1Microvesicle-delivery miR-150 promotes tumorigenesis by up-regulating VEGF,and the neutralization of miR-150 attenuate tumor development显示文摘Tumor-associated macrophages(TAMs)mostly exhibit M2-like(alternatively activated)properties and play posi-tive roles in angiogenesis and tumorigenesis.Vascular en-dothelial growth factor(VEGF)is a key angiogenic factor.During tumor development,TAMs secrete VEGF and other factors to promote angiogenesis;thus,anti-treatment against TAMs and VEGF can repress cancer development,which has been demonstrated in clinical trials and on an experimental level.In the present work,we show that miR-150 is an oncomir because of its promotional effect on VEGF.MiR-150 targets TAMs to up-regulate their secre-tion of VEGF in vitro.With the utilization of cell-derived vesicles,named microvesicles(MVs),we transferred an-tisense RNA targeted to miR-150 into mice and found that the neutralization of miR-150 down-regulates miR-150 and VEGF levels in vivo and attenuates angiogenesis.There-fore,we proposed the therapeutic potential of neutralizing miR-150 to treat cancer and demonstrated a novel,natu-ral,microvesicle-based method for the transfer of nucleic acids.Yuchen Liu Luming Zhao Dameng Li Yuan Yin Chen-Yu Zhang Jing Li Yujing Zhang 2013Protein & Cell2013,4,12:18
2Aqueous extract of Taxus Chinensis (Pilger) Rehd inhibits lung carcinoma A549 cells through the epidermal growth factor receptor/mitogen-activated protein kinase pathway in vitro and in vivo显示文摘OBJECTIVE:To explore the anticancer mechanismof aqueous extract of Taxus Chinensis (Pilger) Rehd (AETC).METHODS: The serum pharmacological method was used to avoid interference from administration of the crude medicinal herbs. Eight purebred New Zealand rabbits were used for preparation of serum containing various concentrations of AETC. Forty-eightBalb/c-nu mice were used for in vivo experiments. The effects of serum containing AETC on the proliferation of A549 cells and expression levels of the epidermal growth factor receptor/mitogen-activated protein kinase(EGFR/MAPK) pathway-related proteins in vitro were investigated. Additionally, the effects on the growth of A549 xenografts in nudemice,and expression levels of the EGFR/MAPK pathway-related proteins in the xenografts,were investigated.RESULTS: 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) assay revealed that the serum containing AETC significantly decreased the viability of A549 cells in a dose-dependent manner.Western blot showed that the serum containing various concentrations of AETC strongly reduced the levels of phospho-Jun N-terminal kinase(p-JNK) and phospho-extracellular signal-regulated kinase1/2(ERK1/2) while it increased the level of p-p38. However, no significant effects on the expression levels of JNK, ERK1/2, and p38 MAPK were found. In addition, an anticancer effect from AETC was observed invivo in the Balb/c-nu mice bearing A549xenografts.CONCLUSION: AETC has significant effects on the growth of A549 xenografts and on the activity of the EGFR/MAPK pathway. 7Astronaut mass measurement using linear acceleration method and the effect of body non-rigidity显示文摘Astronaut's body mass is an essential factor of health monitoring in space.The latest mass measurement device for the International Space Station (ISS) has employed a linear acceleration method.The principle of this method is that the device generates a constant pulling force,and the astronaut is accelerated on a parallelogram motion guide which rotates at a large radius to achieve a nearly linear trajectory.The acceleration is calculated by regression analysis of the displacement versus time trajectory and the body mass is calculated by using the formula m=F/a.However,in actual flight,the device is instable that the deviation between runs could be 6-7 kg.This paper considers the body non-rigidity as the major cause of error and instability and analyzes the effects of body non-rigidity from different aspects.Body non-rigidity makes the acceleration of the center of mass (C.M.) oscillate and fall behind the point where force is applied.Actual acceleration curves showed that the overall effect of body non-rigidity is an oscillation at about 7 Hz and a deviation of about 25%.To enhance body rigidity,better body restraints were introduced and a prototype based on linear acceleration method was built.Measurement experiment was carried out on ground on an air table.Three human subjects weighing 60-70 kg were measured.The average variance was 0.04 kg and the average measurement error was 0.4%.This study will provide reference for future development of China's own mass measurement device.YAN Hui LI LuMing HU ChunHua CHEN Hao HAO HongWei 2011Science China(Physics,Mechanics & Astronomy)2011,54,4:6

