| 1 | Nano-needle strontium-substituted apatite coating enhances osteoporotic osseointegration through promoting osteogenesis and inhibiting osteoclastogenesis显示文摘Implant loosening remains a major clinical challenge for osteoporotic patients.This is because osteoclastic bone resorption rate is higher than osteoblastic bone formation rate in the case of osteoporosis,which results in poor bone repair.Strontium(Sr)has been widely accepted as an anti-osteoporosis element.In this study,we fabricated a series of apatite and Sr-substituted apatite coatings via electrochemical deposition under different acidic conditions.The results showed that Ca and Sr exhibited different mineralization behaviors.The main mineralization products for Ca were CaHPO4⋅2H2O and Ca3(PO4)2 with the structure changed from porous to spherical as the pH values increased.The main mineralization products for Sr were SrHPO4 and Sr5(PO4)3OH with the structure changed from flake to needle as the pH values increased.The in vitro experiment demonstrated that coatings fabricated at high pH condition with the presence of Sr were favorable to MSCs adhesion,spreading,proliferation,and osteogenic differentiation.In addition,Sr-substituted apatite coatings could evidently inhibit osteoclast differentiation and fusion.Moreover,the in vivo study indicated that nano-needle like Sr-substituted apatite coating could suppress osteoclastic activity,improve new bone formation,and enhance bone-implant integration.This study provided a new theoretical guidance for implant coating design and the fabricated Sr-substituted coating might have potential applications for osteoporotic patients. | Zhen Geng Luli Ji Zhaoyang Li Jing Wang Hongyan He Zhenduo Cui Xianjin Yang Changsheng Liu | 2021 | Bioactive Materials2021,6,4: | 4 |

| 4 | Effect of hypotonic medium on aquaporin-4 expression in cultured astrocytes显示文摘BACKGROUND: Aquaporin-4 (AQP4) is abundant in astrocytes, ependymal cells, and the choroid plexus, and is associated with cerebrospinal fluid formation and osmoregulation. AQP4 is speculated to be the hypothalamic osmoreceptor and regulator of water balance. OBJECTIVE: To examine AQP4 expression and its role in cultured rat astrocytes after exposure to hypotonic medium. DESIGN, TIME AND SETTING: Randomized control experiment. This experiment was carried out in the Research Room of Neurobiology, Chongqing University of Medical Science, China, between April and October 2003. MATERIALS: Two-day-old newborn Wistar rats (n =20), weighing 10-15 g, were purchased from the Experimental Animal Center of Chongqing University of Medical Science, China. METHODS: Purified rat cerebral cortical astrocytes were isolated from Wistar rats for in vitro cell culture experiments. The cells were randomly divided into control and hypotonic groups. The in vitro cell edema model was established by exposing astrocytes to hypotonic medium (268, 254, or 240 mmol/L). Cells in the control group were cultured in normal culture medium. MAIN OUTCOME MEASURES: Morphological changes in astrocytes were observed under an inverted microscope and a transmission electron microscope after cells were cultured for 3, 6, 12, or 24 hours with hypotonic medium or normal culture medium. In each group, AQP4 protein and mRNA expression were assessed by immunocytochemistry, in situ hybridization, and reverse transcription polymerase chain reaction at the different time points. RESULTS: After astrocytes were cultured for 3, 6, 12, or 24 hours with hypotonic medium (268, 254, 240 mmol/L), they showed typical features of cell edema. In the control group, no astrocytes developed pathological changes. There were no significant changes in the AQP4 mRNA and protein expression in the control group at any of the time points after astrocytes were cultured with normal culture medium (P > 0.05). Compared with the control group, AQP4 mRNA and protein expression in the hypotonic group were remarkably increased at all time points after astrocytes cultured with hypotonic medium (268, 254, 240 mmol/L; P < 0.05). AQP4 mRNA and protein expression increased with increasing exposure time and with decreasing concentration of the hypotonic medium. CONCLUSION: Hypotonic medium induced cell edema and increased AQP4 mRNA and protein expression. | 5 | A novel snail-inspired bionic design of titanium with strontium-substituted hydroxyapatite coating for promoting osseointegration显示文摘As the population ages,more and more people are suffering from osteoarthritis(OA),resulting