| 1 | Treated dentin matrix induces odontogenic differentiation of dental pulp stem cells via regulation of Wnt/β-catenin signaling显示文摘Treated dentin matrix(TDM)is an ideal scaffold material containing multiple extracellular matrix factors.The canonical Wnt signaling pathway is necessary for tooth regeneration.Thus,this study investigated whether the TDM can promote the odontogenic differentiation of human dental pulp stem cells(hDPSCs)and determined the potential role of Wnt/β-catenin signaling in this process.Different concentrations of TDM promoted the dental differentiation of the hDPSCs and meanwhile,the expression of GSK3βwas decreased.Of note,the expression of the Wnt/β-catenin pathway-related genes changed significantly in the context of TDM induction,as per RNA sequencing(RNA seq)data.In addition,the experiment showed that new dentin was visible in rat mandible cultured with TDM,and the thickness was significantly thicker than that of the control group.In addition,immunohistochemical staining showed lower GSK3βexpression in new dentin.Consistently,the GSK3βknockdown hDPSCs performed enhanced odotogenesis compared with the control groups.However,GSK3βoverexpressing could decrease odotogenesis of TDM-induced hDPSCs.These results were confirmed in immunodeficient mice and Wistar rats.These suggest that TDM promotes odontogenic differentiation of hDPSCs by directly targeting GSK3βand activating the canonical Wnt/β-catenin signaling pathway and provide a theoretical basis for tooth regeneration engineering. | Sirui Liu Jingjing Sun Shuai Yuan Yanyu Yang Yuping Gong Ying Wang Runying Guo Xue Zhang Yiming Liu Hongyan Mi Meiyue Wang Mengzhe Liu Rui Li | 2022 | Bioactive Materials2022,7,1: | 2 |
| 2 | Characterization of Bioaerosol Bacterial Communities During Hazy and Foggy Weather in Qingdao, China显示文摘This study was conducted to evaluate the impact of hazy and foggy weather on the bacterial communities in bioaerosols, for which samples were collected from the Qingdao coastal region on sunny, foggy, and hazy days in January and March 2013. Bacterial community compositions were determined using polymerase chain reaction denaturing gradient gel electrophoresis(PCR-DGGE). The bacterial community diversity was found to be high on foggy and hazy days, and the dominant species differed during hazy weather. The Shannon-Wiener index revealed that the bacterial community diversity of coarse particles was higher than that of fine particles in the bioaerosols. The bacterial community diversity of fine particles significantly correlated with relative humidity(RH; r^2 = 0.986). The cluster analysis results indicated that the bacterial communities on sunny days differed from those on hazy and foggy days. Compared with sunny days, the bacterial communities in the fine particles during hazy weather exhibited greater changes than those in the coarse particles. Most of the sequenced bacteria were found to be closely affiliated with uncultured bacteria. During hazy weather, members of the classes Bacilli and Gammaproteobacteria(Pseudomonas and Acinetobacter) were dominant. The DGGE analysis revealed that Proteobacteria and Firmicutes were the predominant phyla, and their relative percentages to all the measured species changed significantly on hazy days, particularly in the fine particles. | 7 | Biological Effects of Nutrient Nualgi on the Water Ecological Environment in Culture Pond显示文摘In order to study the short-term effects of Nualgi on water quality and phytoplankton in fishpond,we conducted continuous and regular surveys on the phytoplankton community and the water quality indexes in four groups. The four groups were a Nualgi group( M_1 ),a diatom group(M2),a Nualgi and diatom group( M_3 ),and a control group(C). The results showed that there were no significance differences in p H,dissolved oxygen(DO),total nitrogen(TN) and total phosphorus( TP) among the four groups. The IP concentration of groups M_1 and M_3 was significantly lower than that of groups M_2 and C,indicating that Nualgi can reduce the IP concentration of water. The concentrations of NH^+4-N,NO_2^--N and COD_(Mn) of groups M_1 ,M_2 and M_3 were significantly lower than those of group C,suggesting that both Nualgi and dominant diatom species can adjust water quality by reducing the concentrations of NH^+4-N,NO_2^--N and COD_(Mn). In this test,we identified 186 species that belonged to 109 genera of seven phyla. Among them,86 species were Chlorophyta,48 were Bacillariophyta,27 were Cyanophyta,17 were Euglenophyta,three were Dinophyta,three were Cryptophyta and two were Chrysophyta. Both diatom species and Nualgi can promote the growth of Bacillariophyta,restrain the growth of Chlorophyta and Cyanophyta,and thus change phytoplankton community structure. The biomass of Bacillariophyta in groups M_1 ,M_2 and M_3 was larger than that in group C from the 20 ^(th) d of the experiment,while the biomasses of Chlorophyta and Cyanophyta were lower than that in group C. Melosira granulata became the dominant algae species in groups M_1 ,M_2 and M_3 ,while Coelastrum microporum and Crucigenia tetrapedia were the dominant algae species in group Conly. Scenedesmus quadricauda became the dominant algae in groups M_1 ,M_2 and C. There was no remarkable difference in diversity index among the four groups. | 9 | Spatial Inhomogeneity of Atmospheric CO_(2) Concentration and Its Uncertainty in CMIP6 Earth System Models显示文摘This paper provides a systematic evaluation of the ability of 12 Earth System Models(ESMs)participating in the Coupled Model Intercomparison Project Phase 6(CMIP6)to simulate the spatial inhomogeneity of the atmospheric carbon dioxide(CO_(2))concentration.The multi-model ensemble mean(MME)can reasonably simulate the increasing trend of CO_(2) concentration from 1850 to 2014,compared with the observation data from the Scripps CO_(2) Program and CMIP6 prescribed data,and improves upon the CMIP5 MME CO_(2) concentration(which is overestimated after 1950).The growth rate of CO_(2) concentration in the northern hemisphere(NH)is higher than that in the southern hemisphere(SH),with the highest growth rate in the mid-latitudes of the