1Dietary supplementation of Lonicera macranthoides leaf powder improves amino acid profiles in serum and longissimus thoracis muscle of growing-finishing pigs显示文摘A 60-days feeding trial was conducted to determine the effect of dietary supplementation of Lonicera macranthoides leaf(LML) on growth performance, meat quality, and free amino acid profiles in growingfinishing pigs. L. macranthoides leaves were harvested, air-dried and milled to powder. A total of 288 crossbred barrows(Large White x Landrace) with an average initial BW of 54.0 ± 3.4 kg were randomly allocated to 3 treatment groups and fed either a basal diet or a diet supplemented with 0.5% or 1.0% LML powder. Results showed that diet supplemented with LML powder did not affect average daily feed intake(ADFI) and feed: gain ratio(P> 0.05) in growing-finishing pigs. Moreover, diet supplemented with LML powder showed no significant effects on carcass traits(P > 0.05) including dressing percentage,backfat thickness and loin muscle area, and meat quality traits(P > 0.05) including meat color, intramuscular fat and cooking loss. However, diet supplemented with LML powder significantly improved free amino acid profiles in serum and longissimus thoracis muscle in growing-finishing pigs. Most importantly, dietary LML powder increased total free amino acids content(P < 0.001) and essential free amino acid content(P < 0.001) in longissimus thoracis muscle. 2Genetic Analysis of Two Weak Dormancy Mutants Derived from Strong Seed Dormancy Wild Type Rice N22 (Oryza sativa)显示文摘Two weak dormancy mutants,designated Q4359 and Q4646,were obtained from the rice cultivar N22 after treatment with 400 Gy 60Co gamma-radiation.Compared to the N22 cultivar,the dormancy of the mutant seeds was more readily broken when exposed to a period of room temperature storage.The mutants also showed a reduced level of sensitivity to abscisic acid compared to the N22 cultivar,although Q4359 was more insensitive than Q4646.A genetic analysis indicated that in both mutants,the reduced dormancy trait was caused by a single recessive allele of a nuclear gene,but that the mutated locus was different in each case.The results of quantitative trait locus(QTL)mapping,based on the F2 population from Q4359 x Nanjing35,suggested that Q4359 lacks the QTL qSdn-1 and carries a novel allele at QTL qSdn-9, while a similar analysis of the Q4646 x Nanjing35 F2 population suggested that Q4646 lacks QTL qSdn-5, both qSdn-1 and qSdn-5 are major effect seed dormancy QTL in N22.Therefore,these two mutants were helpful to understand the mechanism of seed dormancy in N22.Bingyue Lu KunXie Chunyan Yang Long Zhang Tao Wu Xi Liu Ling Jiang Jianmin Wan 2011Journal of Integrative Plant Biology2011,53,5:7

5SYNTHESIS OF FUNCTIONAL MACROMOLECULE INTERMEDIATE THROUGH ACYLATION CATALYZED BY [Emim]Cl-AlCl_3 IONIC LIQUID显示文摘Acylation reaction of anthracene with oxalyl chloride in the presence of [Emim]C1-A1C13 ionic liquid has been investigated. Pure 1,2-aceanthryenedione, which is used as intermediate of functional aromatic polymer material, was obtained by recrystalling the reaction mixture with aether and was determined by GC/MS, 1HNMR and FTIR analysis. The influences of various parameters, such as the contents of AlCl3 in [Emim]C1-A1C13, the amount of acylation agent, amount of [Emim]C1-A1C13, reaction temperature and reaction time were investigated. The optimum conditions were as follows: the molar fraction of A1C13 in ionic liquid [x(AlCl3)] being 0.67 , molar ratio of ionic liquid to anthracene being 2:1 , molar ratio of oxalyl chloride to anthracene being 2:1, reaction temperature being 40℃ and reaction time being 6h. Under above conditions, the yield and selectivity of 1,2-aceanthrylenedione can reach 91.5% and 98.3% respectively. 6SYNTHESIS OF 1,2-ACEANTHRYLENEDIONE THROUGH ACYLATION OF ANTHRACENE WITH OXALYL CHLORIDE显示文摘Acylation of anthracene and oxalyl chloride in the presence of anhydrous AlCl3 was investigated. Pure 1,2-aceanthrylenedione, which is used as new functional macromolecule, was prepared by extraction and recrystallation. The structure of 1,2-aceanthrylenedione was identified by measure of melting point, GC/MS, FTIR and 1HNMR analyses. The influences of various parameters, such as solvent, dosage of catalyst, molar ratio of anthracene to oxalyl chloride, reaction temperature, and reaction time were studied. The anhydrous AlCl3 was found to catalyze the reaction with high yeild and good selectivity. 