| 1 | QTG-Seq Accelerates QTL Fine Mapping through QTL Partitioning and Whole-Genome Sequencing of Bulked Segregant Samples显示文摘Deciphering the genetic mechanisms underlying agronomic traits is of great importance for crop improvement. Most of these traits are controlled by multiple quantitative trait loci (QTLs), and identifying the underlying genes by conventional QTL fine-mapping is time-consuming and labor-intensive. Here, we devised a new method, named quantitative trait gene sequencing (QTG-seq), to accelerate QTL fine-mapping. QTGseq combines QTL partitioning to convert a quantitative trait into a near-qualitative trait, sequencing of bulked segregant pools from a large segregating population, and the use of a robust new algorithm for identifying candidate genes. Using QTG-seq, we fine-mapped a plant-height QTL in maize (Zea mays L.), qPH7, to a 300-kb genomic interval and verified that a gene encoding an NF-YC transcription factor was the functional gene. Functional analysis suggested that qPH7-encoding protein might influence plant height by interacting with a CO-like protein and an AP2 domain-containing protein. Selection footprint ana卜 ysis indicated that qPH7 was subject to strong selection during maize improvement. | 3 | New lncRNA annotation reveals extensive functional divergence of the transcriptome in maize显示文摘Long non-coding RNAs(lncRNAs), whose sequences are approximately 200 bp or longer and unlikely to encode proteins, may play an important role in eukaryotic gene regulation. Although the latest maize(Zea mays L.) reference genome provides an essential genomic resource, genome-wide annotations of maize lncRNAs have not been updated. Here, we report on a large transcriptomic dataset collected from 749 RNA sequencing experiments across different tissues and stages of the maize reference inbred B73 line and 60 from its wild relative teosinte. We identified 18,165 high-confidence lncRNAs in maize, of which 6,873 are conserved between maize and teosinte. We uncovered distinct genomic characteristics of conserved lncRNAs,non-conserved lncRNAs, and protein-coding transcripts.Intriguingly, Shannon entropy analysis showed that conserved lncRNAs are likely to be expressed similarly to protein-coding transcripts. Co-expression network analysis revealed significant variation in the degree of co-expression. Furthermore, selection analysis indicated that conserved lncRNAs are more likely than non-conserved lncRNAs to be located in regions subject to recent selection, indicating evolutionary differentiation. | 4 | Role of thioredoxin-interacting protein in mediating endothelial dysfunction in hypertension显示文摘Excessive oxidative stress is a major causative factor of endothelial dysfunction in hypertension.As an endogenous pro-oxidant,thioredoxin-interacting protein(TXNIP)contributes to oxidative damage in various tissues.The present study aimed to investigate the role of TXNIP in mediating endothelial dysfunction in hypertension.In vivo,an experimental model of acquired hypertension was established with two-kidney,one-clip(2K1C)surgery.The expression of TXNIP in the vascular endothelial cells of multiple vessels was significantly increased in hypertensive rats compared with sham-operated rats.Resveratrol,a TXNIP inhibitor,suppressed vascular oxidative damage and increased the expression and activity of eNOS in the aorta of hypertensive rats.Notably,impaired endothelium-dependent vasodilation was effectively improved by TXNIP inhibition in hypertensive rats.In vitro,we observed that Ang II increased the expression of TXNIP in primary human aortic endothelial cells(HAECs)and that TXNIP knockdown by RNA interference alleviated cellular oxidative stress damage and mitigated the impaired eNOS activation and intracellular nitric oxide(NO)production observed in Ang Il-treated HAECs.However,inhibiting thioredoxin(TRX)with PX-12 completely blunted the protective effect of silencing TXNIP.In addition,TXNIP knockdown facilitated TRX expression and promoted TRX nuclear translocation to further activate AP1 and REF1.TRX overexpressi on exhibited favorable effects on eNOS/NO homeostasis in Ang 11-treated HAECs.Thus,TXNIP contributes to oxidative stress and endothelial dysfunction in hypertension,and these effects are dependent on the antioxidant capacity of TRX,suggesting that targeting TXNIP may be a novel strategy for antihypertensive therapy. | Ruiyu Wang Yongzheng Guo Lingjiao Li Minghao Luo Linqian Peng Dingyi Lv Zhe Cheng Qian Xue Liang Wang Jing Huang | 2022 | Genes & Diseases2022,9,3: | 1 |

| 6 | Overestimate of remediation efficiency due to residual sodium persulfate in PAHs contaminated soil and a solution显示文摘Oxidation remediation is a commonly used technology for PAHs contaminated soil presently,but the overestimate of efficiency due to ongoing remediation by residual oxidants during extraction and testing has not been paid enough attention.In this study,persulfate was activated by Fe(Ⅱ)to investigate the effects of residual oxidants on PAHs removal during detection process and the elimination effects of adding Na_(2)SO_(3) and extending sampling time on residual oxidants.Results verified that the residual oxidants removed PAHs in extraction process,making the results lower than the actual values:the detection recovery rate η of ΣPAHs and 3-6 ring PAHs ranged from 24.3%(25%Na_(2)S_(2)O_(8)treatment)to 87.4%(5%Na_(2)S_(2)O_(8)+4/4Fe^(2+) treatment),20.1%-99.0%,28.9%-87.9%,20.8%-89.4%,and 1.8.6%-76.9%,respectively.After adding Na_(2)SO_(3),the accuracy of detection results increased significantly:the η of ΣPAHs and 3-6 ring PAHs increased to 64.1%-96.5%,58.8%-95.5%,73.8%-114.4%,60.6%-95.6%,and 45.4%-77.1%,respectively.After 49 days of adding oxidants,residual oxidants had no considerable effect on the detection of PAHs,indicating it was appropriate to start soil remediation verification sampling49 days after the remediation was completed.The observed results will help scientific evaluation of the remediation effects of chemical oxidation on or-ganic contaminated soil. | Liping He Jianhao Tong Yuanqiang Yang Jianxun Wu Linqian Li Zhonghua Wei Wei Long Jingli Pang Jiyan Shi | 2022 | Journal of Environmental Sciences2022,34,3: | 0 |
