1Science and Prediction of Heavy Rainfall over China:Research Progress since the Reform and Opening-Up of New China显示文摘This paper reviews the major progress on development of the science and prediction of heavy rainfall over China since the beginning of the reform and opening-up of new China(roughly between 1980 and 2019).The progress of research on the physical mechanisms of heavy rainfall over China is summarized from three perspectives:1)the relevant synoptic weather systems,2)heavy rainfall in major sub-regions of China,and 3)heavy rainfall induced by typhoons.The development and application of forecasting techniques for heavy rainfall are summarized in terms of numerical weather prediction techniques and objective forecasting methods.Greatly aided by the rapid progress in meteorological observing technology and substantial improvement in electronic computing,studies of heavy rainfall in China have advanced to investigating the evolution of heavy-rain-producing storms and observational analysis of the cloud microphysical features.A deeper and more systematic understanding of the synoptic systems of importance to the production of heavy rainfall has also been developed.Operational forecast of heavy rainfall in China has changed from subjective weather event forecasts to a combination of both subjective and objective quantitative precipitation forecasts,and is now advancing toward probabilistic quantitative precipitation forecasts with the provision of forecast uncertainty information.Yali LUO Jisong SUN Ying LI Rudi XIA Yu DU Shuai YANG Yuanchun ZHANG Jing CHEN Kan DAI Xueshun SHEN Haoming CHEN Feifan ZHOU Yimin LIU Shenming FU Mengwen WU Tiangui XIAO Yangruixue CHEN Huiqi LI Mingxin LI 2020Journal of Meteorological Research2020,34,3:10
22 μm optical frequency comb generation via optical parametric oscillation from a lithium niobate optical superlattice box resonator显示文摘Optical parametric oscillators(OPOs) can downconvert the pump laser to longer wavelengths with octave separation via χ^((2)), which is widely used for laser wavelength extension including mid-infrared(MIR) generation.Such a process can be integrated in monolithic resonators, being compact and low in threshold. In this work, we show that the monolithic χ^((2))mini-OPO can also be used for optical frequency comb generation around 2096 nm and enters the boundary of MIR range. A new geometry called an optical superlattice box resonator is developed for this realization with near-material-limited quality factor of 4.0 × 10^(7). Only a continuous-wave near-infrared pump laser is required, with OPO threshold of 80 mW and output power up to 340 mW. Revival temporal profiles are measured at a detectable repetition frequency of 1.426 GHz, and narrow beat note linewidth of less than 10 Hz shows high comb coherence. These results are in good agreement with our simulation for a stable comb generation. 3The dynamic metabolic profile of Qi-Yu-San-Long decoction in rat urine using UPLC-QTOF-MS^(E) coupled with a post-targeted screening strategy显示文摘Qi-Yu-San-Long decoction(QYSLD)is a traditional Chinese medicine that has been clinically used in the treatment of non-small-cell lung cancer(NSCLC)for more than 20 years.However,to date,metabolicrelated studies on QYSLD have not been performed.In this study,a post-targeted screening strategy based on ultra-performance liquid chromatography coupled with quadrupole time-of-flight full information tandem mass spectrometry(UPLC-QTOF-MS^(E))was developed to identify QYSLD-related xenobiotics in rat urine.The chemical compound database of QYSLD constituents was established from previous research,and metabolites related to these compounds were predicted in combination with their possible metabolic pathways.The metabolites were identified by extracted ion chromatograms using predicted m/z values as well as retention time,excimer ions,and fragmentation behavior.Overall,85 QYSLD-related xenobiotics(20 prototype compounds and 65 metabolites)were characterized from rat urine.The main metabolic reactions and elimination features of QYSLD included oxidation,reduction,decarboxylation,hydrolysis,demethylation,glucuronidation,sulfation,methylation,deglycosylation,acetylation,and associated combination reactions.Of the identified molecules,14 prototype compounds and 58 metabolites were slowly eliminated,thus accumulating in vivo over an extended period,while five prototypes and two metabolites were present in vivo for a short duration.Furthermore,one prototype and five metabolites underwent the process of“appearing-disappearing-reappearing”in vivo.Overall,the metabolic profile and characteristics of QYSLD in rat urine were determined,which is useful in elucidating the active components of the decoction in vivo,thus providing the basis for studying its mechanism of action.Ting Zheng Yue Zhao Ruijuan Li Mengwen Huang An Zhou Zegeng Li Huan Wu 2022Journal of Pharmaceutical Analysis2022,12,5:1
