1CRISPR/Cas9-mediated multiple gene editing in Brassica oleracea var.capitata using the endogenous tRNA-processing system显示文摘Cabbage(Brassica oleracea var.capitata)is a biennial plant with strong self-incompatibility and an obligate requirement for prolonged vernalization by exposure to low temperatures to induce flowering.These characteristics significantly increase the difficulty of exploiting novel germplasm induced by physical or chemical mutagens.In this study,we report a CRISPR/Cas9 gene-editing system based on endogenous tRNA processing to induce high efficiency and inheritable mutagenesis in cabbage.Using the phytoene desaturase gene BoPDS,the S-receptor kinase gene BoSRK,and the male-sterility-associated gene BoMS1 as the target genes,multisite and multiple gene mutations were achieved using a construct with tandemly arrayed tRNA-sgRNA architecture to express multiple sgRNAs.The BoSRK3 gene mutation suppressed self-incompatibility completely,converting the self-incompatible line into a self-compatible line.In addition,the BoMS1 gene mutation produced a completely male-sterile mutant,which was highly cross compatible with its nonmutant isoline at the flowering stage as a result of a simultaneous BoSRK3 gene mutation,enabling the economic propagation of the male-sterile line through bee-mediated cross-pollination.Interestingly,higher site mutation efficiency was detected when a guide sequence was inserted into a location in the tandemly arrayed tRNA-sgRNA architecture that was distal from the upstream Pol III promoter.In addition,mutation sites were also detected in the paralogous genes of the BoPDS and BoSRK genes that had fully consistent sequences or base mismatches but beyond the“seed”region in the spacer sequence compared with the target sgRNAs.Collectively,our results demonstrate that the CRISPR/Cas9 system,coupled with an endogenous tRNA-processing system,is an efficient tool to improve cabbage traits.Cunfa Ma Chenzeng Zhu Min Zheng Mengci Liu Dejun Zhang Baoli Liu Qinfei Li Jun Si Xuesong Ren Hongyuan Song 2019Horticulture Research2019,6,1:8
2Nondestructive detection of infertile hatching eggs based on spectral and imaging information显示文摘In order to quickly distinguish infertile eggs from fertile eggs,the hyperspectral imaging technology consisting of imaging and spectral information was used for detecting the fertile information of eggs.Before hatching eggs were incubated,a hyperspectral imaging system(wavelength between 400 to 1000 nm)was used to acquire the images one-by-one manually.The characteristic information of ratios of length to short axis,elongation,roundness and the ratios of the yolk area to the whole area was extracted based on the images.The normalization method was used as the spectral data preprocessing,and then 155 spectral characteristic variables were extracted from 520 nm waveband through the correlation coefficient method.Principal component analysis(PCA)method was adopted to reduce the dimensions of image-spectrum fusion information;the top six principal components were extracted.Support vector machine(SVM)method was used to establish classification of fertile and infertile eggs models,which are based on image,spectrum and image-spectrum fusion information respectively.The accuracy rates of the SVM models were 84.00%,90.00%and 93.00%respectively.The experimental results show that the model based on image-spectrum fusion information technology is superior to the single information model.Hyperspectral transmission imaging technology is effective and feasible to detect the fertile hatching eggs before incubation.Zhu Zhihui Liu Ting Xie Dejun Wang Qiaohua Ma Meihu 2015International Journal of Agricultural and Biological Engineering2015,8,4:4
3The effects of vertical viscosity coefficients with different distribution characteristics on classical Ekman spiral structure显示文摘The classical Ekman theory tells us that the ocean surface current turns to the right(left) side of wind direction with 45° in the north(south) hemisphere,but the observation and research results show that the surface current deflexion angle is smaller than 45° in the Arctic and high latitude areas while larger than 45° in the low latitude areas.In order to explain these phenomena,a series of idealized numerical experiments are designed to investigate the influence of vertical viscosity coefficients with different vertical distribution characteristics on the classical and steady Ekman spiral structure.Results show that when the vertical viscosity coefficient decreases with water depth,the surface current deflexion angle is larger than 45°,whereas the angle is smaller than 45° when the vertical viscosity coefficient increases with water depth.So the different observed surface current deflexion angles in low latitude sea areas and the Arctic regions should be attributed to the different vertical distribution characteristics of vertical viscosity coefficients in the upper ocean.The flatness of the Ekman spiral is not equal to one and does not show regular behaviors for the numerical experiments with different distribution of vertical viscosity.However,the magnitudes and directions of volume transport of Ekman spirals are almost the same as the results of classical Ekman theory,i.e.,vertical viscosity coefficient distributions have no effect on the magnitudes and directions of volume transport.MA HongYu QIAO FangLi DAI DeJun 2014Science China Earth Sciences2014,57,4:2

