1Current understanding of the molecular mechanisms in Parkinson's disease:Targets for potential treatments显示文摘Gradual degeneration and loss of dopaminergic neurons in the substantia nigra,pars compacta and subsequent reduction of dopamine levels in striatum are associated with motor deficits that characterize Parkinson’s disease(PD).In addition,half of the PD patients also exhibit frontostriatal-mediated executive dysfunction,including deficits in attention,short-term working memory,speed of mental processing,and impulsivity.The most commonly used treatments for PD are only partially or transiently effective and are available or applicable to a minority of patients.Because,these therapies neither restore the lost or degenerated dopaminergic neurons,nor prevent or delay the disease progression,the need for more effective therapeutics is critical.In this review,we provide a comprehensive overview of the current understanding of the molecular signaling pathways involved in PD,particularly within the context of how genetic and environmental factors contribute to the initiation and progression of this disease.The involvement of molecular chaperones,autophagy-lysosomal pathways,and proteasome systems in PD are also highlighted.In addition,emerging therapies,including pharmacological manipulations,surgical procedures,stem cell transplantation,gene therapy,as well as complementary,supportive and rehabilitation therapies to prevent or delay the progression of this complex disease are reviewed.Panchanan Maiti Jayeeta Manna Gary L.Dunbar 2017Translational Neurodegeneration2017,6,1:27

4Hepatorenal syndrome: Update on diagnosis and treatment显示文摘Acute kidney injury(AKI) is a common complication in patients with end-stage liver disease and advanced cirrhosis regardless of the underlying cause. Hepatorenal syndrome(HRS), a functional form of kidney failure, is one of the many possible causes of AKI. HRS is potentially reversible but involves highly complex pathogenetic mechanisms and equally complex clinical and therapeutic management. Once HRS has developed, it has a very poor prognosis. 6Potential advantages of acute kidney injury management by mesenchymal stem cells显示文摘Mesenchymal stem cells are currently considered as a promising tool for therapeutic application in acute kidney injury(AKI) management. AKI is characterized by acute tubular injury with rapid loss of renal function. After AKI, inflammation, oxidative stress and excessive deposition of extracellular matrix are the molecular events that ultimately cause the end-stage renal disease. Despite numerous improvement of supportive therapy, the mortality and morbidity among patients remain high. Therefore, exploring novel therapeutic options to treat AKI is mandatory. Numerous evidence in animal models has demonstrated the capability of mesenchymal stem cells(MSCs) to restore kidney function after induced kidney injury. After infusion, MSCs engraft in the injured tissue and release soluble factors and microvesicles that promote cell survival and tissue repairing. Indeed, the main mechanism of action of MSCs in tissue regeneration is the paracrine/endocrine secretion of bioactive molecules. MSCs can be isolated from several tissues, including bone marrow, adipose tissue, and blood cord; pre-treatment procedures to improve MSCs homing and their paracrine function have been also described. 8Renal function and physical fitness after 12-mo supervised training in kidney transplant recipients显示文摘AIM To evaluate the effect of a 12-mo supervised aerobic and resistance training, on renal function and exercise capacity compared to usual care recommendations.METHODS Ninety-nine kidney transplant recipients(KTRs) were assigned to interventional exercise(Group A; n = 52) and a usual care cohort(Group B; n = 47). Blood and urine chemistry, exercise capacity, muscular strength, anthropometric measures and health-related quality of life(HRQo L) were assessed at baseline, and after 6 and 12 mo. Group A underwent a supervised training three times per week for 12 mo. Group B received only general recommendations about home-based physical activities.RESULTS Eighty-five KTRs completed the study(Group A, n = 44; Group B, n = 41). After 12 mo, renal function remained stable in both groups. Group A significantly increased maximum workload(+13 W, P = 0.0003), V'O2 peak(+3.1 mL/kg per minute, P = 0.0099), muscular strength in plantar flexor(+12 kg, P = 0.0368), height in the countermovement jump(+1.9 cm, P = 0.0293) and decreased in Body Mass Index(-0.5 kg/m^2, P = 0.0013). HRQo L significantly improved in physical function(P = 0.0019), physical-role limitations(P = 0.0321) and social functioning scales(P = 0.0346). Noimprovements were found in Group B.CONCLUSION Twelve-month of supervised aerobic and resistance training improves the physiological variables related to physical fitness and cardiovascular risks without consequences on renal function. Recommendations alone are not sufficient to induce changes in exercise capacity of KTRs. 10Retroperitoneal paragangliomas : 4 个案例的报告显示文摘 We reviewed the data of all patients managed for retroperitoneal paragangliomas(PGLs)between June 2010and June 2011 to present our experience concerning this uncommon entity to highlight diagnostic and therapeutic challenges of retroperitoneal PGLs.All patients were admitted to the department of general and hepatobiliary surgery in the regional hospital of Jendouba,Tunisia.The size of the tumor was taken at its largest dimension,as determined in a computed tomography(CT)scan and pathological reports.There were 4 patients(all women)with a median age of 48 years(range46-56 years).Abdominal pain was the commonest presentation.CT showed and localized the tumors which were all retroperitoneal.All