10Carbothermic reduction of vanadium titanomagnetite with the assistance of sodium carbonate显示文摘The carbothermic reduction of vanadium titanomagnetite concentrate(VTC)with the assistance of Na_(2)CO_(3)was conducted in an argon atmosphere between 1073 and 1473 K.X-ray diffraction and scanning electron microscopy were used to investigate the phase transformations during the reaction.By investigating the reaction between VTC and Na_(2)CO_(3),it was concluded that molten Na_(2)CO_(3)broke the structure of titanomagnetite by combining with the acidic oxides(Fe_(2)O_(3),TiO_(2),Al_(2)O_(3),and SiO_(2))to form a Na-rich melt and release FeO and MgO.Therefore,Na_(2)CO_(3)accelerated the reduction rate.In addition,adding Na_(2)CO_(3)also benefited the agglomeration of iron particles and the slag–metal separation by decreasing the viscosity of the slag.Thus,Na_(2)CO_(3)assisted carbothermic reduction is a promising method for treating VTC at low temperatures.Luming Chen Yulan Zhen Guohua Zhang Desheng Chen Lina Wang Hongxin Zhao Fancheng Meng Tao Qi 2022International Journal of Minerals,Metallurgy and Materials2022,29,2:5
11shRNA-mediated Slc38a1 silencing inhibits migration, but not invasiveness of human pancreatic cancer cells显示文摘Objective:Early metastasis is a major biological feature of pancreatic cancer.The current study examined whether silencing Slc38a1,a gene involved in energy metabolism,using short hairpin RNA(shRNA)could inhibit the growth,migration,and invasiveness of pancreatic cancer cells.Methods:A series of Slc38a1 shRNAs were designed and cloned into the pGPU6/GFP/Neo vectors.An shRNA with the most efficacious inhibitory action on SCL38A1 expression(65%inhibition)upon screening in DH5αbacteria was used to transfect SW1990 human pancreatic cancer cells.Cell growth,migration,and invasiveness were examined using cell counting kit-8,Boyden chamber without and with Matrigel,respectively.Results:Transfection of SW1990 cells with the SLCs38A1 shRNA significantly decreased the proliferation(P<0.0001)and migratory potential(by 46.7%,P=0.0399)of the cancer cells.Invasiveness,however,was not affected.Conclusions:Inhibiting Slc38a1 using shRNA technology could decrease the growth and migration of representative pancreatic cancer cells.However,the fact that invasiveness was not affected suggested that SLC38A1 is unlikely to be responsible for early metastasis.Jing Xie Zhen Chen Luming Liu Ping Li Xiaoyan Zhu Huifeng Gao Zhiqiang Meng 2013Chinese Journal of Cancer Research2013,25,5:5

13Tunable and switchable harmonic h-shaped pulse generation in a 3.03 km ultralong mode-locked thulium-doped fiber laser显示文摘We experimentally demonstrated a type of tunable and switchable harmonic h-shaped pulse generation in a thulium-doped fiber(TDF) laser passively mode locked by using an ultralong nonlinear optical loop mirror.The total cavity length was ~3.03 km, the longest ever built for a TDF laser to our best knowledge, which resulted in an ultralarge anomalous dispersion over -200 ps^2 around the emission wavelength. The produced h-shaped pulse can operate either in a fundamental or in a high-order harmonic mode-locking(HML) state depending on pump power and intra-cavity polarization state(PS). The pulse duration, no matter of the operation state, was tunable with pump power. However, pulse breaking and self-organizing occurred, resulting in high-order HML,when the pump power increased above a threshold. At a fixed pump power, the order of HML was switchable from one to another by manipulating the PS. Switching from the 8 th up to the 48 th order of HML was achieved with a fixed pump power of ~4.15 W. Our results revealed the detailed evolution and switching characteristics of the HML and individual pulse envelope with respect to both the pump power and PS. We have also discussed in detail the mechanisms of both the h-shaped pulse generation and the switching of its HML. 14Proteasomal deubiquitinase UCH37 inhibits degradation of β-catenin and promotes cell proliferation and motility显示文摘The ubiquitin–proteasome system degrades most cellular proteins in eukaryotes.UCH37,also known as UCH-L5,is a deubiquitinase binding to Rpn13,a receptor for ubiquitinated substrates in the 26 S proteasome.But,it remains unclear how UCH37 influences the proteasomal degradation of the ubiquitinated substrates.Because deletion of UCH37 is embryonically lethal in mice,this study aims to investigate the role of UCH37 in proteasomal degradation by constructing the UCH37-deficient cell lines using CRISPR/Cas9 technology.Our results demonstrated that deletion of UCH37 decreased the levels of proteasomal Rpn13,implying that UCH37 might facilitate incorporation of Rpn13 into the proteasome.Meanwhile,deletion of UCH37 decreased the levels of β-catenin and the early endosomal protein Rab8.β-Catenin interacts with TCF/LEF to control transcription,and is involved in development,tissue homeostasis and tumorigenesis.We further found that deletion of UCH37 increased the levels of the ubiquitinated β-catenin and accelerated the hydrogen peroxide-stimulated degradation of β-catenin.Deletion of UCH37 also down-regulated the transcription of c-Myc,a downstream effector of β-catenin,and inhibited cell proliferation and motility.These results raise the possibility that UCH37 maintains the homeostasis of proteasomal degradation reciprocally by assisting the recruitment of the ubiquitin receptor Rpn13 into the proteasome and by reversing ubiquitination of certain critical substrates of the 26 S proteasome.Zijian Li Luming Zhou Tianxia Jiang Libin Fan Xiaoying Liu Xiaobo Qiu 2019Acta Biochimica et Biophysica Sinica2019,51,3:4
