| 1 | Phase I study of CAR-T cells with PD-1 and TCR disruption in mesothelin-positive solid tumors显示文摘Programmed cell death protein-1(PD-1)-mediated immunosuppression has been proposed to contribute to the limited clinical efficacy of chimeric antigen receptor T(CAR-T)cells in solid tumors.We generated PD-1 and T cell receptor(TCR)deficient mesothelin-specific CAR-T(MPTK-CAR-T)cells using CRISPR-Cas9 technology and evaluated them in a dose-escalation study.A total of 15 patients received one or more infusions of MPTK-CAR-T cells without prior lymphodepletion.No dose-limiting toxicity or unexpected adverse events were observed in any of the 15 patients.The best overall response was stable disease(2/15 patients).Circulating MPTK-CAR-T cells peaked at days 7–14 and became undetectable beyond 1 month.TCR-positive CAR-T cells rather than TCR-negative CAR-T cells were predominantly detected in effusion or peripheral blood from three patients after infusion.We further confirmed the reduced persistence of TCR-deficient CAR-T cells in animal models.Our results establish the preliminary feasibility and safety of CRISPR-engineered CAR-T cells with PD-1 disruption and suggest that the natural TCR plays an important role in the persistence of CAR-T cells when treating solid tumors. | Zhenguang Wang Na Li Kaichao Feng Meixia Chen Yan Zhang Yang Liu Qingming Yang Jing Nie Na Tang Xingying Zhang Chen Cheng Lianjun Shen Jiaping He Xun Ye Wei Cao Haoyi Wang Weidong Han | 2021 | Cellular & Molecular Immunology2021,18,9: | 7 |
| 2 | Identification and Characterization of the Populus AREB/ABF Subfamily显示文摘Abscisic acid (ABA) is a major plant hormone that plays an important role in responses to abiotic stresses. The ABA-responsive element binding protein/ABRE-binding factor (AREB/ABF) gene subfamily contains crucial transcription factors in the ABA-mediated signaling pathway. In this study, a total of 14 putative AREB/ABF members were identified in the Populus trichocarpa Torr. & Gray. genome using five AREB/ABF amino acid sequences from Arabidopsis thaliana L. as probes. The 14 putative Populus subfamily members showed high protein similarities, especially in the basic leucine zipper (bZIP) domain region. A neighbor-joining analysis combined with gene structure data revealed homology among the 14 genes. The expression patterns of the Populus AREB/ABF subfamily suggested that the most abundant transcripts of 11 genes occurred in leaf tissues, while two genes were most transcribed in root tissues. Significantly, eight Populus AREB/ABF gene members were upregulated after treatment with 100 μM exogenous ABA, while the other six members were downregulated. We identified the expression profiles of the subfamily members in Populus tissues and elucidated different response patterns of Populus AREB/ABF members to ABA stress. | 3 | Genome sequencing and phylogenetic analysis of allotetraploid Salix matsudana Koidz显示文摘Polyploidy is a common phenomenon among willow species.In this study,genome sequencing was conducted for Salix matsudana Koidz(also named Chinese willow),an important greening and arbor tree species,and the genome of this species was compared with those of four other tree species in Salicaceae.The total genome sequence of S.matsudana was 655.72 Mb in size,with repeated sequences accounting for 45.97%of the total length.In total,531.43 Mb of the genome sequence could be mapped onto 38 chromosomes using the published genetic map as a reference.The genome of S.matsudana could be divided into two groups,the A and B genomes,through homology analysis with the genome of Populus trichocarpa,and the A and B genomes contained 23,985 and 25,107 genes,respectively.4DTv combined transposon analysis predicted that allotetraploidy in S.matsudana appeared~4 million years ago.The results from this study will help reveal the evolutionary history of S.matsudana and lay a genetic basis for its breeding. | Jian Zhang Huwei Yuan Yujuan Li Yanhong Chen Guoyuan Liu Meixia Ye Chunmei Yu Bolin Lian Fei Zhong Yuna Jiang Jichen Xu | 2020 | Horticulture Research2020,7,1: | 3 |
| 4 | Greenspace’s value orientations of ecosystem service and socioeconomic service in China显示文摘Background:Natural ecosystems provide necessary services for human beings,including ecosystem service values(ESVs)and socioeconomic service values(SSVs).The value orientations of ESVs and SSVs are mainly related to people’s interaction with nature.This study reclassified greenspace from a perspective of exposed and non-exposed greenspace based on the level of interaction by people and greenspace.We applied an expert questionnaire to survey the SSVs value orientations of forestland,grassland,wetland,and water bodies,and quantitatively compared the value orientations between the ESVs and SSVs of greenspace in China.Result:(1)The values of exposed greenspace were relatively far higher than nonexposed greenspace,as it had both ESVs and SSVs.(2)The forestland and grassland had relatively high ESVs and SSVs,and are the priority for both the exposed and non-exposed greenspace.(3)Wetland had relatively high ESVs but low SSVs,which was unpopular for exposed greenspace.(4)The ESVs and SSVs of water body were relatively balanced.Conclusion:Greenspace had both ESVs and SSVs when they are exposed to human.Our study provided an innovative perspective to explore the value orientations of greenspace,which provides an actionable scientific basis for greenspace planning,design and construction in human habitat. | Xia Yao Tao Lin Shoukai Sun Guoqin Zhang Hao Zhou Laurence Jones Wenhui Liu Yiyi Huang Meixia Lin Junmao Zhang Yuan Chena Hong Ye | 2022 | Ecosystem Health and Sustainability2022,8,1: | 1 |
