3Registration of intervention trials of Traditional Chinese Medicine for four neurological diseases on Chinese Clinical Trial Registry and Clinical Trials.gov:a narrative review显示文摘OBJECTIVE:To analyze the current status of clinical trial registration of Traditional Chinese Medicine(TCM)for the treatment of neurological diseases.METHODS:Interventional clinical trials of TCM treatment for ischemic stroke,hemorrhagic stroke,vascular cognitive impairment,tension-type headache before September 22,2020 on the platform of Chinese Clinical Trial Registry(Chi CTR),and Clinical Trials.gov were searched.Two researchers independently selected the literature and extracted data.RESULTS:A total of 180 interventional clinical trials were included for analysis.Out of 180 trials,127 were from Chi CTR and 53 from Clinical Trials.gov.The countries primary sponsoring the included trials were China(176,97.8%),and the common categories of primary sponsors were hospital(131,72.8%).Among the study design,the largest proportion of allocation was randomized(172,95.6%),interventional model assignment was parallel(163,90.6%),masking was double blind 49(27.2%),and the sample size was≤400(144,80.0%).The trials were most carried out at a single center(102,56.7%).Among the included studies,112(62.2%)registered on Chi CTR attached the ethical approval documents.In terms of trial stages,50(27.7%)studies were in phaseⅣ.The mostly used intervention was Chinese herbal medicines(99,55%),acupuncture(68,37.8%)was the second.By searching the registration number on China National Knowledge Infrastructure Database and Pub Med,38(21.1%)registered trials were published,including 25 protocol studies and 14 research results with one(NCT02275949)published both the protocol and the results.CONCLUSIONS:Irregular and inadequate reporting,untimely update and publication,insufficient information on traditional medicine unique characteristics,and lack of international collaborations are the problems existing in the interventional clinical registration trials of traditional medicine treatment on neurological diseases.More efforts need to be made from the above aspects to standardize and improve the registration of traditional medicine trials.WEI Jingjing GUO Rongjuan FU Guojing LIANG Xiao XU Zhenmin JIA Min ZENG Zixiu DU Wanqing JIAO Weiwei SUN Linjuan LIU Hongmei GUO Chunli TONG Chenguang ZHANG Yunling LIAO Xing 2022Journal of Traditional Chinese Medicine2022,42,1:1
4Safety Analysis of Riding at Intersection Entrance Using Video Recognition Technology显示文摘To study riding safety at intersection entrance,video recognition technology is used to build vehicle-bicycle conflict models based on the Bayesian method.It is analyzed the relationship among the width of nonmotorized lanes at the entrance lane of the intersection,the vehicle-bicycle soft isolation form of the entrance lane of intersection,the traffic volume of right-turning motor vehicles and straight-going non-motor vehicles,the speed of right-turning motor vehicles,and straight-going non-motor vehicles,and the conflict between right-turning motor vehicles and straight-going nonmotor vehicles.Due to the traditional statistical methods,to overcome the discreteness of vehicle-bicycle conflict data and the differences of influencing factors,the Bayesian random effect Poisson-log-normal model and random effect negative binomial regression model are established.The results show that the random effect Poisson-log-normal model is better than the negative binomial distribution of random effects;The width of non-motorized lanes,the form of vehicle-bicycle soft isolation,the traffic volume of right-turning motor vehicles,and the coefficients of straight traffic volume obey a normal distribution.Among them,the type of vehicle-bicycle soft isolation facilities and the vehicle-bicycle traffic volumes are significantly positively correlated with the number of vehicle-bicycle conflicts.The width of non-motorized lanes is significantly negatively correlated with the number of vehicle-bicycle conflicts.Peak periods and flat periods,the average speed of right-turning motor vehicles,and the average speed of straight-going non-motor vehicles have no significant influence on the number of vehicle-bicycle conflicts.Xingjian Xue Linjuan Ge Longxin Zeng Weiran Li Rui Song Neal N.Xiong 2022Computers, Materials & Continua2022,,9:1
