| 4 | Hypoxia inducible factor(HIF) in the tumor microenvironment:friend or foe?显示文摘Hypoxia acts as an important regulator of physiological and pathological processes. Hypoxia inducible factors(HIFs) are the central players involved in the cellular adaptation to hypoxia and are regulated by oxygen sensing EGLN prolyl hydroxylases.Hypoxia affects many aspects of cellular growth through both redox effects and through the stabilization of HIFs. The HIF isoforms likely have differential effects on tumor growth via alteration of metabolism, growth, and self-renewal and are likely highly context-dependent. In some tumors such as renal cell carcinoma, the EGLN/HIF axis appears to drive tumorigenesis,while in many others HIF1 and HIF2 may actually have a tumor suppressive role. An emerging role of HIF biology is its effects on the tumor microenvironment. The EGLN/HIF axis plays a key role in regulating the function of the various components of the tumor microenvironment, which include cancer-associated fibroblasts, endothelial cells, immune cells, and the extracellular matrix(ECM). | 10 | Celastrus orbiculatus extract induces mitochondrial-mediated apoptosis in human hepatocellular carcinoma cells显示文摘OBJECTIVE:To investigate the apoptotic effects and underlying molecular mechanisms of Celastrus orbiculatus(C.orbiculatus) extract in human hepatocellular carcinoma cells.METHODS:Human hepatocellular carcinoma cells(HCCLM6) were treated with C.orbiculatus extract(COE) at different nontoxic concentrations(10,20,40,80,and 160 μg/mL).The effect of COE on HC-CLM6 viability was examined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) assays.Cellular apoptosis following COE treatment was assessed by flow cytometry and western blot analysis.RESULTS:COE significantly inhibited cell viability and induced apoptosis of HCCLM6 cells in a dose-dependent manner.Apoptosis was accompanied by increased Bax expression and decreased Bcl-2 expression.In addition,COE treatment led to the release of cytochrome c,activation of caspase-3,and cleavage of poly(ADP-ribose) polymerase(PARP).Furthermore,activation of extracellular signal-regulated kinase(ERK),p38 kinase,and c-Jun N-terminal kinase(JNK) phosphorylation,and down-regulation of Akt phosphorylation was observed.CONCLUSION:COE induces mitochondrial-mediated,caspase-dependent apoptosis in HCCLM6 cells,which might be attributed to the activation of mitogen-activated protein kinase(MAPK) and inhibition of Akt signaling pathways.These data suggest that COE may be a potential treatment for human hepatocellular carcinoma. | Hua Zhang Yayun Qian Yanqing Liu Guoqing Li Pingfang Cui Yaodong Zhu Hui Ma Xue Ji Shiyu Guo Hisamits Tadashi | 2012 | Journal of Traditional Chinese Medicine2012,32,4: | 18 |

| 12 | NOD2 3020insC frameshift mutation is not associated with inflammatory bowel disease in Chinese patients of Hart nationality显示文摘AIM: An insertion mutation at nucleotide 3020 (3020insC) in the Caspase recruitment domain gene (CARD15), originally reported as NOD2, is strongly associated with Crohn's disease. The C-insertion mutation at nucleotide 3020 (3020inC) in the leucine-rich repeat (LRR) region results in a frameshift in the 10^th LRR followed by a premature stop codon. This truncation mutation is responsible for the inability to activate nuclear factor (NF)-κB in response to bacterial lipopolysaccharide (LPS). The present study aimed to genotype NOD2/CARD15 gene 3020insC frameshift mutation in Chinese patients with inflammatory bowel disease. METHODS: We genotyped an insertion polymorphism affecting the leucine-rich region of the protein product by the allele specific PCR in 74 unrelated patients with ulcerative colitis of Han nationality in Hubei Province of China, 15 patients with Crohn's disease and 172 healthy individuals. RESULTS: No significant differences were found in the genotype and allele frequendes of the C-insertion mutation of NOD2 gene among patients with Crohn's disease and ulcerative colitis and healthy controls. | 13 | Effect and mechanism of Irbesartan on occurrence of ventricular arrhythmias in rats with myocardial ischemia through connexin43(cx43)显示文摘Objective: To explore the effect and mechanism of angiotensin Ⅱ receptor blockers-Irbesartan on occurrence of ventricular arrhythmias in rats with myocardial ischemia. Methods: Rats with embryonic cardiomyocytes-H9c2 were randomly divided into control group, ischemia group, Irbesartan group and Irbesartan+ischemia group. The cell viability of rats in each group was tested using MTT. Real-time PCR was employed to detect the expression of connexin43(Cx43) mR NA and western blot to detect the expression of Cx43 and phosphorylated Cx43. SD rats were randomly divided into the sham-operation group(SO), myocardial infarction group(MI), Irbesartan group and MI+ Irbesartan group, with 10 rats in each group. HE staining was employed to observe the change in the pathomorphology of left ventricular tissue and TUNEL method to analyze the cell apoptosis in the tissue. The immunofluorescence was adopted to observe the expression and distribution of Cx43 in the left ventricular myocardium and study the change in the expression of Cx43 in the cardiac muscular tissue at mR NA and protein level. Results: The intervention of Irbesartan in the condition of ischemia indicated the significant decrease in the number of necrotic cells. The expression of Cx43 was significantly decreased under the culture of ischemia(P<0.05), but in the presence of Irbesartan, the expression of Cx43 was increased compared with the ischemia group(P<0.01). The results of WB assay showed the similar trend of change at mRNA