6Sulfation of chondroitin and bile acids converges to antagonize Wnt/β-catenin signaling and inhibit APC deficiency-induced gut tumorigenesis显示文摘Sulfation is a crucial and prevalent conjugation reaction involved in cellular processes and mammalian physiology.3’-Phosphoadenosine 5’-phosphosulfate(PAPS)synthase 2(PAPSS2)is the primary enzyme to generate the universal sulfonate donor PAPS.The involvement of PAPSS2-mediated sulfation in adenomatous polyposis coli(APC)mutation-promoted colonic carcinogenesis has not been reported.Here,we showed that the expression of PAPSS2 was decreased in human colon tumors along with cancer stages,and the lower expression of PAPSS2 was correlated with poor prognosis in advanced colon cancer.Gut epithelial-specific heterozygous Apc deficient and Papss2-knockout(Apc^(Δgut-Het)Papss2^(Δgut))mice were created,and the phenotypes were compared to the spontaneous intestinal tumorigenesis of Apc^(Δgut-Het)mice.Apc^(Δgut-Het)Papss2^(Δgutmice) were more sensitive to gut tumorigenesis,which was mechanistically accounted for by the activation of Wnt/β-catenin signaling pathway due to the suppression of chondroitin sulfation and inhibition of the farnesoid X receptor(FXR)-transducin-like enhancer of split 3(TLE3)gene regulatory axis.Chondroitin sulfate supplementation in Apc^(Δgut-Het)Papss2^(Δgutmice) alleviated intestinal tumorigenesis.In summary,we have uncovered the protective role of PAPSS2-mediated chondroitin sulfation and bile acids-FXR-TLE3 activation in the prevention of gut carcinogenesis via the antagonization of Wnt/β-catenin signaling.Chondroitin sulfate may be explored as a therapeutic agent for Papss2 deficiency-associated colonic carcinogenesis.Pengfei Xu Yue Xi Jong-Won Kim Junjie Zhu Min Zhang Meishu Xu Songrong Ren Da Yang Xiaochao Ma Wen Xie 2024Acta Pharmaceutica Sinica B2024,14,3:0
7Frustration recognition from speech during game interaction using wide residual networks显示文摘Background Although frustration is a common emotional reaction while playing games,an excessive level of frustration can negatively impact a user's experience,discouraging them from further game interactions.The automatic detection of frustration can enable the development of adaptive systems that can adapt a game to a user's specific needs through real-time difficulty adjustment,thereby optimizing the player's experience and guaranteeing game success.To this end,we present a speech-based approach for the automatic detection of frustration during game interactions,a specific task that remains under explored in research.Method The experiments were performed on the Multimodal Game Frustration Database(MGFD),an audiovisual dataset-collected within the Wizard-of-Oz framework-that is specially tailored to investigate verbal and facial expressions of frustration during game interactions.We explored the performance of a variety of acoustic feature sets,including Mel-Spectrograms,Mel Frequency Cepstral Coefficients(MFCCs),and the low-dimensional knowledge-based acoustic feature set eGeMAPS.Because of the continual improvements in speech recognition tasks achieved by the use of convolutional neural networks(CNNs),unlike the MGFD baseline,which is based on the Long Short Term Memory(LSTM)architecture and Support Vector Machine(SVM)classifier-in the present work,we consider typical CNNs,including ResNet,VGG,and AlexNet.Furthermore,given the unresolved debate on the suitability of shallow and deep networks,we also examine the performance of two of the latest deep CNNs:WideResNet and EfficientNet.Results Our best result,achieved with WideResNet and Mel-Spectrogram features,increases the system performance from 58.8%unweighted average recall(UAR)to 93.1%UAR for speech-based automatic frustration recognition.Meishu SONG Adria MALLOL-RAGOLTA Emilia PARADA-CABALEIRO Zijiang YANG Shuo LIU Zhao REN Ziping ZHAO Björn WSCHULLER 2021Virtual Reality & Intelligent Hardware2021,3,1:0
8Transgenic overexpression of steroid sulfatase alleviates cholestasis显示文摘Background and Aim:Sulfotransferase(SULT)-mediated sulfation and steroid sulfatase(STS)-mediated desulfation represent two critical mechanisms that regulate the chemical and functional homeostasis of endogenous and exogenous molecules.STS catalyzes the hydrolysis of steroid sulfates to form hydroxysteroids.Oxygenated cholesterol derivative oxysterols are known to be endogenous ligands of the liver X receptor(LXR),a nuclear receptor with anti-cholestasis activity,whereas the sulfated oxysterols antagonize LXR signaling.The conversion of sulfated oxysterols to their non-sulfated counterparts is catalyzed by STS.The aim of this study is to determine whether STS can alleviate cholestasis by increasing the activity of LXR.Methods:Liver-specific STS transgenic mice were created and subject to the lithocholic acid(LCA)-induced model of cholestasis.Results:Transgenic overexpression of STS in the liver promoted bile acid elimination and alleviated LCAinduced cholestasis.The protective effect of the STS transgene was associated with the activation of LXR and induction of LXR target genes,likely because of the increased conversion of the antagonistic oxysterol sulfates to the agonistic oxysterols.Conclusions:STS has a novel function in controlling the homeostasis of bile acids by regulating endogenous LXR ligands.Mengxi Jiang Meishu Xu Songrong Ren Kyle W.Selcer Wen Xie 2017Liver Research2017,1,1:0

10A Fast Detection Method of Network Crime Based on User Portrait显示文摘In order to quickly and accurately find the implementer of the network crime,based on the user portrait technology,a rapid detection method for users with abnormal behaviors is proposed.This method needs to construct the abnormal behavior rule base on various kinds of abnormal behaviors in advance,and construct the user portrait including basic attribute tags,behavior attribute tags and abnormal behavior similarity tags for network users who have abnormal behaviors.When a network crime occurs,firstly get the corresponding tag values in all user portraits according to the category of the network crime.Then,use the Naive Bayesian method matching each user portrait,to quickly locate the most likely network criminal suspects.In the case that no suspect is found,all users are audited comprehensively through matching abnormal behavior rule base.The experimental results show that,the accuracy rate of using this method for fast detection of network crimes is 95.9%,and the audit time is shortened to 1/35 of that of the conventional behavior audit method.Yabin Xu Meishu Zhang Xiaowei Xu 2021Journal of Information Hiding and Privacy Protection2021,3,1:0