4Pancreatic cancer cells render tumor-associated macrophages metabolically reprogrammed by a GARP and DNA methylation-mediated mechanism显示文摘How tumor-associated macrophages transit from a predominant antitumor M1-like phenotype to a protumoral M2-like phenotype during the development of pancreatic ductal adenocarcinoma (PDA) remains to be elucidated. We thus conducted a study by employing a PDA-macrophage co-culture system, an “orthotopic” PDA syngeneic mouse model, and human PDA specimens, together with macrophages derived from GARP knockout mice and multiple analytic tools including whole-genome RNA sequencing, DNA methylation arrays, multiplex immunohistochemistry, metabolism measurement, and invasion/metastasis assessment. Our study showed that PDA tumor cells, through direct cell–cell contact, induce DNA methylation and downregulation of a panel of glucose metabolism and OXPHOS genes selectively in M1-like macrophages, leading to a suppressed glucose metabolic status in M1-like but not in M2-like macrophages. Following the interaction with PDA tumor cells, M1-like macrophages are reprogrammed phenotypically to M2-like macrophages. The interaction between M1-like macrophages and PDA cells is mediated by GARP and integrin αV/β8, respectively. Blocking either GARP or integrin would suppress tumor-induced DNA methylation in Nqo-1 gene and the reprogramming of M1-like macrophages. Glucose-response genes such as Il-10 are subsequently activated in tumor-educated M1-like macrophages. Partly through Il-10 and its receptor Il-10R on tumor cells, M1-like macrophages functionally acquire a pro-cancerous capability. Both exogenous M1-like and M2-like macrophages promote metastasis in a mouse model of PDA while such a role of M1-like macrophages is dependent on DNA methylation. 5黑蒜提取液对恶性黑色素瘤B16细胞中Akt和PTEN表达的影响及意义(英文)显示文摘Objective: The aim of this study was to investigate the effect on Akt and PTEN expression and the mechanism of proliferation and apoptosis of melanoma B16 cells treated with chamaejasme extract. Methods: The expressions of Akt and PTEN of B16 cells treated with different concentrations of chamaejasme extract were detected with immunohistochemical method, cell apoptosis index(AI) was calculated with in situ labeling method. Results: Akt expressions of B16 cells treated with different concentrations of chamaejasme extract were significantly lower than the control group, while the PTEN expressions significantly up-regulated, and the effects appeared to be dose-related(P < 0.05). The Akt/AI of B16 cells treated with different concentrations of chamaejasme extract was significantly lower than the control group, all the parameters had extremely difference(F = 24.58, P < 0.05). 6RhoGEF Trio Regulates Radial Migration of Projection Neurons via Its Distinct Domains显示文摘The radial migration of cortical pyramidal neurons(PNs)during corticogenesis is necessary for establishing a multilayered cerebral cortex.Neuronal migration defects are considered a critical etiology of neurodevelopmental disorders,including autism spectrum disorders(ASDs),schizophrenia,epilepsy,and intellectual disability(ID).TRIO is a high-risk candidate gene for ASDs and ID.However,its role in embryonic radial migration and the etiology of ASDs and ID are not fully understood.In this study,we found that the in vivo conditional knockout or in utero knockout of Trio in excitatory precursors in the neocortex caused aberrant polarity and halted the migration of late-born PNs.Further investigation of the underlying mechanism revealed that the interaction of the Trio N-terminal SH3 domain with Myosin X mediated the adherence of migrating neurons to radial glial fibers through regulating the membrane location of neuronal cadherin(N-cadherin).Also,independent or synergistic