in an increasing requirement for joint implants.Surface modification to improve the topology and composition of the implants has been proved to be an effective way to improve the primary stability and long-term success rate of joint implants.In this work,a bionic micro/nano-structure accompanied with a strontium-substituted hydroxyapatite(SrHA)coating was fabricated on titanium(Ti)surface via electrochemical corrosion,ultrasonic treatment,and hydrothermal deposition methods.Thein vitro study demonstrated that the bionic structure and the bioactive apatite could synergistically increase the expressions of integrin-related gene(ITGα5β1)and osteoblastic genes(Col-I and OCN),and thus promote osteoblast growth.In addition,owing to the anti-bone resorption property of Sr^(2+),the coating could effectively inhibit osteoclast differentiation and proliferation.In a word,the prepared samples not only promoted osteogenesis but also inhibited osteoclastogenesis.The in vivo experiment via a rabbit model found that the bionic structured surface provided the pore for new bone ingrowth,which was beneficial to the mechanical interlocking between the implant and bone.Moreover,the bionic structure and bioactive SrHA coating had a synergistic effect on promoting bone formation,osseointegration,and bone-implant bonding strength.This study therefore presented a new strategy to fabricate bio-functionalized Ti-based implants for potential application in orthopedics field. | Zhen Geng Xueping Li Luli Ji Zhaoyang Li Zhenduo Cui Jing Wang Jingyuan Cui Xianjin Yang Changsheng Liu | 2021 | Journal of Materials Science & Technology2021,,20: | 0 |
| 6 | Oblique Cutting Based Mechanical Model for Insertion Torque of Dental Implant显示文摘The insertion torque of a dental implant is an important indicator for the primary stability of dental implants.Thus,the preoperative prediction for the insertion torque is crucial to improve the success rate of implantation surgery.In this present research,an alternative method for prediction of implant torque was proposed.First,the mechanical model for the insertion torque was established based on an oblique cutting process.In the proposed mechanical model,three factors,including bone quality,implant geometry and surgical methods were considered in terms of bone-quality coefficients,chip load and insertion speeds,respectively.Then,the defined bone-quality coefficients for cancellous bone with the computed tomography(CT)value of 235–245,345–355 and 415–425 Hu were obtained by a series of insertion experiments of IS and ITI implants.Finally,the insertion experiments of DIO implants were carried out to verify the accuracy of developed model.The predicted insertion torques calculated by the mechanical model were compared with those acquired by insertion experiments,with good agreement,the relative error being less than 15%.This method allows the insertion torque for different implant types to be quickly established and enhances prediction accuracy by considering the effects of implants’geometries and surgical methods. | Luli Li Song Zhang Quhao Li Cuirong Bian Airong Zhang | 2022 | Chinese Journal of Mechanical Engineering2022,35,3: | 0 |
| 7 | Genetic Diversity and Population Structure Analysis of Barley Landraces from Shanghai Region Using Genotyping-by-Sequencing显示文摘Barley(Hordeum vulgare L.)is an important economic crop for food,feed and industrial raw materials.In the present research,112 barley landraces from the Shanghai region were genotyped using genotyping-by-sequencing(GBS),and the genetic diversity and population structure were analyzed.The results showed that 210,268 Single Nucleotide Polymorphisms(SNPs)were present in total,and the average poly-morphism information content(PIC)was 0.1642.Genetic diversity and population structure analyses suggested that these barley landraces were differentiated and could be divided into three sub-groups,with morphological traits of row-type and adherence of the hulls the main distinguishing factors between groups.Genotypes with similar or duplicated names were also investigated according to their genetic backgrounds and seed appearances.This study provided valuable information on barley landraces from the Shanghai region,and showed that all these barley landraces should be protected and used for future breeding programs. | Luli Li Nigel G.Halford Huihui Wang Yingjie Zong Zhenzhu Guo Ruiju Lu Chenghong Liu Zhiwei Chen | 2023 | Phyton-International Journal of Experimental Botany2023,92,4: | 0 |
