| 3 | The study on the mechanical properties of PU/MF double shell self-healing microcapsules显示文摘The self-healing microcapsules can be buried in the coating to improve the anticorrosive ability.In this paper,self-healing microcapsules of polyurea(PU)/melamine resin(MF)double shell were prepared by in-situ polymerization and interfacial polymerization with isocyanate as the core material.Scanning electron microscope was used to observe the microcapsule morphology.The structures of microcapsules prepared with different chain extenders were characterized by Fourier transform infrared spectroscopy.The micromanipulation system was used to loading–holding,loading–unloading and loading to rupture individual microcapsules,so as to explore the mechanical properties of microcapsules.The Young’s modulus corresponding to microcapsules was calculated by mathematical model fitting.The self-healing properties of microcapsule coating were characterized by optical microscope.The experimental results showed that the microcapsule shell prepared under optimized conditions had a complete morphology and good mechanical properties.The microcapsule was in the elastic deformation stage under small deformation,and the plastic deformation stage under large deformation.The Young’s modulus range of microcapsules was 9.29–14.51 MPa,and the corresponding Young’s modulus could be prepared by adjusting the process.The surface crack of the coating containing microcapsule could heal itself after48 h in a humid environment. | Guohao Du Jianfeng Hu Jianhui Zhou Guangwu Wang Shengli Guan Hailing Liu Man Geng Chuang Lv Yaoqiang Ming Jinqing Qu | 2020 | Chinese Journal of Chemical Engineering2020,28,5: | 4 |
| 4 | Metabolic regulation of T cell development by Sinl-mTORC2 is mediated by pyruvate kinase M2显示文摘Glucose metabolism plays a key role in thymocyte development. The mammalian target of rapamycin complex 2 (mT0RC2) is a critical regulator of cell growth and metabolism, but its role in early thymocyte development and metabolism has not been fully studied. We show here that genetic ablation of Sinl, an essential component of mTORC2, in T lineage cells results in severely impaired thymocyte development at the CD4^- CD8^- double negative (DN) stages but not at the CD4^+ CD8^+ double positive (DP) or later stages. Notably, Sinl-deficient DN thymocytes show markedly reduced proliferation and glycolysis.Importantly, we discover that the M2 isoform of pyruvate kinase (PKM2) is a novel and crucial Sinl effector in promoting DN thymocyte development and metabolism. At the molecular level, we show that Sinl-mTORC2 controls PKM2 expression through an AKT-dependent PPAR-y nuclear translocation. | 5 | Reproductive toxicity and underlying mechanisms of di(2-ethylhexyl) phthalate in nematode Caenorhabditis elegans显示文摘DEHP(di(2-ethylhexyl) phthalate) is an endocrine disruptor commonly found in plastic products that has been associated with reproduction alterations, but the effect of DEHP on toxicity is still widely unknown. Using DEHP concentrations of 10, 1, and 0.1 mg/L, we showed that DEHP reduced the reproductive capacity of Caenorhabditis elegans after 72 hr. of exposure. DEHP exposure reduced the reproductive capacity in terms of decreased brood sizes, egg hatchability(0.1, 1 and 10 mg/L), and egg-laying rate(1 and 10 mg/L), and increased numbers of fertilized eggs in the uterus(1 and 10 mg/L). DEHP also caused damage to gonad development. DEHP decreased the total number of germline cells, and decreased the relative area of the gonad arm of all exposure groups, with worms in the 1 mg/L DEHP exposure group having the minimum gonad arm area. Additionally, DEHP caused a significant concentration-dependent increase in the expression of unc-86. Autophagy and ROS contributed to the enhancement of DEHP toxicity in reducing reproductive capacity, and glutathione peroxidase and superoxide dismutase were activated as the antioxidant defense in this study. Hence, we found that DEHP has a dual effect on nematodes. Higher concentration(10 mg/L) DEHP can inhibit the expression of autophagy genes( atg-18, atg-7, bec-1, lgg-1 and unc-51), and lower concentrations(0.1 and 1 mg/L) can promote the expression of autophagy genes. | 6 | Optimization of Hydrolysis Conditions for the Isolation of Angiotensin-I Converting Enzyme(ACE) Inhibitory Peptides from Rhopilema hispidum显示文摘To obtain the maximum angiotensin-I converting enzyme(ACE) inhibitory activity, the protein hydrolysis conditions of the jellyfish Rhopilema hispidum were optimized using response surface methodology(RSM). Trypsin was selected to produce R. hispidum protein hydrolysates(RPH) with ACE inhibitory activity. The optimal parameters for producing protein hydrolysates with the highest ACE inhibitory activity were as follows: hydrolysis time 5 h, hydrolysis temperature 50℃, and the enzyme-to-substrate ratio 6%. Under these conditions, the ACE inhibitory rate of RPH could reach 64.28% ± 5.72%. In addition, RPH contained high levels of Gly, Glu, Pro, Ala, Asp and Arg, with a molecular weight distribution range of 0.32–6.84 kDa. The following three novel ACE inhibitory peptides were isolated and identified: Ile-Gly-Glu-Thr-Gly-Pro, Gly-Ala-Thr-Gly-Pro-Ala-Gly-Tyr-Val and Gly-AlaPhe-Gly-Pro-Gly-Gly-Leu-Val-Gly-Arg-Pro. The IC_(50) values of the ACE inhibitory activity of these three purified peptides were 19.07, 27.42 and 31.26 μmol L^(-1), respectively. Results showed that 2.5%(mass fraction)Li 2O would be needed to rule out the voltage pulse in the first part of discharge curves for the FeS 2 powder of small particle size (<44 μm). After thermal decomposition, the FeS 2 had transformed to Fe (1- x ) S where x =0.024~0.066. The deficiency of the cathode FeS 2 would make discharge voltage decrease 0.4 V. | 11 | 2-(2-Benzofuranyl)-2-imidazoline treatment within 5 hours after cerebral ischemia/reperfusion protects the brain显示文摘We previously demonstrated that administering 2-(2-benzofuranyl)-2-imidazolin(2-BFI), an imidazoline I2 receptor agonist, immediately after ischemia onset can protect the brain from ischemic insult. However, immediate administration after stroke is difficult to realize in the clinic. Thus, the therapeutic time window of 2-BFI should be determined. Sprague-Dawley rats provided by Wenzhou Medical University in China received right middle cerebral artery occlusion for 120 minutes, and were treated with 2-BFI(3 mg/kg) through the caudal vein at 0, 1, 3, 5, 7, and 9 hours after reperfusion. Neurological function was assessed using the Longa's method. Infarct volume was measured by 2,3,5-triphenyltetrazolium chloride assay. Morphological changes in the cortical penumbra were observed by hematoxylin-eosin staining under transmission electron microscopy. The apoptosis levels in the ipsilateral cortex were examined with terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling(TUNEL) assay. The protein expression of Bcl-2 and BAX was detected using immunohistochemistry. We found the following: Treatment with 2-BFI within 5 hours after reperfusion obviously improved neurological function. Administering 2-BFI within 9 hours after ischemia/reperfusion decreased infarct volume and alleviated apoptosis. 2-BFI administration at different time points after reperfusion alleviated the pathological damage of the ischemic penumbra and reduced the number of apoptotic neurons, but the protective effect was more obvious when administered within 5 hours. Administration of 2-BFI within 5 hours after reperfusion remarkably increased Bcl-2 expression and decreased BAX expression. To conclude, 2-BFI shows potent neuroprotective effects when administered within 5 hours after reperfusion, seemingly by up-regulating Bcl-2 and down-regulating BAX expression.
