| 1 | Super-resolution dipole orientation mapping via polarization demodulation显示文摘Fluorescence polarization microscopy(FPM)aims to detect the dipole orientation of fluorophores and to resolve structural information for labeled organelles via wide-field or confocal microscopy.Conventional FPM often suffers from the presence of a large number of molecules within the diffraction-limited volume,with averaged fluorescence polarization collected from a group of dipoles with different orientations.Here,we apply sparse deconvolution and least-squares estimation to fluorescence polarization modulation data and demonstrate a super-resolution dipole orientation mapping(SDOM)method that resolves the effective dipole orientation from a much smaller number of fluorescent molecules within a sub-diffraction focal area.We further apply this method to resolve structural details in both fixed and live cells.For the first time,we show that different borders of a dendritic spine neck exhibit a heterogeneous distribution of dipole orientation.Furthermore,we illustrate that the dipole is always perpendicular to the direction of actin filaments in mammalian kidney cells and radially distributed in the hourglass structure of the septin protein under specific labelling.The accuracy of the dipole orientation can be further mapped using the orientation uniform factor,which shows the superiority of SDOM compared with its wide-field counterpart as the number of molecules is decreased within the smaller focal area.Using the inherent feature of the orientation dipole,the SDOM technique,with its fast imaging speed(at sub-second scale),can be applied to a broad range of fluorescently labeled biological systems to simultaneously resolve the valuable dipole orientation information with super-resolution imaging. | Karl Zhanghao Long Chen Xu-San Yang Miao-Yan Wang Zhen-Li Jing Hong-Bin Han Michael Q Zhang Dayong Jin Jun-Tao Gao Peng Xi | 2016 | Light(Science & Applications)2016,5,1: | 11 |

| 6 | Four-sample lactose hydrogen breath test for diagnosis of lactose malabsorption in irritable bowel syndrome patients with diarrhea显示文摘AIM: To validate 4-sample lactose hydrogen breath testing(4SLHBT) compared to standard 13-sample LHBT in the clinical setting.METHODS: Irritable bowel syndrome patients with diarrhea(IBS-D) and healthy volunteers(HVs) were enrolled and received a 10 g, 20 g, or 40 g doselactose hydrogen breath test(LHBT) in a randomized, double-blinded, controlled trial. The lactase gene promoter region was sequenced. Breath samples and symptoms were acquired at baseline and every 15 min for 3 h(13 measurements). The detection rates of lactose malabsorption(LM) and lactose intolerance(LI) for a 4SLHBT that acquired four measurements at 0, 90, 120, and 180 min from the same data set were compared with the results of standard LHBT.RESULTS: Sixty IBS-D patients and 60 HVs were studied. The genotype in all participants was C/C-13910. LM and LI detection rates increased with lactose dose from 10 g, 20 g to 40 g in both groups(P < 0.001). 4SLHBT showed excellent diagnostic concordance with standard LHBT(97%-100%, Kappa 0.815-0.942) with high sensitivity(90%-100%) and specificity(100%) at all three lactose doses in both groups.CONCLUSION: Reducing the number of measurements from 13 to 4 samples did not significantly impact on the accuracy of LHBT in health and IBS-D. 4SLHBT is a valid test for assessment of LM and LI in clinical practice. | Jian-Feng Yang Mark Fox Hua Chu Xia Zheng Yan-Qin Long Daniel Pohl Michael Fried Ning Dai | 2015 | World Journal of Gastroenterology2015,21,24: | 4 |

| 10 | Mesencephalic astrocyte-derived neurotrophic factor ameliorates steatosis in HepG2 cells by regulating hepatic lipid metabolism显示文摘BACKGROUND Nonalcoholic fatty liver disease (NAFLD) is a global metabolism-associated liver disease.Mesencephalic astrocyte-derived neurotrophic factor (MANF) is a newly discovered secreted protein that is involved in metabolic homeostasis.However,much remains to be discovered about its function in hepatic lipid metabolism;thus,we assessed whether MANF could regulate hepatic metabolism.AIM To establish in vivo and in vitro NAFLD models to explore the role of MANF in hepatic lipid metabolism.METHODS HepG2 cells treated with free fatty acids (FFAs) and ob/ob mice were used as NAFLD models.Liver tissues collected from wild type and ob/ob mice were used to detect MANF expression.Cells were treated with FFAs for different durations.Moreover,we used lentiviral constructs to establish overexpression and knockdown cell models in order to interfere with MANF expression levels and observe whether MANF influences hepatic steatosis.Western blot analysis and quantitative real-time PCR were used to detect protein and gene expression,and oil red O staining was used to visualize intracellular lipid droplets.RESULTS Hepatic MANF protein and mRNA expression in wild type mice were 10-fold and 2-fold higher,respectively,than those in ob/ob mice.The MANF protein was temporarily increased by 1.3-fold after stimulation with FFAs for 24 h and gradually decreased to 0.66-fold that of the control at the 72 h time point in HepG2 cells.MANF deficiency upregulated the expression of genes involved infatty acid synthesis,cholesterol synthesis,and fatty acid uptake and aggravated HepG2 cell steatosis,while MANF overexpression inhibited fatty acid synthesis and uptake and cholesterol synthesis,and rescued HepG2 cells from FFAsinduced steatosis.Furthermore,a significant decrease in triglyceride levels was observed in the MANF overexpression group compared with the control group(0.4288±0.0081 mmol/g vs 0.3746±0.0121 mmol/g,P <0.05) upon FF As treatment.There was also a 17%decrease in intracellular total cholesterol levels between the MANF overexpression group and the control group (0.1301±0.0059mmol/g vs 0.1088±0.0009 mmol/g,P <0.05) upon FF As treatment.Moreover,MANF suppressed lipid deposition in HepG2 cells.CONCLUSION Our findings indicate that MANF improves the phenotype of liver cell steatosis and may be a potential therapeutic target in hepatic steatosis processes. | Miao He Cong Wang Xiao-Hong Long Jia-Jia Peng Dong-Fang Liu Gang-Yi Yang Michael D Jensen Li-Li Zhang | 2020 | World Journal of Gastroenterology2020,26,10: | 3 |

| 16 | Graphene-loaded metal wire grating for deep and broadband THz modulation in total internal reflection geometry显示文摘We employed a metallic wire grating loaded with graphene and operating in total internal reflection (TIR) geometry to realize deep and broadband THz modulation. The non-resonant field enhancement effect of the evanescent wave in TIR geometry and in the subwavelength wire grating was combined to demonstrate a ~77% modulation depth (MD) in the frequency range of 0.2–1.4 THz. This MD, achieved electrically with a SiO_2∕Si gated graphene device, was 4.5 times higher than that of the device without a metal grating in transmission geometry. By optimizing the parameters of the metallic wire grating, the required sheet conductivity of graphene for deep modulation was lowered to 0.87 mS.