15Damage assessment for seismic response of recycled concrete filled steel tube columns显示文摘A model for evaluating structural damage of recycled aggregate concrete filled steel tube(RCFST) columns under seismic effects is proposed in this paper. The proposed model takes the lateral deformation and the effect of repeated cyclic loading into account. Available test results were collected and utilized to calibrate the parameters of the proposed model. A seismic test for six RCFST columns was also performed to validate the proposed damage assessment model. The main test parameters were the recycled coarse aggregate(RCA) replacement percentage and the bond-slip property. The test results indicated that the seismic performance of the RCFST member depends on the RCA contents and their damage index increases as the RCA replacement percentage increases. It is also indicated that the damage degree of RCFST changes with the variation of the RCA replacement percentage. Finally, comparisons between the RCA contents, lateral deformation ratio and damage degree were implemented. 16Anti-tumor effect of the extract from Qingyihuaji formula on pancreatic cancer by down-regulating Notch-4 and Jagged-1显示文摘OBJECTIVE: To investigate, in terms of Notch signaling pathway, the effect on pancreatic cancer of the extract of an anti-tumor prescription — Qingyihuaji formula(QYHJ) — from Traditional Chinese Medicine(TCM).METHODS: Nude mice were implanted subcutaneously with human pancreatic cancer cell line SW1990 and then randomly divided into four groups: Control, QYHJ extract, Gemcitabine, and Combination of QYHJ extract and gemcitabine.Treatments were given for 21 days and tumor growth was evaluated simultaneously. Then, expression of Notch receptors(Notch-1, Notch-2,Notch-3, and Notch-4) and their Jagged ligands(Jagged-1 and Jagged-2) in dissected tumor tissuewere detected by real-time quantitative reverse transcription-polymerase chain reaction and Western blot. Finally, immunohistochemistry was performed to detect CD133, a marker of pancreatic cancer stem cells(CSCs), to evaluate the impact of QYHJ extract on pancreatic CSCs.RESULTS: QYHJ extract treatment effectively inhibited the tumor growth in nude mice. The expression of both Notch-4 and Jagged-1 were decreased significantly in QYHJ treatment groups(P < 0.05), while gemcitabine alone had no significant effect in down-regulating Jagged-1(P > 0.05).No significant difference was observed in the expression of Notch-1, Notch-2, Notch-3, and Jagged-2 between three treatment groups and control group(P > 0.05). Moreover, immunohistochemical analysis showed that the number of CD133 positive cells was significantly reduced by QYHJ treatment(P < 0.05), and the combined treatment was more effective than gemcitabine alone(P < 0.05).CONCLUSION: The role of the extract in pancreatic cancer treatment was associated with down-regulation of Notch-4 and Jagged-1 in Notch signaling pathway. 17Nickel catalysts supported on MgO with different specific surface area for carbon dioxide reforming of methane显示文摘In this paper, three kinds of MgO with different specific surface area were prepared, and their effects on the catalytic performance of nickel catalysts for the carbon dioxide reforming of methane were investigated. The results showed that MgO support with the higher specific surface area led to the higher dispersion of the active metal, which resulted in the higher initial activity. On the other hand, the specific surface area of MgO materials might not be the dominant factor for the basicity of support to chemisorb and activate CO2, which was another important factor for the performance of catalysts. 20Real-time dynamics of soliton triplets in fiber lasers显示文摘The evolution of soliton molecules emphasizes the complex soliton dynamics akin to matter molecules.Beyond the simplest soliton molecule-a soliton pair constituted by two bound pulses-soliton molecules with more constituents have more degrees of freedom because of the temporal pulse separations and relative phases.Here we detailedly characterize the transient dynamics of soliton triplets in fiber lasers by using the dispersive Fourier transform measurement.A particular form of leading,central,and tailing pulses is constructed to shed new light on more intriguing scenarios and fuel the molecular analogy.Especially the vibrating dynamics of the central and tailing pulses are captured near the regime of equally spaced soliton triplets,which is reminiscent of the recurrent timing jitters within multi-pulse structures.Further insights enable acess into a universal form of unequally spaced soliton triplets interpreted as 2+1 soliton molecules.Different binding strengths of intramo-lecular and intermolecular bonds are validated with respect to the diverse internal motions involved in this soliton triplet molcule.All these findings unveil the transient dynamics with more degrees of freedom as well as highlight the possible application for all-optical bit storage.YIYANG LUO RAN XIA PERRY PING SHUM WENJUN NI YUSONG LIU HUY QUOC LAM QIZHEN SUN XIAHUI TANG LUMING ZHAO 2020Photonics Research2020,8,6:3
