1Effect of dietary soy oil, glucose, and glutamine on growth performance, amino acid profile, blood profile, immunity, and antioxidant capacity in weaned piglets显示文摘Weaning stress results in gastrointestinal dysfunction and depressed performance in pigs. This study aimed to investigate the effect of soy oil, glucose, and glutamine on the growth and health of weaned piglets. Compared with those in the glutamine group, piglets in the glucose and soy oil groups had greater average daily gain, average daily feed intake, and gain: feed ratio from day 0 to 14, and gain: feed ratio for the overall period. There were no differences with regard to serum amino acids among the three groups on day 14, except glycine and threonine. The serum concentration of histidine, serine, threonine, proline, and cysteine was the highest in the glutamine group, while the content of glycine and lysine in the soy oil group on day 28 was the highest among all groups. Piglets fed with glutamine had greater serum glucose and creatinine on day 14, high-density lipoprotein on day 28, and serum IgG and IgM on day 28. Piglets in the glutamine group demonstrated lower serum total superoxide dismutase on day 14 and 28; however, they demonstrated higher total superoxide dismutase and total antioxidant capacity in the duodenum and ileum on day 14. 2Dietary high protein-induced diarrhea and intestinal inflammation by activation of NF-kB signaling in piglets显示文摘The present study aimed to investigate whether inflammation-associated responses in piglets are induced by high protein(HP)through activating nuclear factor kappa B(NF-κB)signaling.Sixteen piglets(35 d of age,Duroc×[Landrace×Yorkshire],weaned at d 21,initial BW=9.70±0.11 kg)were allocated to 18%and 26%CP(HP group)at random,comprising 8 replicate pens per treatment.The piglets were slaughtered to collect intestinal tissues when apparent,persistent,and stable diarrhea syndromes happened(on d 12).No significant differences were observed in their growth performance(P>0.05),but reduction by 19.11%,25.31%,23.64%of ADFI,ADG,and G:F,respectively was detected in the HP group.The HP group had greater(P=0.002)diarrhea rates.Furthermore,dietary HP had lower ileal villus height(VH;P=0.048),ratio of villus height to crypt depth(VH/CD ratio;P=0.016),and colonic CD(P=0.034),as well as had the trend(P=0.075)to reduce the ileal villus absorptive area.Moreover,HP diets significantly elevated the goblet cell numbers in the ileal villi(P=0.016)and colonic crypts(P<0.001)and up-regulated(P=0.012)the mRNA expression of mucin2(Muc2)in the ileum.In addition,HP diets increased the myeloperoxidase concentration in the ileum(P=0.002)and colon(P=0.007)of piglets.Dietary HP significantly down-regulated the mRNA expression of tumor necrosis factor-α(TNF-α;P<0.001)in the ileum,induced nitric oxide synthase(iNOS;P=0.040)and interleukin-22(IL-22;P=0.008)in the colon,and inclined to down-regulate interleukin-1β(IL-1β;P=0.076)expression in the colon.The relative protein abundance of Galectin-3(P=0.046)in the colon and the ratio of phosphorylation NF-κB to NF-κB(p-NF=κB/NF-κB ratio)in the ileum of HP piglets were also greater(P=0.038).These results suggest that dietary HP may cause diarrhea in piglets by activating NF-icB signaling induced intestinal inflammation.Lanmei Yin Jun Li Meiwei Wang Qiye Wang Jianzhong Li Nengshui Ding Huansheng Yang Yulong Yin 2021Animal Nutrition2021,,4:3
3UPLC-MS metabolomics provides insights into the diff erences between black- and white-shelled Pacifi c oysters Crassostrea gigas显示文摘A variety of shell colors are one of the most fundamental characteristics of molluscs,which have importantly ecological and economic signifi cance.The Pacifi c oyster Crassostrea gigas is distributed in many sea areas around the world and also an aquacultured mollusc with high nutritional value.In this study,the whole soft body and the mantle tissue of black-shelled Pacifi c oyster(BSO)and white-shelled Pacifi c oyster(WSO)with starkly diff erent melanin contents were compared,and the diff erences in physiology and metabolism between BSO and WSO were analyzed.The results of physiological indicators suggested BSO show more melanin,more dry matter,more crude lipid content,and stronger ability to scavenge free radicals than WSO.The altered metabolites of glycerophospholipids,fatty acyls,and steroids revealed diff erent regulatory mechanisms of lipids.The correlation analysis of metabolomics and previously published RNAseq data suggested that BSO and WSO mainly diff ered in the basal metabolic processes,such as lipid,amino acid and purine metabolisms.This study provides insights into the changes in the physiological indictors and the metabolites of oysters with varying melanin content.Xi CHEN Qiuyun JIANG Hongce SONG Lingling LI Chaoyi XIE Baoyu HUANG Yaqiong LIU Meiwei ZHANG Lei WEI Xiaotong WANG 2021Journal of Oceanology and Limnology2021,39,1:2
4A secreted phospholipase A_(2) (BmsPLA_(2)) regulates melanization of immunity through BmDDC in the silkworm Bombyx mori显示文摘Insect immune-associated phospholipase A_(2) (PLA_(2)) is an important target of pathogen invasion. Melanization, an effective defense response, has significant correlations with other immune responses to coordinate immune attack against invaders. However, the effect of PLA_(2) on melanization has not yet been reported in insects or other arthropods. In this work, we cloned a PLA_(2) gene (BmsPLA_(2)), and its protein had characteristic features of secreted PLA_(2) (sPLA_(2)). After injection of bacteria, BmsPLA_(2) expression and sPLA_(2) activity in hemolymph significantly increased. BmsPLA_(2) fluorescence was transferred from the cytoplasm to the cell membranes of circulating hemocytes. These results indicated that BmsPLA_(2) was related to hemolymph immunity in silkworms. Interestingly, reducing BmsPLA_(2) by RNA interference decreased melanosis (melanistic hemocytes) levels in vivo and in vitro, while BmsPLA_(2) overexpression had the opposite effect. The larval survival and melanization rate in the hemocoel both slowed depending on the PLA_(2) inhibitor dosage. These results demonstrated that BmsPLA_(2) plays a role in melanization during the immune process of silkworms. Surprisingly, the level of BmDDC matched the degree of melanization in various observations. BmDDC expression showed a significant increase, with the peak occurring later than that of BmsPLA_(2) after injection of bacteria, implying that BmsPLA_(2) was activated prior to BmDDC. Moreover, the alteration of BmsPLA_(2) by RNA interference or overexpression led to altered BmDDC levels. These results suggested that BmsPLA_(2) regulates the melanization response in silkworms through BmDDC. 5Surface activity of a series of fluoroether betaine amphoteric surfactants: Oxygen roles显示文摘As PFOS, PFOA and their derivatives were banned according to the Stockholm Convention for their potential bioaccumulation and toxicity, people attempted to substitute the legacy fluorosurfactants with short-chain ones. Although short-chain alternatives can alleviate bioaccumulation, surface activity was compromised. Fluorine industry kept seeking for effective solution. In this work, we prepared and investigated a series of fluoroether betaine surfactants for their surface activity and spreading property. The role of oxygen on surface activity was discussed. We found that insertion of oxygen atoms into fluorinated chain could increase hydrophobicity and thus enhance surface activity. The contribution of one oxygen is approximately half of that of a difluoromethylene group by experience. Moreover, introducing oxygen diversified the structure to fill in the gap of surface activity between short and long fluorosurfactants.
