1Simultaneous gene editing of three homoeoalleles in self-incompatible allohexaploid grasses显示文摘Clustered regularly interspaced palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)system has been widely used for precise gene editing in plants.However,simultaneous gene editing of multiple homoeoalleles remains challenging,especially in self-incompatible polyploid plants.Here,we simultaneously introduced targeted mutations in all three homoeoalleles of two genes in the self-incompatible allohexaploid tall fescue,using both CRISPR/Cas9 and LbCas12a(LbCpf1)systems.Loss-of-function mutants of FaPDS exhibited albino leaves,while knockout of FaHSP17.9 resulted in impaired heat resistance in T0 generation of tall fescue.Moreover,these mutations were inheritable.Our findings demonstrate the feasibility of generating loss-of-function mutants in T0 generation polyploid perennial grasses using CRISPR/Cas systems.Liang Zhang Tao Wang Guangyang Wang Aoyue Bi Misganaw Wassie Yan Xie Liwen Cao Huawei Xu Jinmin Fu Liang Chen Yang Zhao Tao Hu 2021Journal of Integrative Plant Biology2021,63,8:3

9Floristic composition and structure of the Kibate Forest along environmental gradients in Wonchi,Southwestern Ethiopia显示文摘Dry evergreen montane forests in Ethiopia are severely threatened.The status of species composition and structure of forest vegetation are important indicators to understand the trends of threats on local plant communities.In the present study,we examined the floristic composition and structure of the Kibate Forest,Wonchi Highland,Ethiopia along environmental gradients.Sixty-six(30 m×30 m)plots were established every 100 m interval along altitudinal gradients(2811‒3073 m a.s.l.)in five transect lines for vegetation and environmental data collection.In total,125 vascular plant species belonging to 104 genera and 52 families were identified.Eighteen species(14%)were endemic to Ethiopia and Eritrea.The two most dominant families,Asteraceae(29 species)and Lamiaceae(eight species)accounted for 30%of the total number of species.The highest number of species(54%)was herbs.Four major community types(viz.,Olinia rochetiana-Myrsine melanophloeos,Ilex mitis-Galiniera saxifraga,Erica arborea-Protea gaguedi,and Hagenia abyssinica-Juniperus procera)were identified.The highest species richness,evenness,diversity,and importance value index were in community types 2 and 4.About 82%of the species and all endemic taxa except five were recorded in these two community types.The most dominant woody species were O.rochetiana,E.arborea,Olea europaea subsp.cuspidata,Myrica salicifolia,I.mitis var.mitis,and H.abyssinica with different patterns of population structure.The results show that there was a weak correlation between species richness and altitude.Our findings confirm that environmental variables both with interactions(such as altitude)and without interactions(such as livestock grazing)significantly(p<0.05)affect species richness.Anthropogenic activities and overgrazing by livestock appear to be the main threat in community types 2 and 3.Urgent management practices and conservation measures such as prohibiting forest clearing and overgrazing and planting indigenous trees through community participation should be considered in community types that are rich in endemic species but are highly threatened.Misganaw Meragiaw Zerihun Woldu Vegard Martinsen Bal R.Singh 2021Journal of Forestry Research2021,32,6:0
10Genome-wide identification of the CONSTANS-LIKE(COL)family and mechanism of fruit senescence regulation by PpCOL8 in sand pear(Pyrus pyrifolia)显示文摘Pyrus pyrifolia Nakai‘Whangkeumbae'is a sand pear fruit with excellent nutritional quality and taste.However,the industrial development of pear fruit is significantly limited by its short shelf life.Salicylic acid(SA),a well-known phytohormone,can delay fruit senescence and improve shelf life.However,the mechanism by which SA regulates CONSTANS-LIKE genes(COLs)during fruit senescence and the role of COL genes in mediating fruit senescence in sand pear are poorly understood.In this study,22 COL genes were identified in sand pear,including four COLs(Pp COL8,Pp COL9a,Pp COL9b,and Pp COL14)identified via transcriptome analysis and 18 COLs through genome-wide analysis.These COL genes were divided into three subgroups according to the structural domains of the COL protein.Pp COL8,with two B-box motifs and one CCT domain,belonged to the first subgroup.In contrast,the other three Pp COLs,Pp COL9a,Pp COL9b,and Pp COL14,with similar conserved protein domains and gene structures,were assigned to the third subgroup.The four COLs showed different expression patterns in pear