overexpression of RAC1 and RHOA showed different phenotypic recoveries of the abnormal neuronal migration by affecting the morphological transition and/or the glial fiber-dependent locomotion.Taken together,our findings clarify a novel mechanism of Trio in regulating N-cadherin cell surface expression via the interaction of Myosin X with its N-terminal SH3 domain.These results suggest the vital roles of the guanine nucleotide exchange factor 1(GEF1)and GEF2 domains in regulating radial migration by activating their Rho GTPase effectors in both distinct and cooperative manners,which might be associated with the abnormal phenotypes in neurodevelopmental disorders.Chengwen Wei Mengwen Sun Xiaoxuan Sun Hu Meng Qiongwei Li Kai Gao Weihua Yue Lifang Wang Dai Zhang Jun Li 2022Neuroscience Bulletin2022,38,3:1
7Biogenesis of flavor-related linalool is diverged and genetically conserved in tree peony (Paeonia × suffruticosa)显示文摘Floral scent is an important and genetically complex trait in horticultural plants.Tree peony(Paeonia×suffruticosa)originates in the Pan-Himalaya and has nine wild species divided into two subsections,Delavayanae and Vaginatae.Their f lowers are beloved worldwide for their sweet f loral fragrance,yet the f lavor-related volatiles and underlying biosynthetic pathways remain unknown.Here,we characterized the volatile blends of all wild tree peony species and found that the flavor-related volatiles were highly divergent,but linalool was a unique monoterpene in subsect.Delavayanae.Further detection of volatiles in 97 cultivars with various genetic backgrounds showed that linalool was also the characteristic aroma component in Paeonia delavayi hybrid progenies,suggesting that linalool was conserved and dominant within subsect.Delavayanae and its hybrids,instead of species and cultivars from subsect.Vaginatae.Global transcriptome analysis of all wild tree peony species and 60 cultivars revealed five candidate genes that may be involved in key steps of linalool biosynthesis;especially the expressions of three TPS genes,PdTPS1,PdTPS2,and PdTPS4,were significantly positively correlated with linalool emissions across tree peony cultivars.Further biochemical evidence demonstrated that PdTPS1 and PdTPS4were the pivotal genes determining the species-specific and cultivar-specific emission of linalool.This study revealed a new insight into floral scent divergence in tree peony and would greatly facilitate our understanding of the phylogeny and evolution of Paeonia.Shanshan Li Ling Zhang Miao Sun Mengwen Lv Yong Yang Wenzhong Xu Liangsheng Wang 2023Horticulture Research2023,10,2:0
8Causal relationship between hypertension and epilepsy:a mendelian randomization study显示文摘Background Recent studies suggest that hypertension may increase the risk of epilepsy onset,revealing intricate interactions between cardiovascular health and neurological disorders,thus emphasizing the significance of conducting further investigations into their connection.This study aimed to investigate the potential causality between hypertension,either in systolic or diastolic blood pressure,and epilepsy,using a Mendelian randomization strategy.Methods A two-sample Mendelian randomization design was used in this study.We extracted data from the UK Biobank,FinnGen,and the International Consortium of Blood Pressure,utilizing blood pressure-related single nucleotide polymorphisms as instrumental variables to evaluate the influence of hypertension on the risk of epilepsy.Inverse variance weighted,weighted median,and MR-Egger approaches were used for analysis.Results There was a potential association between hypertension,primarily in systolic blood pressure,and an elevated epilepsy risk,while the relationship between hypertension in diastolic blood pressure and epilepsy risk remained inconclusive.Sensitivity analyses suggest an absence of substantial heterogeneity and confounding effects,suggesting the reliability of our findings.Conclusions Our study lays the groundwork for further investigations into the mechanisms of this causal relationship,which may potentially involve vascular change,neuroinflammatory pathways,and alterations in cerebral blood flow,which are crucial for understanding the complex hypertension-epilepsy nexus.Zhen Sun Tong Jiang Mengwen Zhang Yulong Li Jing Zhang Yanping Sun Xiaofeng Yu 2024Acta Epileptologica2024,7,1:0
