3Development of A Neonatal Mouse Model for Coxsackievirus B1 Antiviral Evaluation显示文摘Coxsackievirus B1(CVB1) is a leading causative agent of severe infectious diseases in humans and has been reported to be associated with outbreaks of aseptic meningitis, myocarditis, and the development of chronic diseases such as type 1 diabetes mellitus(T1DM). There is no approved vaccine or effective antiviral therapy to treat CBV1 infection. And animal models to assess the effects of antiviral agents and vaccine remain limited. In this study, we established a neonatal mouse model of CVB1 using a clinically isolated strain to characterize the pathological manifestations of virus infection and to promote the development of vaccines and antiviral drugs against CVB1. One-day-old BALB/c mice were susceptible to CVB1 infection by intraperitoneal injection. Mice challenged with CVB1 at a low dose [10 median tissue culture infective dose(TCID_(50))] exhibited a series of clinical symptoms, such as inactivity, emaciation, limb weakness, hair thinning,hunching and even death. Pathological examination and tissue viral load analysis showed that positive signals of CVB1 were detected in the heart, spinal cord, limb muscle and kidney without pathological damage. Particularly, CVB1 had a strong tropism towards the pancreas, causing severe cellular necrosis with inflammatory infiltration, and was spread by viraemia. Notably, the monoclonal antibody(mAb) 6H5 and antisera elicited from CVB1-vaccinated mice effectively protected the mice from CVB1 infection in the mouse model. 4The quantitative proteome atlas of a model cyanobacterium显示文摘Cyanobacteria are a group of oxygenic photosynthetic bacteria with great potentials in biotechnological applications and advantages as models for photosynthesis research. The subcellular localizations of the majority of proteins in any cyanobacteria remain undetermined, representing a major challenge in using cyanobacteria for both basic and industrial researches. Here, using label-free quantitative proteomics, we map 2027 proteins of Synechocystis sp. PCC6803, a model cyanobacterium, to different subcellular compartments and generate a proteome atlas with such information. The atlas leads to numerous unexpected but important findings, including the predominant localization of the histidine kinases Hik33 and Hik27 on the thylakoid but not the plasma membrane. Such information completely changes the concept regarding how the two kinases are activated. 5Determination of total flavonoids and its antioxidant activity in the exocarp of Aronia melanocarpa显示文摘As a kind of berry food rich in flavonoids,the determination method of total flavonoids content of Aronia melanocarpa is of great significance.This paper compares the applicability of NaNO_(2)-Al(NO_(3))_(3)-NaOH colorimetry and AlCl_(3)colorimetry to total flavonoids content of the exocarp of Aronia melanocarpa by Ultraviolet-visible Spectrophotometry.At the same time,we determined hydroxyl radical(OH·-)inhibition rate,superoxide anion radical(O_(2)·-)inhibition rate and 1,1-diphenyl-2-picrylhydrazyl radical free radical(DPPH·)scavenging rate of total flavonoids extracted from Aronia melanocarpa.It is found that AlCl_(3)colorimetry has good repeatability,precision,and accuracy.The method is suitable for the determination of total flavonoids in the exocarp of Aronia melanocarpa.Thus,by the established method the content of total flavonoids in the exocarp of Aronia melanocarpa was determined as high as(36.711±0.034)mg/g.The results of antioxidant activity showed superoxide anion radical scavenging ability,hydroxyl radical scavenging ability and DPPH radical scavenging ability of the total flavonoids extract reached 45.33%,87.87%and 98.05%,respectively,indicating that it had good antioxidant activity.Longfa Li Weizhuo Xu Wei Xu Ming Song 2022Journal of Polyphenols2022,4,2:0