NH.The MME can also reasonably simulate the seasonal amplitude of CO_(2) concentration,which is larger in the NH than in the SH and grows in amplitude after the 1950s(especially in the NH).Although the results of the MME are reasonable,there is a large spread among ESMs,and the difference between the ESMs increases with time.The MME results show that regions with relatively large CO_(2) concentrations(such as northern Russia,eastern China,Southeast Asia,the eastern United States,northern South America,and southern Africa)have greater seasonal variability and also exhibit a larger inter-model spread.Compared with CMIP5,the CMIP6 MME simulates an average spatial distribution of CO_(2) concentration that is much closer to the site observations,but the CMIP6-inter-model spread is larger.The inter-model differences of the annual means and seasonal cycles of atmospheric CO_(2) concentration are both attributed to the differences in natural sources and sinks of CO_(2) between the simulations. | Chengjun XIE Tongwen WU Jie ZHANG Kalli FURTADO Yumeng ZHOU Yanwu ZHANG Fanghua WU Weihua JIE He ZHAO Mengzhe ZHENG | 2023 | Advances in Atmospheric Sciences2023,40,11: | 0 |
| 10 | Sanguinarine suppresses cell proliferation, migration and invasion in nasopharyngeal carcinoma via inhibiting mTOR signaling显示文摘OBJECTIVE: To confirm the anti-NPC effect of sanguinarine(SA) through a series of wet experiments.METHODS: NPC cell viability was determined by proliferation experiment. Cell clone formation experiment, cell scratch test, transwell migration and invasion experiment and flow cytometry-based cell apoptosis assay were further performed. In addition, Western blotting was performed to investigate the cell signaling pathway. All the relevant experimental data were statistically processed using SPSS 16.0 software.RESULTS: The results showed that sanguinarine represented a time and dose dependent inhibition effects on NPC cell proliferation including the low differentiated CNE2 cells and high metastatic 5-8F cells, along with the cell cloning ability reduction. In addition, sanguinarine has a certain inhibitory effect on the invasion and migration of NPC cells. Mechanistically, sanguinarine displayed the anti-NPC effects mainly involved into the suppression of m TOR signaling and cell apoptosis, which is closely associated with the tumor growth and metastatic malignancy. | 11 | An Adaptive DDoS Detection and Classification Method in Blockchain Using an Integrated Multi-Models显示文摘With the rising adoption of blockchain technology due to its decentralized,secure,and transparent features,ensuring its resilience against network threats,especially Distributed Denial of Service(DDoS)attacks,is crucial.This research addresses the vulnerability of blockchain systems to DDoS assaults,which undermine their core decentralized characteristics,posing threats to their security and reliability.We have devised a novel adaptive integration technique for the detection and identification of varied DDoS attacks.To ensure the robustness and validity of our approach,a dataset amalgamating multiple DDoS attacks was derived from the CIC-DDoS2019 dataset.Using this,our methodology was applied to detect DDoS threats and further classify them into seven unique attack subcategories.To cope with the broad spectrum of DDoS attack variations,a holistic framework has been pro-posed that seamlessly integrates five machine learning models:Gate Recurrent Unit(GRU),Convolutional Neural Networks(CNN),Long-Short Term Memory(LSTM),Deep Neural Networks(DNN),and Support Vector Machine(SVM).The innovative aspect of our framework is the introduction of a dynamic weight adjustment mechanism,enhancing the system’s adaptability.Experimental results substantiate the superiority of our ensemble method in comparison to singular models across various evaluation metrics.The framework displayed remarkable accuracy,with rates reaching 99.71%for detection and 87.62%for classification tasks.By developing a comprehensive and adaptive methodology,this study paves the way for strengthening the defense mechanisms of blockchain systems against DDoS attacks.The ensemble approach,combined with the dynamic weight adjustment,offers promise in ensuring blockchain’s enduring security and trustworthiness. | Xiulai Li Jieren Cheng Chengchun Ruan Bin Zhang Xiangyan Tang Mengzhe Sun | 2023 | Computers, Materials & Continua2023,77,12: | 0 |

| 14 | Characteristics and Influencing Factors of the Microbial Concentration and Activity in Atmospheric Aerosols over the South China Sea显示文摘Oceans are important sources of microbes in atmospheric aerosols;however, information about the characteristics of airborne microbes and their influencing factors over oceans is lacking. Here we report the characteristics of the microbial abundance and activity in aerosols sampled near the sea surface over the South China Sea(SCS) from May to June 2016. The airborne microbial concentration range in the aerosols was 1.68?105 to 4.84?105 cells m-3 over the SCS, reflecting an average decrease of 40% – 54% over the SCS compared with that in the samples from the coastal region of Qingdao. About 63% – 76% of the airborne microbes occurred in coarse particles(> 2.1 ?m), with a variable size distribution over the SCS. The microbial activity range in aerosols, measured by the fluorescein diacetate(FDA) hydrolysis method, was 2.09 – 11.97 ng m-3 h-1 sodium fluorescein(SF) over the SCS, which was 15% – 79% lower than that over the coastal region. These values reflected a different spatial distribution over the SCS from that of the microbial concentration. Except for certain samples, all samples had 68% of the microbial activity occurring in coarse particles. Correlation analysis showed that the microbial abundance and activity were positively correlated with the aerosol, organic carbon(OC), and water-soluble organic carbon(WSOC) concentrations, indicating that the airborne microbes may be related to the reactions of certain water-soluble organic chemicals in the atmosphere. Moreover, the concentrations of airborne microbes were significantly negatively correlated with the horizontal offshore distance.
