| 2 | Application of an antibody chip for screening differentially expressed proteins during peach ripening and identification of a metabolon in the SAM cycle to generate a peach ethylene biosynthesis model显示文摘Peach(Prunus persica)is a typical climacteric fruit that produces ethylene rapidly during ripening,and its fruit softens quickly.Stony hard peach cultivars,however,do not produce large amounts of ethylene,and the fruit remains firm until fully ripe,thus differing from melting flesh peach cultivars.To identify the key proteins involved in peach fruit ripening,an antibody-based proteomic analysis was conducted.A mega-monoclonal antibody(mAb)library was generated and arrayed on a chip(mAbArray)at a high density,covering~4950 different proteins of peach.Through the screening of peach fruit proteins with the mAbArray chip,differentially expressed proteins recognized by 1587 mAbs were identified,and 33 corresponding antigens were ultimately identified by immunoprecipitation and mass spectrometry.These proteins included not only important enzymes involved in ethylene biosynthesis,such as ACO1,SAHH,SAMS,and MetE,but also novel factors such as NUDT2.Furthermore,protein–protein interaction analysis identified a metabolon containing SAHH and MetE.By combining the antibody-based proteomic data with the transcriptomic and metabolic data,a mathematical model of ethylene biosynthesis in peach was constructed.Simulation results showed that MetE is an important regulator during peach ripening,partially through interaction with SAHH. | Wenfang Zeng Liang Niu Zhaohui Wang Xiaobei Wang Yan Wang Lei Pan Zhenhua Lu Guochao Cui Weining Weng Mingqiao Wang Xun Meng Zhiqiang Wang | 2020 | Horticulture Research2020,7,1: | 2 |
| 3 | Prediction of a common neutralizing epitope of H5N1 avian influenza virus by in silico molecular docking显示文摘The H5N1 avian influenza virus (AIV) has widely spread in Asia, Europe and Africa, making a large amount of economic loss. Recently, our research group has screened a common neutralizing mono- clonal antibody named 8H5, which can neutralize almost all H5 subtype AIV ever isolated so far. Obvi- ously, this monoclonal antibody would benefit for research and development of the universal AIV vac- cine and design of the drug against H5N1 AIV in high mutation rate. In this study, the homology mod- eling was applied to generate the 3D structure of 8H5 Fab fragment, and 'canonical structure' method was used to define the specified loop conformation of CDR regions. The model was subjected to en- ergy minimization in cvff force field with Discovery module in Insight II program. The resulting model has correct stereochemistry as gauged from the Ramachandran plot calculation and good 3D-structure compatibility as assessed by interaction energy analysis, solvent accessible surface (SAS) analysis, and Profiles-3D approach. Furthermore, the 8H5 Fab model was subjected to docking with three H5 subtype hemagglutinin (HA) structures deposited in PDB (ID No: 1jsm, 2ibx and 2fk0) respectively. The result indicates that the three docked complexes share a common binding interface, but differ in bind- ing angle related with HA structure similarity between viral subtypes. In the light of the three HA inter- faces with structural homology analysis, the common neutralizing epitope on HA recognized by 8H5 consists of 9 incontinuous amino acid residues: Asp68, Asn72, Glu112, Lys113, Ile114, Pro118, Ser120, Tyr137, Tyr252 (numbered as for 1jsm sequence). | 4 | Delivery of pOXR1 through an injectable liposomal nanoparticle enhances spinal cord injury regeneration by alleviating oxidative stress显示文摘Oxidation resistance 1(OXR1)is regarded as a critical regulator of cellular homeostasis in response to oxidative stress.However,the role of OXR1 in the neuronal response to spinal cord injury(SCI)remains undefined.On the other hand,gene therapy for SCI has shown limited success to date due in part to the poor utility of conventional gene vectors.In this study,we evaluated the function of OXR1 in SCI and developed an available carrier for delivering the OXR1 plasmid(pOXR1).We found that OXR1 expression is remarkably increased after SCI and that this regulation is protective after SCI.Meanwhile,we assembled cationic nanoparticles with vitamin E succinate-graftedε-polylysine(VES-g-PLL)(Nps).The pOXR1 was precompressed with Nps and then encapsulated into cationic liposomes.The particle size of pOXR1 was compressed to 58 nm,which suggests that pOXR1 can be encapsulated inside liposomes with high encapsulation efficiency and stability to enhance the transfection efficiency.The agarose gel results indicated that Nps-pOXR1-Lip eliminated the degradation of DNA by DNase I and maintained its activity,and the cytotoxicity results indicated that pOXR1 was successfully transported into cells and exhibited lower cytotoxicity.Finally,Nps-pOXR1-Lip promoted functional recovery by alleviating neuronal apoptosis,attenuating oxidative stress and inhibiting inflammation.Therefore,our study provides considerable evidence that OXR1 is a beneficial factor in resistance to SCI and that Nps-Lip-pOXR1 exerts therapeutic effects in acute traumatic SCI. | Jing Zhang Yao Li Jun Xiong Helin Xu Guanghen Xiang Mingqiao Fan Kailiang Zhou Yutian Lin Xiangxiang Chen Lin Xie Hongyu Zhang Jian Wang Jian Xiao | 2021 | Bioactive Materials2021,6,10: | 2 |

| 6 | Luminescence properties of composite material Sr_(2)MgSi_(2)O_(7):Eu^(2+),Dy^(3+)/light conversion agent with multilayer structure显示文摘In order to improve the red luminescent properties,Sr_(2)MgSi_(2)O_(7):Eu^(2+),Dy^(3+)was selected as a blue persistent luminescent donor phosphor,while light conversion agent was utilized to tune the persistent luminescent spectra from blue to red.Composite red luminescent material Sr_(2)MgSi_(2)O_(7):Eu^(2+),Dy^(3+)/light conversion agent(SMED/LCA)was fabricated with light conversion agent and Sr_(2)MgSi_(2)O_(7):Eu^(2+),Dy^(3+)at a certain mass ratio.SiO_(2)(Al2 O_(3) or MgF2)were coated on the surface of SMED/LCA through heterogeneous deposition method.The structural and optical characteristics of the resulting samples were launched in terms of X-ray diffraction and emission spectrum as well as afterglow brightness.The results demonstrate that the emission spectrum exhibits two emission bands,and the peaks are located at around 470 and 615 nm.SiO_(2),Al_(2)O_(3) and MgF_(2) are coated on the surface of SMED/LCA like a protective shell to maintain its stability and luminescent properties,the afterglow initial brightness is still up to 0.37 cd/m^(2) and the afterglow color purity calculated from CIE color coordinates is basically unchanged. | Liubin Zheng Yanan Zhu Zengyuan Pang Mingqiao Ge | 2022 | Journal of Rare Earths2022,40,1: | 1 |
