1Selection and identification of a novel ssDNA aptamer targeting human skeletal muscle显示文摘Skeletal muscle disorders have posed great threats to health.Selective delivery of drugs and oligonucleotides to skeletal muscle is challenging.Aptamers can improve targeting efficacy.In this study,for the first time,the human skeletal muscle-specific ssDNA aptamers(HSM01,etc.)were selected and identified with Systematic Evolution of Ligands by Exponential Enrichment(SELEX).The HSM01 ssDNA aptamer preferentially interacted with human skeletal muscle cells in vitro.The in vivo study using tree shrews showed that the HSM01 ssDNA aptamer specifically targeted human skeletal muscle cells.Furthermore,the ability of HSM01 ssDNA aptamer to target skeletal muscle cells was not affected by the formation of a disulfide bond with nanoliposomes in vitro or in vivo,suggesting a potential new approach for targeted drug delivery to skeletal muscles via liposomes.Therefore,this newly identified ssDNA aptamer and nanoliposome modification could be used for the treatment of human skeletal muscle diseases.Shuming Sun Han Liu Yan Hu Yanpeng Wang Mingri Zhao Yijun Yuan Yafei Han Yingying Jing Jin Cui Xiaoxiang Ren Xiao Chen Jiacan Su 2023Bioactive Materials2023,,2:3
2Sorl1 knockout inhibits expression of brain-derived neurotrophic factor:involvement in the development of late-onset Alzheimer's disease显示文摘Sortilin-related receptor 1(SORL1)is a critical gene associated with late-onset Alzheimer’s disease.SORL1 contributes to the development and progression of this neurodegenerative condition by affecting the transport and metabolism of intracellularβ-amyloid precursor protein.To better understand the underlying mechanisms of SORL1 in the pathogenesis of late-onset Alzheimer s disease,in this study,we established a mouse model of SorI1 gene knockout using cluste red regularly inters paced short palindro mic repeats-associated protein 9 technology.We found that Sorl1-knocko ut mice displayed deficits in learning and memory.Furthermore,the expression of brain-derived neurotrophic factor was significantly downregulated in the hippocampus and co rtex,and amyloidβ-protein deposits were observed in the brains of 5orl1-knockout mice.In vitro,hippocampal neuronal cell synapses from homozygous Sorl1-knockout mice were impaired.The expression of synaptic proteins,including Drebrin and NR2B,was significantly reduced,and also their colocalization.Additionally,by knocking out the Sorl1 gene in N2a cells,we found that expression of the N-methyl-D-aspartate receptor,NR2B,and cyclic adenosine monophosphate-response element binding protein was also inhibited.These findings suggest that SORL1 participates in the pathogenesis of late-onset Alzheimer s disease by regulating the N-methyl-D-aspartate receptor NR2B/cyclic adenosine monophosphate-response element binding protein signaling axis.Mingri Zhao Xun Chen Jiangfeng Liu Yanjin Feng Chen Wang Ting Xu Wanxi Liu Xionghao Liu Mujun Liu Deren Hou 2024Neural Regeneration Research2024,19,7:2
3Mechanisms of oil displacement by ASP-foam and its influencing factors显示文摘ASP-foam(ASPF) is a system prepared by injecting natural gas into the conventional alkalisurfactant-polymer(ASP) system. Foam can be formed in the porous media by the interaction of gas and surfactant in the ASP system. With the ASPF system,oil recovery is improved as the interfacial tension(IFT) is reduced to a relatively low level,and the swept volume is enlarged. In this paper,four surfactants were evaluated and characterized by IFT between ASP system and oil and the foaming performance. Alkyl benzene sulfonate(ORS-41) was chosen as the surfactant to best reduce IFT between displacement fluids and oil and improve the foaming performance. The mechanisms of ASPF flooding were studied in this paper,the results show that the ASPF flooding not only enlarges the swept volume but also enhances the displacement effi ciency. The effects of reservoir heterogeneity,the gas-liquid ratio of ASPF system,and the concentrations of polymer and surfactant on the displacement effi ciency were studied. A fi eld trial of ASPF flooding has also been conducted. Both the laboratory results and the fi eld trial results show that the ASPF flooding can signifi cantly increase the oil recovery,with a 30% increase in the proportion of the original oil in place recovered compared with water flooding.Wu Wenxiang Pan Jianhua Guo Mingri 2010Petroleum Science2010,7,1:1