8Unraveling the stabilization mechanism of solid electrolyte interface on ZnSe by rGO in sodium ion battery显示文摘Transition metal selenides have been widely studied as anode materials of sodium ion batteries(SIBs),however,the investigation of solid-electrolyte-interface(SEI)on these materials,which is critical to the electrochemical performance of SIBs,remains at its infancy.Here in this paper,ZnSe@C nanoparticles were prepared from ZIF-8 and the SEI layers on these electrodes with and without reduced graphene oxide(rGO)layers were examined in details by X-ray photoelectron spectroscopies at varied charged/discharged states.It is observed that fast and complicated electrolyte decomposition reactions on ZnSe@C leads to quite thick SEI film and intercalation of solvated sodium ions through such thick SEI film results in slow ion diffusion kinetics and unstable electrode structure.However,the presence of rGO could efficiently suppress the decomposition of electrolyte,thus thin and stable SEI film was formed.ZnSe@C electrodes wrapped by rGO demonstrates enhanced interfacial charge transfer kinetics and high electrochemical performance,a capacity retention of 96.4%,after 1000 cycles at 5 A/g.This study might offer a simple avenue for the designing high performance anode materials through manipulation of SEI film.Shuang Men Hui Zheng Dejun Ma Xiaolian Huang Xiongwu Kang 2021Journal of Energy Chemistry2021,30,3:1

13NUMERICAL STUDY OF TWO INTERFACE METHODS FOR COMPRESSIBLE MULTI-FLUID FLOWS显示文摘Two interface capturing methods are studied for multi fluid flows, governed by the stiffened gas equation of state. The mixture type interface capturing algorithm uses a simple volume fraction model Euler equations written in a quasi conservative form, which is solved by a standard high resolution piecewise parabolic method (PPM) with multi fluid Riemann solver. The level set interface capturing method uses a narrow band ghost fluid method (GFM) with no numerical smearing. 15Topography modulates effects of nitrogen deposition on microbial resource limitation in a nitrogen-saturated subtropical forest显示文摘Background:Nitrogen(N)saturation theory proposes that an ecosystem might switch from N limitation to carbon(C),phosphorus(P),or other nutrient limitations if it receives continuous N input.Yet,after N limitation is removed,which nutrient is the most limited and whether topography modulates such change is rarely tested at a microbial level.Here,we conducted a two-year N addition experiment under two different topography positions(i.e.a slope and a valley)in a N-saturated subtropical forest.Soil enzyme activity was measured,and ecoenzymatic stoichiometry indexes were calculated as indicators of microbial resource limitation.Results:In the valley,two-year N addition changed the activity of all studied enzymes to various degrees.As a result,microbial C limitation was aggravated in the valley,and consequently microbial decomposition of soil labile organic C increased,but microbial P limitation was alleviated due to the stoichiometry balance.On the slope,however,N addition did not significantly change the activity of the studied enzymes,and did not alter the status of microbial resource limitation.Conclusions:These results indicate that C is a more limited element for microbial growth than P after removing N limitation,but we also highlight that topography can regulate the effect of N deposition on soil microbial resource limitation in subtropical forests.These findings provide useful supplements to the N saturation theory.Hao Chen Junjie Tang Xibin Sun Kayan Ma Huaihai Chen Dejun Li 2021Forest Ecosystems2021,8,4:0
