1Combined therapy with transcatheter arterial chemoembolization and percutaneous microwave coagulation for small hepatocellular carcinoma显示文摘AIM:To assess the efficacy of combined transcatheter arterial chemoembolization(TACE)and percutaneous microwave coagulation therapy(PMCT)for small hepatocellular carcinoma(HCC). METHODS:Thirty-five patients with a total of 41 HCC nodules(≤3 cm in diameter)were treated with TACE followed by computed tomograghy(CT)-guided percutaneous microwave coagulation therapy(PMCT) within 1-3 wk. RESULTS:By biopsies and enhanced CT scans, complete necrosis of the tumor and 3-5 mm of the surrounding non-cancerous area were observed in 34 foci.In seven foci,incomplete necrosis of the surrounding parenchyma was observed.Serum alpha- fetoprotein(AFP)levels returned to normal 10 d after treatment in 25 patients who originally had high serum AFP levels.The follow-up period was 6-31 mo,and all patients remained alive.One patient had a recurrence in the subsegments of the liver,and another patient had a recurrence near the original lesion. 5The Qitai radio telescope显示文摘This study presents a general outline of the Qitai radio telescope(QTT)project.Qitai,the site of the telescope,is a county of Xinjiang Uygur Autonomous Region of China,located in the east Tianshan Mountains at an elevation of about 1800 m.The QTT is a fully steerable,Gregorian-type telescope with a standard parabolic main reflector of 110 m diameter.The QTT has adopted an umbrella support,homology-symmetric lightweight design.The main reflector is active so that the deformation caused by gravity can be corrected.The structural design aims to ultimately allow high-sensitivity observations from 150 MHz up to115 GHz.To satisfy the requirements for early scientific goals,the QTTwill be equipped with ultra-wideband receivers and large field-of-view multi-beam receivers.A multi-function signal-processing system based on RFSo C and GPU processor chips will be developed.These will enable the QTT to operate in pulsar,spectral line,continuum and Very Long Baseline Interferometer(VLBI)observing modes.Electromagnetic compatibility(EMC)and radio frequency interference(RFI)control techniques are adopted throughout the system design.The QTT will form a world-class observational platform for the detection of lowfrequency(nano Hertz)gravitational waves through pulsar timing array(PTA)techniques,pulsar surveys,the discovery of binary black-hole systems,and exploring dark matter and the origin of life in the universe.The QTT will also play an important role in improving the Chinese and international VLBI networks,allowing high-sensitivity and high-resolution observations of the nuclei of distant galaxies and gravitational lensing systems.Deep astrometric observations will also contribute to improving the accuracy of the celestial reference frame.Potentially,the QTT will be able to support future space activities such as planetary exploration in the solar system and to contribute to the search for extraterrestrial intelligence.Na Wang Qian Xu Jun Ma Zhiyong Liu Qi Liu Hailong Zhang Xin Pei Maozheng Chen Richard N.Manchester Kejia Lee Xingwu Zheng Hans J.Kärcher Wulin Zhao Hongwei Li Dongwei Li Martin Süss Matthias Reichert Zhongyi Zhu Congsi Wang Mingshuai Li Rui Li Ning Li Guljaina Kazezkhan Wenming Yan Gang Wu Lang Cui Ming Zhang Haitao Li 2023Science China(Physics,Mechanics & Astronomy)2023,66,8:14

7Dihydroartemisinin regulates the immune system by promotion of CD8^+T lymphocytes and suppression of B cell responses显示文摘Artemisia annua is an anti-fever herbal medicine first described in traditional Chinese medicine 1,000 years ago.Artemisinin,the extract of A.annua,and its derivatives(dihydroartemisinin(DHA),artemether,and artesunate)have been used for the treatment of malaria with substantial efficacy.Recently,DHA has also been tested for the treatment of lupus erythematosus,indicating that it may function to balance the immune response in immunocompromised individuals.In the present study,the regulatory effect of artemisinin on the murine immune system was systematically investigated in mice infected with two different protozoan parasites(Toxoplasma gondii and Plasmodium berghei).Our results revealed that the mouse spleen index significantly increased(spleen enlargement)in the healthy mice after DHA administration primarily due to the generation of an extra number of lymphocytes and CD8^+T lymphocytes in both the spleen and circulation.DHA could increase the proportion of T helper cells and CD8^+T cells,as well as decrease the number of splenic and circulatory B cells.Further,DHA could reduce the production of proinflammatory cytokines.Our study revealed that apart from their anti-parasitic activity,artemisinin and its derivatives can also actively modulate the immune system to directly benefit the host.Ting Zhang Yiwei Zhang Ning Jiang Xu Zhao Xiaoyu Sang Na Yang Ying Feng Ran Chen Qijun Chen 2020Science China(Life Sciences)2020,63,5:12