tissues and were preferentially expressed at the early stage of fruit development.Moreover,the expression of Pp COL8 was inhibited by exogenous SA treatment,while SA up-regulated the expression of Pp COL9a and Pp COL9b.Interestingly,Pp COL8 interacts with Pp MADS,a MADS-box protein preferentially expressed in fruit,and SA up-regulated its expression.While the production of ethylene and the content of malondialdehyde(MDA)were increased in Pp COL8-overexpression sand pear fruit,the antioxidant enzyme(POD and SOD)activity and the expression of Pp POD1 and Pp SOD1 in the sand pear fruits were down-regulated,which showed that Pp COL8 promoted sand pear fruit senescence.In contrast,the corresponding changes were the opposite in Pp MADS-overexpression sand pear fruits,suggesting that Pp MADS delayed sand pear fruit senescence.The co-transformation of Pp COL8 and Pp MADS also delayed sand pear fruit senescence.The results of this study revealed that Pp COL8 can play a key role in pear fruit senescence by interacting with Pp MADS through the SA signaling pathway.Yue Xu Shurui Song Huiying Wang Xilong Cao Xinran Zhao Wenli Wang Liyue Huo Yawei Li Misganaw Wassie Bin Lu Liang Chen Haiyan Shi 2024Journal of Integrative Agriculture2024,23,4:0
11A synthetic light-inducible photorespiratory bypass enhances photosynthesis to improve rice growth and grain yield显示文摘Bioengineering of photorespiratory bypasses is an effective strategy for improving plant productivity by modulating photosynthesis.In previouswork,two photorespiratory bypasses,theGOC andGCGT bypasses,increased photosynthetic rates but decreased seed-setting rate in rice(Oryza sativa),probably owing to excess photosynthate accumulation in the stem.To solve this bottleneck,we successfully developed a newsynthetic photorespiratory bypass(called theGMAbypass)in rice chloroplasts by introducing Oryza sativa glycolate oxidase 1(OsGLO1),Cucurbita maxima malate synthase(CmMS),and Oryza sativa ascorbate peroxidase7(OsAPX7)into the rice genome using a high-efficiency transgene stacking system.Unlike the GOC and GCGT bypass genes driven by constitutive promoters,OsGLO1 in GMA plants was driven by a light-inducible Rubisco small subunit promoter(pRbcS);its expression dynamically changed in response to light,producing a more moderate increase in photosynthate.Photosynthetic rates were significantly increased inGMA plants,and grain yieldswere significantly improved under greenhouse and field conditions.Transgenic GMA rice showed no reduction in seed-setting rate under either test condition,unlike previous photorespiratory-bypass rice,probably reflecting proper modulation of the photorespiratory bypass.Together,these results imply that appropriate engineering of the GMA bypass can enhance rice growth and grain yield without affecting seed-setting rate.Huawei Xu Huihui Wang Yanwen Zhang Xiaoyi Yang Shufang Lv Dianyun Hou Changru Mo Misganaw Wassie Bo Yu Tao Hu 2023Plant Communications2023,4,6:0
12EROSION AND SEDIMENT MANAGEMENT STRATEGIESFOR THE ILLINOIS RIVER显示文摘The Illinois River is the single most important river in the state of Illinois. Approximately 95 percent of the urban areas and half of the agricultural lands in the state are located within the Illinois River basin. The Illinois Waterway, which consists of the Illinois River and its tributary streams. is the only waterway that links the Great Lakes to the Mississippi River and thus the Gulf of Mexico. The Illinois Waterway is therefore an important commercial waterway. Because of its strategic location in the state and because it is downstream of the Chicago Metropolitan Area, the Illinois River has experienced significant human influences over the years. The most significant influences have been related to commercial navigation, municipal and industrial waste discharges, and agricultural practices in the watershed. Because of the lack of a basinwide comprehensive management plan. many of the environmental and ecological problems in the Illinois River basin have not been addressed adequately over the years. The most serious and difficult to manage problem facing the Illinois River is the problem of sedimentation in the river channel and backwater lakes. Many of the backwater lakes have lost over 70 percent of their capacities to sediment accumulation and might not last too long unless appropriate rehabilitation and management techniques are applied in the near future. Problems in the Illinois River basin are not, however, limited only to sedimentation. Issues such as water quality, nonpoint source pollution, flooding, degradation of aquatic habitats, and water based recreation need to be addressed.
