2Ganoderma lucidum spore oil(GLSO),a novel antioxidant, extends the average life span in Drosophila melanogaster显示文摘In ancient China,Ganoderma lucidum was believed to be a medical fungus that could increase lifespan.Recently, pharmacologic studies have found that polysaccharide peptides and triterpenoids extracted from Ganoderma lucidum have various physiological effects as active compounds. However, the effects of spore oil isolated from Ganoderma lucidum remains unknown. In this study, the biological effects of Ganoderma lucidum spore oil(GLSO)were evaluated using a Drosophila melanogaster model.Compared with untreated groups, groups treated with GLSO had significantly longer average and maximum lifespan in both normal conditions and under oxidative stress. The activities of various antioxidant enzymes were measured to determine the antioxidant effect of GLSO.GLSO treatment markedly enhanced total superoxide dismutase(SOD)and catalase(CAT)activity and decreased levels of malondialdehyde(MDA).Further,we found dose-dependent increases in the mRNA expression of Cu,Zn-SOD,Mn-SOD,and CAT in GLSO-treated groups. These results suggest that GLSO may effectively eliminate free radicals and extend lifespan in Drosophila.Future work should investigate the value of GLSO as a functional food for the prevention of aging in larger animal models.Yi Zhang Hongfei Cai Zhu Tao Cheng Yuan Zhaojian Jiang Juyan Liu Hiroshi Kurihara Wendong Xu 2021Food Science and Human Wellness2021,10,1:3

5Total glucosides of Rhizoma Smilacis Glabrae:a therapeutic approach for psoriasis by regulating Th17/Treg balance显示文摘Total glucosides of Rhizoma Smilacis Glabrae(RSG)are selective immunosuppressants that exhibit primary efficacy in the treatment of rheumatoid arthritis through targeted inhibition of activated T cells. In this study,we aimed to investigate the potential application of RSG in the treatment of psoriasis and elucidate its mechanism of action and material basis. Our findings revealed significant improvements upon administration of RSG in an imiquimod(IMQ)-induced psoriasis model. These improvements were characterized by a remarkable increase in the number of tail scales in mice and a substantial amelioration of skin erythema, ulceration, and flaking.By transcriptome sequencing and T-cell flow sorting assay,we identified notable effects of RSG on the modulation of various cellular processes.Specifically,RSG prominently down-regulated the Th17/Treg ratio in damaged skin tissues and reduced the proportion of G2 phase cells.Furthermore,RSG exhibited a stimulatory effect on the proliferation and differentiation of epithelial cells.Of particular interest,we discovered thatβ-sitosterol,sitostenone,stigmasterol,smiglanin,and cinchonain Ib displayed potent inhibitory effects on the IL-17-mediated inflammatory response in HaCaT cells. In summary, our study highlights the therapeutic potential of RSG in the treatment of psoriasis, attributed to its ability to regulate the Th17/Treg balance. These findings contribute to the development of new indications for RSG and provide a solid theoretical foundation for further exploration in this field.TANG Yingzhan YU Jingyi ZHAO Wen LIU Juyan PENG Hongying ZHANG Haoran JIANG Zhenzhou YU Qinwei ZHANG Luyong 2023Chinese Journal of Natural Medicines2023,21,8:0
