1Enzymatic hydrolysis of silkworm pupa and its allergenicity evaluation by animal model with different immunization routes显示文摘Silkworm pupa is a nourishing food with high nutritional value, but its consumption has been greatly limited given its allergenicity.Enzyme hydrolytic technique is recognized as an effective method to reduce the allergenicity of protein. In this study,we aimed to investigate the effect of enzymolysis on the allergenicity of silkworm pupa.Crude silkworm pupa protein was extracted through alkali extraction and acid precipitation, which included 5 proteins with the molecular weights ranging from 34 kDa to 76 kDa,and silkworm pupa were then hydrolyzed by alkaline protease. The allergenicity of silkworm pupa protein and its enzymatic hydrolysates was evaluated by establishing BALB/c mice model, and the mice were immunized via intragastric gavage and intraperitoneal injection, respectively. The results indicated that the intraperitoneal inj ection immunization route induced more by detecting with antibodies, histamine and Th2-related cytokines. Moreover, mice treated with silkworm pupa protein peptide displayed no obvious allergic symptoms, indicating that enzyme hydrolytic technique could significantly reduce the allergenicity of silkworm pupa.Yan Dai Meijia Huang Yujuan Xu Lixia Mu Jingyan Gao Hongbing Chen Zhihua Wu Anshu Yang Yong Wu Xin Li 2023Food Science and Human Wellness2023,12,3:3

6Expressions of matrix metalloproteinase-9 and tissue inhibitor of metalloproteinase-1 in malignant peripheral nerve sheath tumor显示文摘BACKGROUND: Matrix metalloproteinase-9 (MMP-9) can degrade collagen Ⅳ (the main structural ingredient of basilar membrane), and it also plays an important role in tumor vascularization, tumor cell progression, formation of metastatic focus, etc. Tissue inhibitor of metalloproteinase-1 (TIMP-1) can bind with MMP-9 to form 1∶1 compound and inhibit its activity, and can negatively regulate the tumor progression and metastasis. OBJECTIVE: To analyze the relationship of MMP-9 and TIMP-1 expressions with the pathological grade, metastasis and prognosis of malignant peripheral nerve sheath tumor (MPNST). DESIGN: An observational comparative experiment. SETTING: Heze Medical College. PARTICIPANTS: Fifty-eight surgical pathological samples, which were clearly diagnosed to be MPNST, were collected from the pathological laboratory archives in the Department of Pathology, Heze Municipal Hospital from January 1988 to December 2003. The MPNST pathological types were common tumor in 53 cases, malignant triton tumor in 2 cases, epithelial MPNST in 2 cases and MPNST with gland differentiation in 1 case. The pathological grade was grade 1 in 11 cases, grade 2 in 24 cases and grade 3 in 23 cases. Besides, the resected tumor samples of 20 patients with benign peripheral nerve tumor (10 cases of nerve sheath tumor and 10 cases of neurofibromatosis) and the normal peripheral nerves (by-products of some surgeries) of 5 patients were also collected. The samples were used with the approval of the patients. Rat-anti-human MMP-9, TIMP-1 monoclonal antibody and S-P kit were purchased from Fuzhou Maixin Biotechnology, Co.,Ltd. METHODS: The documented paraffin blocks were again prepared to sections of 5 μm. The expressions of MMP-9 and TIMP-1 in the samples were detected with mmunohistochemical S-P method. The relationships of the MPNST severity, recurrence, metastasis and survival rate with the expressions of MMP-9 and TIMP-1 were analyzed. MAIN OUTCOME MEASURES: Relationships of MMP-9 and TIMP-1 expressions with the MPNST severity and prognosis. RESULTS: ① Expressions of MMP-9 and TIMP-1 in three tissues: MMP-9 and TIMP-1 stainings were mainly observed in cytoplasm. Among the 58 MPNST patients, the MMP-9 expression was significantly higher than those in normal peripheral nerve and benign tumor (P < 0.05), while the TIMP-1 expression in MPNST was lower than those in normal peripheral nerve and benign tumor (P < 0.05). ② Relationship of MMP-9 and TIMP-1 expressions with the severity and prognosis of MPNST: The expressions of both proteins were observed in the four subtypes. The positive expression of MMP-9 in the MPNST patients of grades 2-3 was significantly higher than that in the MPNST patients of grade 1 (P < 0.05), while the expression of MMP-9 was significantly lower than that in the MPNST patients of grade 1 (P < 0.05). The metastatic rate was positively correlated with MMP-9 expression (r =1.696, P < 0.05), but negatively correlated with TIMP-1 expression (r =-2.125, P < 0.05). CONCLUSION: MMP-9 and TIMP-1 are associated with MPNST pathological grades and metastasis, and can be used as the indicators for judging the severity and prognosis of MPNST.Yunfei Qi Yingjun Mu Lixia Pei 2007Neural Regeneration Research2007,2,8:0