patients had successfu surgical resection of the tumors under invasive arterial blood pressure monitoring.One patient underwent surgery for a presumed tumor of the pancreatic head.The fresh-mount microscopic study of the peroperative biopsy yielded inflammatory tissue without malignancy and no resection was performed.Final histologica examination of the biopsy concluded PGL.A secondlaparotomy was performed and the tumor was entirely resected.The diagnosis was made after surgery by histology in all patients.The control of the blood pressure was improved after surgery in 3 patients.Paragangliomas are rare tumors.The retroperitoneal localization is uncommon.Complete surgical resection remains the only curative treatment but it is often challenging as these tumors are located near multiple vital blood vessels.Helmi Kallel Hassen Hentati Amine Baklouti Ali Gassara Ahmed Saadaoui Ghassen Halek Sana Landolsi MA El Ouaer Wajdi Chaieb Fethia Maamouri Saber Manna? 2014World Journal of Gastrointestinal Surgery2014,6,4:3
11Diagnostic and therapeutic potential of exosomal miRNAs in Alzheimer’s disease显示文摘Alzheimer’s disease(AD)is a primary cause of dementia.AD is a neurodegenerative disorder,characterized by synapses loss,extracellular amyloid plaques composed of the amyloid-βpeptide(Aβ)and intracellular aggregates of hyperphosphorylated tau protein.AD is a complex disease linked to multiple interacting factors,both environmental and genetic,which can contribute to the onset and severity of the disease.Longitudinal studies have highlighted several cardiovascular risk factors that can increase the risk of AD.The genetic landscape of AD has changed dramatically in recent decades.Early studies identified mutations in the amyloid precursor protein gene(APP)as well as proteins that are involved in the enzymatic cleavage of APP to toxicβ-amyloid(Aβ),namely presenilin-1 and presnilin-2.However,these mutations were found in familial cases of early-onset AD,while the causes of sporadic late-onset AD are still unknown.The latest advances in Genomewide Association Studies(GWAS),sequencing,and bioinformatics have begun to unravel the complex genetic architecture of the sporadic form of AD.GWAS were able to uncover common variants with high frequency in the population that individually carried low risk(Robinson et al.,2017).The advent of next-generation and thirdgeneration sequencing platforms shows great promise in further unravelling the genetics of AD.Ida Manna Selene De Benedittis Enrico Iaccino Andrea Quattrone Aldo Quattrone 2021Neural Regeneration Research2021,16,11:3

18Geochemical contamination in the mine affected soil of Raniganj Coalfield——A river basin scale assessment显示文摘The study aimed to assess the heavy metals(K, Ca, Ti, Cr, Mn, Fe, Ni, Cu, Zn, As, Pb, Sr, Zr) contamination in the soil of mine affected Singaran river basin and to analyse spatial variation in the contamination level considering 32 soil samples. Elemental analysis of soil samples has been performed through Energy Dispersive X-ray Analysis(EDX) to quantify the elemental concentration(mg kgà1). Heavy metal concentrations have been assessed through geo-accumulation index(Igeo) and enrichment factor(EF).Indices showed soils have moderate accumulation of most of the metals with moderate enrichment of Sr,Zr, Zn, Cu and Ni. Soil contamination level assessment has been carried out using indices like Contamination Factor(CF), degree of contamination(C_(deg)), modified degree of contamination(m C_(deg)) and Pollution Load Index(PLI). CF shows moderate to considerable contamination by Sr, Zr, Ca, Cu, Mn, Zn and Ni. Mean indices values(m C_(deg)and PLI for the entire basin are 3.38 and 2.23 respectively) show low to moderate level of soil contamination. These indices result have been mapped and analysed in GIS platform to get spatial variation of pollution level. Opencast mines dominate middle catchment area and so is comparatively contaminated. Sample sites 11, 18 and 25 evidenced high values of all indices of pollution load. From the ecological standpoint Ecological Risk Factor(Er) and Potential Ecological Risk Index(RI) have been estimated to assess regional threat to native soil environment and it shows low ecological risk potential. 19Entanglement-based quantum key distribution with a blinking-free quantum dot operated at a temperature up to 20 K显示文摘Entanglement-based quantum key distribution(QKD)promises enhanced robustness against eavesdropping and compatibility with future quantum networks.Among other sources,semiconductor quantum dots(QDs)can generate polarization-entangled photon pairs with near-unity entanglement fidelity and a multiphoton emission probability close to zero even at maximum brightness.These properties have been demonstrated under resonant two-photon excitation(TPE)and at operation temperatures below 10 K.However,source blinking is often reported under TPE conditions,limiting the maximum achievable photon rate.In addition,operation temperatures reachable with compact cryocoolers could facilitate the widespread deployment of QDs,e.g.,in satellite-based quantum communication.We demonstrate blinking-free emission of highly entangled photon pairs from GaAs QDs embedded in a p-i-n diode.High fidelity entanglement persists at temperatures of at least 20 K,which we use to implement fiber-based QKD between two buildings with an average raw key rate of 55 bits/s and a qubit error rate of 8.4%.We are confident that by combining electrical control with already demonstrated photonic and strain engineering,QDs will keep approaching the ideal source of entangled photons for real world applications.Christian Schimpf Santanu Manna Saimon F.Covre da Silva Maximilian Aigner Armando Rastelli 2021Advanced Photonics2021,3,6:2