16Excess PbI_(2)evolution for triple-cation based perovskite solar cells with 21.9%efficiency显示文摘The triple cation mixed perovskites(Cs FAMA)are known as one of the most efficient candidates for perovskite solar cells(PSCs).It is found that the power conversion efficiency(PCE)of triple-cation based devices would increase with the test time extending,and the maximum efficiency is normally obtained after several days aging storage.Here,the relationship between enhanced device performance,excess PbI_(2)and its evolution in triple cation perovskite films of initial days was systematically explored.The Cs FAMA-PSCs are prepared by two-step methods under two environmental conditions,including in the glove box and the ambient air(30%humidity).After 7 days testing,the maximum PCE of PSCs under two conditions dramatically increased 12.4%and 12.2%,reached 21.68%and 21.89%,respectively.At initial days,the XRD peak intensities of perovskite phase gradually decreased and those corresponding to PbI_(2)increased.Along with time-resolved photoluminescence(TRPL)and kelvin probe force microscopy(KPFM),it was found that the defects were passivated with the evolution of PbI_(2).This work reveals the excess PbI_(2)and its evolution in perovskite film,which can further supplement the understanding of PbI_(2)defect passivation.Zhu Ma Dejun Huang Qianyu Liu Guangyuan Yan Zheng Xiao Dong Chen Jiaxuan Zhao Yan Xiang Changtao Peng Haijin Li Meng Zhang Wenfeng Zhang Lianfeng Duan Yuelong Huang 2022Journal of Energy Chemistry2022,31,3:0
17Transmission of Carbapenem Resistance Between Human and Animal NDM-Positive Escherichia coli Strains显示文摘Although carbapenem use is prohibited in animals in China,carbapenem-resistant Escherichia coli(CREC),especially New Delhi metallo-β-lactamase(NDM)-producing strains,are widely prevalent in foodproducing animals.At present,the impact of livestock-associated CREC strains on human populations at the national level is unknown.Here,we conduct a retrospective cross-sectional study to investigate the prevalence of CREC from clinical settings across 22 Chinese provinces or municipalities and analyze anthropogenic factors associated with their presence.We also ascertain the blaNDMand blaKPCabundance among pig and chicken farms and present a detailed genomic framework for CREC of animal and human origin.Overall,631/29799(2.1%)clinical Escherichia coli(E.coli)isolates were identified as CREC.Multivariable analysis revealed that being male,an age below 1,an age between 13 and 18,provinces with greater chicken production,and provinces with higher pig production were associated with higher odds of CREC infection.In general,73.8%(n=45/61)of pig farms and 62.2%(n=28/45)of chicken farms had a blaNDMabundance of 1×10^(-5)to 1×10^(-3)and 1×10^(-3)to 1×10^(-2),respectively.Among all the Chinese NDM-positive E.coli(n=463)available at the National Center for Biotechnology Information(NCBI),the genomic analysis revealed that blaNDM-5and Inc X3 were the predominant carbapenemase gene-plasmid combination,while a highly homogeneous relationship between NDM-positive isolates from humans and animals was demonstrated at the plasmid and core genome levels.All the findings suggest frequent CREC transmission between humans and animals,indicating that further discussions on the use of antibiotics in animals and humans are needed,both in China and across the globe.Yingbo Shen Fupin Hu Yongqiang Wang Dandan Yin Lu Yang Yiqiang Chen Chunyan Xu Jiyun Li Junyao Jiang Xueyang Wang Yulin Fu Dongyan Shao Dejun Liu Tengfei Ma Chang Cai Zhangqi Shen Shaolin Wang Juan Li Rong Zhang Yuebin Ke Congming Wu Jianzhong Shen Timothy R.Walsh Yang Wang 2022Engineering2022,8,8:0
18Numerical Simulation of a Multi-Frequency Resistivity Logging-While-Drilling Tool Using a Highly Accurate and Adaptive Higher-Order Finite Element Method显示文摘A novel,highly efficient and accurate adaptive higher-order finite element method(hp-FEM)is used to simulate a multi-frequency resistivity loggingwhile-drilling(LWD)tool response in a borehole environment.Presented in this study are the vector expression of Maxwell’s equations,three kinds of boundary conditions,stability weak formulation of Maxwell’s equations,and automatic hpadaptivity strategy.The new hp-FEM can select optimal refinement and calculation strategies based on the practical formation model and error estimation.Numerical experiments show that the new hp-FEM has an exponential convergence rate in terms of relative error in a user-prescribed quantity of interest against the degrees of freedom,which provides more accurate results than those obtained using the adaptive h-FEM.The numerical results illustrate the high efficiency and accuracy of the method at a given LWD tool structure and parameters in different physical models,which further confirm the accuracy of the results using the Hermes library(http://gffzz741bd96fb8484393hb9vu6q00x9p66ufq.ffgz.tsg.suse.edu.cn/hermes)with a multi-frequency resistivity LWD tool response in a borehole environment.Zhonghua Ma Dejun Liu Hui Li Xinsheng Gao 2012Advances in Applied Mathematics and Mechanics2012,4,4:0