74-35 Identification of Mutant Gene Induced by Carbon Ion Beams in Arabidopsis by Whole Genome Re-sequencing and Rough Map-based Cloning显示文摘In recent years, heavy ion beams have been recognized as an effective and efficient physical mutagen due to their capacity to induce mutations with high frequency and broad spectrum.Nowadays,it is not so difficult to produce various mutants in plants. However,to mine the responsible mutated genes has been an important challenge. For mutation isolation,map-based cloning is one of the major traditional ways to isolate the mutant genes that control traits of interest in forward genetics studies. However, the process of map-based cloning is usually complicated and time-consuming.Du Yan Li Wenjian Yu Lixia Luo Shanwei Feng Hui Chen Yuze Mu Jinhu Cui Tao Chen Xia Zhou Libin 2016IMP & HIRFL Annual Report2016,,1:0
84-42 Study on the Mechanism of Flower Pigment Mutation Induced by Carbon Ions in Geranium(Pelargonium×hortorum)显示文摘Flower color as the important economic indicator for ornamental plants has become the primary target for ornamental improving and breeding.Heavy ion mutation technology as a unique and efficient mutagen has been widely used in germplasm innovation and plant breeding. In our study,150 young shoots of Pelargonium were exposed to 80 MeV/u carbon ions and finally one stable genetic flower color mutant was obtained at dosage of 30 Gy.Comparing with wild type, the physiological indexes of mutant displayed significant differences. The color of petal in mutant was changed from red to light pink, and the color of peduncle and torus was changed from green to red,as well as the pistils and stamens’color were changed from yellow green to purplish red.Yu Lixia Zhou Libin Du Yan Luo Shanwei Feng Hui Mu Jinhu Chen Yuze Li Wenjian 2016IMP & HIRFL Annual Report2016,,1:0
94-44 Phenotype Screening of Arabidopsis thaliana Irradiated by Carbon Ion Beams Based on High-throughput Imaging Technique显示文摘In recent years, with the development of robotics, sensors and kinds of imaging apparatuses, researchers have developed varieties of automation, high precision and throughput phenotype analysis platforms by ceaselessly hard working and exploring[1,2].Although these platforms have improved the development of phenomics to some degree, there are still some problems in need of resolution. For example, researchers have not yet carried out an effective evaluation of the relationship between plant phenotype and genotype.Certainly,it is the key to solve these problems that how to analyze thoroughly and availably the“big data”obtained from the“high throughput”of the phenotypic analysis platform[3].Based on these issues,we attempted to analyze the huge amounts of data produced by Scanalyzer HTS,and summed up a set of research methods to analyze phenomics data.Mu Jinhu Chen Yuze Du Yan Yu Lixia Yang Jiangyan Luo Shanwei Feng Hui Cui Tao Chen Xia Li Wenjian Zhou Libin 2016IMP & HIRFL Annual Report2016,,1:0

123-36 Optimization of ISSR-PCR System for the Mutants of Arabidopsis thaliana Induced by Carbon Ion Irradiations显示文摘Arabidopsis thaliana is one of the most commonly used model plants due to its short life cycle and concise genome. As a novel physical mutagen, heavy ion beams have been widely used in plant breeding. The most significant advantages of heavy ion beams are the higher linear energy transfer (LET) and relative biological effectiveness (RBE), inducing a broad spectrum of mutation phenotypes even at low irradiation[1]. In order to investigate the effects on DNA behavior of Arabidopsis thaliana mutants induced by carbon ion irradiations, inter-simple sequence repeat (ISSR), a DNA-based molecular marker technique, was performed to assess the genetic polymorphism.Feng Hui Zhou Libin Du Yan Luo Shanwei Chen Yuze Mu Jinhu Yu Lixia Li Wenjian 2015IMP & HIRFL Annual Report2015,,1:0