level. There was the significant difference in the score of ventricular arerythmia between MI group and SO group(P<0.01). The incidence of ventricular tachycardia or ventricular fibrillation was significantly increased compared with the one in SO group(P<0.05). There was the significant difference in the overall score between MI+Irbesartan group and MI group(P<0.05). The expression of Cx43 in the cardiac muscular tissue in MI group was significantly decreased(P<0.01 vs SO group). But the expression of Cx43 was increased after the treatment with Irbesartan. Conclusions: Irbesartan can inhibit the injury of H9c2 cardiomyocytes and the decreased expression of Cx43 that are induced by the ischemic myocardial infarction. In compliance with data standards and structures of the International Nucleotide Sequence Database Collaboration(INSDC), GSA adopts four data objects(Bio Project, Bio Sample,Experiment, and Run) for data organization, accepts raw sequence reads produced by a variety of sequencing platforms, stores both sequence reads and metadata submitted from all over the world,and makes all these data publicly available to worldwide scientific communities. | 18 | Gray bootstrap method for estimating frequency-varying random vibration signals with small samples显示文摘During environment testing, the estimation of random vibration signals(RVS) is an important technique for the airborne platform safety and reliability. However, the available methods including extreme value envelope method(EVEM), statistical tolerances method(STM) and improved statistical tolerance method(ISTM) require large samples and typical probability distribution. Moreover, the frequency-varying characteristic of RVS is usually not taken into account. Gray bootstrap method(GBM) is proposed to solve the problem of estimating frequency-varying RVS with small samples. Firstly, the estimated indexes are obtained including the estimated interval, the estimated uncertainty, the estimated value, the estimated error and estimated reliability. In addition, GBM is applied to estimating the single flight testing of certain aircraft. At last, in order to evaluate the estimated performance, GBM is compared with bootstrap method(BM) and gray method(GM) in testing analysis. | 19 | Effect of allogeneic umbilical cord mesenchymal stem cell transplantation in a rat model of hepatic cirrhosis显示文摘OBJECTIVE: To determine the effects of human umbilical cord mesenchymal stem cell(UCMSC) transplantation, alone or in combination with tanshinone IIA(Tan IIA) on hepatic cirrhosis in rats.METHODS: A rat model of cirrhosis was established. Rats were divided into control, UCMSC, and UCSMC plus Tan IIA groups. Rats in the UCMSC group were injected via the tail vein with 0.2 m L Di I-labeled UCMSC suspension. Intraperitoneal Tan IIA injections(20 mg/kg) were started on the day ofUCMSC transplantation in the UCMSC plus Tan IIA group, and continued for 7 consecutive days thereafter. Rats were sacrificed 1 day, 3 days, 1 month,and 3 months after transplantation and the numbers of Di I-labeled UCMSCs colonizing the liver were determined. Albumin(ALB) and alanine aminotransferase(ALT) levels were measured in venous blood, and m RNA and protein expression levels of human ALB and cytokeratin(CK)-18 in liver tissues were determined by reverse transcription-polymerase chain reaction and western blotting, respectively.RESULTS: Serum ALT levels were significantly lower and serum ALB levels significantly higher in rats in the UCMSC group compared with the control group(P < 0.05). Hepatic CK-18 and ALB m RNA and protein expression levels increased after transplantation, and were significantly higher in the UCMSC plus Tan IIA group compared with the UCMSC group(P < 0.05).CONCLUSION: Human UCMSCs transplanted into rats with liver cirrhosis can grow and differentiate into hepatocyte-like cells resulting in improved liver function in vivo. | 20 | Agricultural information dissemination using ICTs:A review and analysis of information dissemination models in China显示文摘Over the last three decades,China’s agriculture sector has been transformed from the traditional to modern practice through the effective deployment of Information and Communication Technologies(ICTs).Information processing and dissemination have played a critical role in this transformation process.Many studies in relation to agriculture information services have been conducted in China,but few of them have attempted to provide a comprehensive review and analysis of different information dissemination models and their applications.This paper aims to review and identify the ICT based information dissemination models in China and to share the knowledge and experience in applying emerging ICTs in disseminating agriculture information to farmers and farm communities to improve productivity and economic,social and environmental sustainability.The paper reviews and analyzes the development stages of China’s agricultural information dissemination systems and different mechanisms for agricultural information service development and operations.Seven ICT-based information dissemination models are identified and discussed.Success cases are presented.The findings provide a useful direction for researchers and practitioners in developing future ICT based information dissemination systems.It is hoped that this paper will also help other developing countries to learn from China’s experience and best practice in their endeavor of applying emerging ICTs in agriculture information dissemination and knowledge transfer. | Yun Zhang Lei Wang Yanqing Duan | 2016 | Information Processing in Agriculture2016,3,1: | 12 |