5Analysis of Wellbore Flow in Shale Gas Horizontal Wells显示文摘Theflow behavior of shale gas horizontal wells is relatively complex,and this should be regarded as the main reason for which conventional pipeflow models are not suitable to describe the related dynamics.In this study,numerical simulations have been conducted to determine the gas-liquid distribution in these wells.In particular,using the measuredflow pressure data related to 97 groups of shale gas wells as a basis,9 distinct pipeflow models have been assessed,and the models displaying a high calculation accuracy for different water-gas ratio(WGR)ranges have been identified.The results show that:(1)The variation law of WGR in gas well satisfies a power function relation.(2)The well structure is the main factor affecting the gas-liquid distribution in the wellbore.(3)The Beggs&Brill,Hagedorn&Brown and Gray models exhibit a high calculation accuracy.Linjuan Zeng Daogang Cai Yunhai Zhao Changqing Ye Chengcheng Luo 2023Fluid Dynamics & Materials Processing2023,19,11:0
6Niclosamide(NA)overcomes cisplatin resistance in human ovarian cancer显示文摘Ovarian cancer(OC)is one of the most lethal malignancies of the female reproduc-tive system.OC patients are usually diagnosed at advanced stages due to the lack of early diag-nosis.The standard treatment for OC includes a combination of debulking surgery and platinum-taxane chemotherapy,while several targeted therapies have recently been approved for maintenance treatment.The vast majority of OC patients relapse with chemoresistant tu-mors after an initial response.Thus,there is an unmet clinical need to develop new therapeu-tic agents to overcome the chemoresistance of OC.The anti-parasite agent niclosamide(NA)has been repurposed as an anti-cancer agent and exerts potent anti-cancer activities in human cancers including OC.Here,we investigated whether NA could be repurposed as a therapeutic agent to overcome cisplatin-resistant(CR)in human OC cells.To this end,we first established two CR lines SKOV3CR and OVCAR8CR that exhibit the essential biological characteristics of cisplatin resistance in human cancer.We showed that NA inhibited cell proliferation,sup-pressed cell migration,and induced cell apoptosis in both CR lines at a low micromole range.Mechanistically,NA inhibited multiple cancer-related pathways including AP1,ELK/SRF,HIF1,and TCF/LEF,in SKOV3CR and OVCAR8CR cells.NA was further shown to effectively inhibit xenograft tumor growth of SKOV3CR cells.Collectively,our findings strongly suggest that NA may be repurposed as an efficacious agent to combat cisplatin resistance in chemoresistant hu-man OC,and further clinical trials are highly warranted.Linjuan Huang Jing Zhang Youling Deng Hao Wang Piao Zhao Guozhi Zhao Wei Zeng Yonghui Wang Connie Chen William Wagstaff Rex C.Haydon Russell R.Reid Tong-Chuan He Le Shen Hue H.Luu Ling Zhao 2023Genes & Diseases2023,10,4:0
7Development of a simplified and inexpensive RNA depletion method for plasmid DNA purification using size selection magnetic beads(SSMBs)显示文摘Plasmid DNA(pDNA)isolation from bacterial cells is one of the most common and critical steps in molecular cloning and biomedical research.Almost all pDNA purification in-volves disruption of bacteria,removal of membrane lipids,proteins and genomic DNA,purifi-cation of pDNA from bulk lysate,and concentration of pDNA for downstream applications.While many liquid-phase and solid-phase pDNA purification methods are used,the final pDNA preparations are usually contaminated with varied degrees of host RNA,which cannot be completely digested by RNase A.To develop a simple,cost-effective,and yet effective method for RNA depletion,we investigated whether commercially available size selection magnetic beads(SSMBs),such as Mag-Bind®TotalPure NGS Kit(or Mag-Bind),can completely deplete bacterial RNA in pDNA preparations.In this proof-of-principle study,we demonstrated that,compared with RNase A digestion and two commercial plasmid affinity purification kits,the SSMB method was highly efficient in depleting contaminating RNA from pDNA minipreps.Gene transfection and bacterial colony formation assays revealed that pDNA purified from SSMB method had superior quality and integrity to pDNA samples cleaned up by RNase A digestion and/or commercial plasmid purification kits.We further demonstrated that the SSMB method completely depleted contaminating RNA in large-scale pDNA samples.Furthermore,the Mag-bind-based SSMB method costs only 5-10%of most commercial plasmid purification kits on a per sample basis.Thus,the reported SSMB method can be a valuable and inexpensive tool for the removal of bacterial RNA for routine pDNA preparations.Xi Wang Ling Zhao Xiaoxing Wu Huaxiu Luo Di Wu Meng Zhan Jing Zhang Mikhail Pakvasa William Wagstaff Fang He Yukun Mao Yongtao Zhang Changchun Niu Meng Wu Xia Zhao Hao Wang Linjuan Huang Deyao Shi Qing Liu Na Ni Kai Fu Kelly Hynes Jason Strelzow Mostafa El Dafrawy Tong-Chuan He Hongbo Qi Zongyue Zeng 2021Genes & Diseases2021,8,3:0