7Junctional and somatic hypermutation-induced CX4C motif is critical for the recognition of a highly conserved epitope on HCV E2 by a human broadly neutralizing antibody显示文摘Induction of broadly neutralizing monoclonal antibodies(bNAbs)that bind to the viral envelope glycoproteins is a major goal of hepatitis C virus(HCV)vaccine research.The study of bNAbs arising in natural infection is essential in this endeavor.We generated a human antibody,8D6,recognizing the E2 protein of HCV isolated from a chronic hepatitis C patient.This antibody shows broadly neutralizing activity,which covers a pan-genotypic panel of cell culture-derived HCV virions(HCVcc).Functional and epitope analyses demonstrated that 8D6 can block the interaction between E2 and CD81 by targeting a highly conserved epitope on E2.We describe how the 8D6 lineage evolved via somatic hypermutation to achieve broad neutralization.We found that the V(D)J recombination-generated junctional and somatic hypermutation-induced disulfide bridge(C-C)motif in the CDRH3 is critical for the broad neutralization and binding activity of 8D6.This motif is conserved among a series of broadly neutralizing HCV antibodies,indicating a common binding model.Next,the 8D6 inferred germline(iGL)was reconstructed and tested for its binding affinity and neutralization activity.Interestingly,8D6 iGL-mediated relatively strong inhibition of the 1b genotype PR79L9 strain,suggesting that PR79L9 may serve as a potential natural viral strain that provides E2 sequences that induce bNAbs.Overall,our detailed epitope mapping and genetic studies of the HCV E2-specific mAb 8D6 have allowed for further refinement of antigenic sites on E2 and reveal a new mechanism to generate a functional CDRH3,while its iGL can serve as a probe to identify potential HCV vaccine strains.Chunyan Yi Jing Xia Lan He Zhiyang Ling Xuesong Wang Yu Yan Jiangjun Wang Xinhao Zhao Weiguo Fan Xiaoyu Sun Ronghua Zhang Sheng Ye Rongguang Zhang Yongfen Xu Liyan Ma Yaguang Zhang Honglin Zhou Zhong Huang Junqi Niu Gang Long Junxia Lu Jin Zhong Bing Sun 2021Cellular & Molecular Immunology2021,18,3:0
8CGG repeat expansion in LOC642361/NUTM2B-AS1 typically presents as oculopharyngodistal myopathy显示文摘CGG repeat expansions in LOC642361/NUTM2B-AS1 have recently been identified as a cause of oculopharyngeal myopathy with leukoencephalopathy.However,since only three patients from a single family were reported,it remains unknown whether their clinicopathological features are typical for CGG repeat expansions in LOC642361/NUTM2B-AS1.Here,using repeat-primed-polymerase chain reaction and long-read sequencing,we identify 12 individuals from 3 unrelated families with CGG repeat expansions in LOC642361/NUTM2B-AS1,typically presenting with oculopharyngodistal myopathy.The CGG repeat expansions range from 161 to 669 repeat units.Most of the patients present with ptosis,restricted eye movements,dysphagia,dysarthria,and diffuse limb muscle weakness.Only one patient shows T2-weighted hyperintensity in the cerebellar white matter surrounding the deep cerebellar nuclei on brain magnetic resonance imaging.Muscle biopsies from three patients show a myopathic pattern and rimmed vacuoles.Analyses of muscle biopsies suggest that CGG repeat expansions in LOC642361/NUTM2B-AS1 may deleteriously affect aggrephagic capacity,suggesting that RNA toxicity and mitochondrial dysfunction may contribute to pathogenesis.Our study thus expands the phenotypic spectrum for the CGG repeat expansion of LOC642361/NUTM2B-AS1 and indicates that this genetic variant typically manifests as oculopharyngodistal myopathy with chronic myopathic changes with rimmed vacuoles and filamentous intranuclear inclusions in muscle fibers.Yan Shi Chunyan Cao Yiheng Zeng Yuanliang Ding Long Chen Fuze Zheng Xuejiao Chen Fanggui Zhou Xiefeng Yang Jinjing Li Liuqing Xu Guorong Xu Minting Lin Hiroyuki Ishiura Shoji Tsuji Ning Wang Zhiqiang Wang Wan-Jin Chen Kang Yang 2024Journal of Genetics and Genomics2024,51,2:0
9Primary metabolite contents are correlated with seed protein and oil traits in near-isogenic lines of soybean显示文摘Soybean(Glycine max[L.]Merr.)is an important source of human dietary protein and vegetable oil.A strong negative correlation between protein and oil contents has hindered efforts to improve soybean seed quality.The metabolic and genetic bases of soybean seed composition remain elusive.We evaluated metabolic diversity in a soybean near-isogenic line(NIL)population derived from parents(JD12 and CMSD)with contrasting seed oil contents.Using GC-TOF/MS,we compared seed primary metabolites of high protein/low oil lines,low protein/high oil lines,and their parents.Principal-components analysis showed that metabolic profiles of all progeny lines could be discriminated based on protein and oil contents.Univariate analysis revealed wide variation and transgressive segregation of metabolites in the population.Twenty-eight annotated metabolites,in particular free asparagine,free 3-cyanoalanine,and L-malic acid,were correlated with seed protein content or seed oil content or seed protein and oil content.These results shed light on the metabolic and genetic basis of soybean seed composition.Jie Wang Pengfei Zhou Xiaolei Shi Na Yang Long Yan Qingsong Zhao Chunyan Yang Yuefeng Guan 2019The Crop Journal2019,7,5:0
