normal and tumor tissues based on RNA-sequencing datasets from The Cancer Genome Atlas and Genotype-Tissue Expression databases.Forty-seven cases of LUAD tissues and their matching adjacent tissues were collected,and TRIM59 expression in tissue samples was demonstrated by immunohistochemistry.All tissue specimens were obtained under the approval of the Medical Ethics Committee of the Second Affiliated Hospital of Zhejiang University School of Medicine(approval No.IR2019001101;approved on April 3,2019).The immune cell scores were calculated using the CIBERSORT database.The Tumor Immune Estimation Resource database was used to analyze the correlation between TRIM59 and immune cell activities.Results:TRIM59 was up-regulated in most cancer types.High TRIM59 expression predicted a worse prognosis in patients with LUAD(overall survival,P=0.00096;disease-specific survival,P=0.00056;disease-free interval,P=0.0009;progression-free interval,P=0.0012).Moreover,TRIM59 was highly expressed in patients with LUAD who had a poorer prognosis.TRIM59 also showed a significant correlation with the ESTIMATE score(P=0.04)and stromal score(P=0.005)in patients with LUAD.Notably,a significant correlation between TRIM59 and the tumor mutation burden was found in LUAD but in no other cancer types(P<0.001).Further investigation showed that TRIM59 had a significant correlation with gene markers on neutrophils and dendritic cells.Conclusion:TRIM59 is a potential prognosticator in LUAD and may be correlated with immune cell identification,immune cell infiltration,and immunotherapy checkpoints in LUAD. | Lingge Yang Yuan Wu Liangkun You Binbin Xie Jun Lou Weidong Han Kai Wang | 2022 | Journal of Bio-X Research2022,5,2: | 0 |