8PI3K/AKT/mTOR signaling pathway inhibitors in proliferation of retinal pigment epithelial cells显示文摘AIM: To determine whether the PI3K/AKT/mTOR pathway is activated in proliferative vitreoretinopathy (PVR) in homo- sapiens. METHODS: The retina of controls and patients with PVR were collected and their levels of PI3K, phospho-AKT, phospho-mTOR, phospho-p70S6k and phospho-4EBP-1 were determined by Western blot. The cultured human retinal pigment epithelial cell line D407 was treated with a specific mTOR inhibitor, rapamycin (RAPA) or a PI3K inhibitor, LY294002, of various concentrations and durations. Cell morphology was observed by phase contrast microscopy and the proliferation and apoptosis of treated cells were determined by MTT assay and flow cytometry. RESULTS: Levels of PI3K, phospho-AKT, phospho-mTOR, phospho-P70S6K and phospho-4EBP1 was increased in the retina in PVR (P <0.05). In D407 cells, both RAPA and LY294002 significantly inhibited cell proliferation and cell cycle progression, and promoted apoptosis (P <0.05); mor- phologically, the cells became smaller. Both RAPA and LY294002 reduced levels of phospho-AKT, phospho-mTOR, phospho-p70S6k and phospho-4EBP1 expression (P <0.05). RAPA, but not LY294002, had no significant effect on PI3K expression. CONCLUSION: PI3K/AKT/mTOR signaling pathway is highly activated in the retinal pigment epithelial cells of PVR. 10MicroRNA Expression Signature in Gastric Cancer显示文摘Objective:To identify the miRNA specific signature as novel diagnostic and prognostic tools for gastric cancer. Methods:miRNAs expression profiling of 3 normal gastric tissues,24 malignant tissues,gastric cancer cell SGC7901 and normal gastric cell GES-1 were detected using microarray technology. The hierarchical clustering algorithm of the Cluster software was used to analyse the miRNAs expression of all samples. The expression levels of miR-433 and miR-9 which were significantly down-regulated in gastric cancer tissues and SGC7901 cells by microarray technology were validated by quantitative Real-time PCR(qRT-PCR). Results:Differential expressions of 26 individual miRNAs between normal samples(including 3 normal gastric tissues and GES-1 cells) and carcinomas(including 24 malignant tissues and SGC7901 cells) were discovered,19 of them showing down-regulation and 7 up-regulation in carcinoma samples. Hierarchical clustering of the cancer samples by their miRNA expression accurately separated the carcinomas from normal samples and further their histotypes of carcinomas. 11Characterization and allergic role of IL-33-induced neutrophil polarization显示文摘Neutrophils are involved in the pathogenesis of allergy.However,the contribution of the different functionally polarized neutrophils in allergy needs to be clarified.We sought to define the characteristics of interleukin(IL)-33-induced neutrophils and the involvement of this subset of polarized neutrophils in allergic pathogenesis.Freshly isolated neutrophils were treated with different cytokines and the cytokine expression levels were detected by realtime PCR.The gene expression profile of IL-33-induced neutrophils was determined by microarray assay.Adoptive transfer assay was used to investigate the function of IL-33-induced neutrophils in an ovalbumin(OVA)-induced allergic asthma model.IL-33-treated neutrophils selectively produced IL-4,IL-5,IL-9 and IL-13(referred as to N(IL-33)cells)and displayed a distinctive gene expression profile in sharp contrast to resting and lipopolysaccharide(LPS)-treated neutrophils.IL-33-induced neutrophils expressed high Levels of IL-1R2 on cell surface,whereas resting and LPS-treated neutrophils did not,indicating IL-1R2 might be used as a biomarker for N(IL-33)cells.Importantly,N(IL-33)neutrophils exist in the lungs of OVA-induced allergic asthma mice.Adoptive transfer of N(IL-33)neutrophils significantly promotes the severity of the lung pathogenesis in this model.IL-33 induces neutrophil polarization through c-Jun N-terminal kinase-and nuclear factor-κB-dependent pathways.A previously unappreciated neutrophil polarization driven by IL-33 with unique cell surface markers and cytokine/chemokineproducing gene profile was defined.The newly identified N(IL-33)subpopulation may have significant contribution to IL-33-related pathogenesis.Bo Sun Linnan Zhu Yaling Tao Hai-Xi Sun Yang Li Peng Wang Yuzhu Hou Yang Zhao Xiaodong Zhang Lianfeng Zhang Ning Na Yong Zhao 2018Cellular & Molecular Immunology2018,15,8:9
12Nano/Micro HKUST-1 Fabricated by Coordination Modulation Method at Room Temperature显示文摘A simple and fast route for the synthesis of metal-organic framework(MOF) particles was presented.Cu 3(BTC) 2(HKUST-1,BTC=1,3,5-benzenetricarboxylate),one of the most well-known MOFs,was synthesized at room temperature via coordination modulation method.By adding different modulators(monocarboxylic acids) into the reaction system,the morphologies of HKUST-1 crystals were tuned from nano spheres to micro octahedrons at room temperature without any complex equipment.X-Ray diffractions and gas sorption measurements revealed highly crystalline particles with large Brunauer-Emmett-Teller(BET) surface areas(1116―1273 m 2 /g) and total pore volumes(0.62―0.73 cm 3 /g).The significantly small particle sizes and high capacity of gas sorption are considered advantageous for envisaged application in practical industrial process.NA Li-yan HUA Rui-nian NING Gui-ling OU Xiao-xia 2012Chemical Research in Chinese Universities2012,28,4:8

16The association between the genetic polymorphisms of LMP2/LMP7 and the outcomes of HCV infection among drug users显示文摘Objective:To investigate a possible association of LMP2/LMP7 genes with chronic hepatitis C virus(HCV) infection,and to assess whether LMP2/LMP7 genes could influence the outcomes of HCV infection among drug users.Methods:Genomic DNAs of 362 anti-HCV sero-positive drug users and 225 control drug users were extracted from the peripheral blood leukocytes.The sero-positive patients were divided into those who had persistent infection and those who had spontaneously cleared the infection.Polymorphisms of LMP genes were determined by PCR combined with restriction fragment length polymorphism(RFLP).Results:The distribution of LMP2 genotypes among the control,persistent infection and spontaneous clearance groups were not different.However,the LMP7 codon 145 Gln/Lys,Lys/Lys,and Gln/Lys+Lys/Lys genotypes were found significantly more frequent in the persistent infection group than in control group(OR=1.75,95%CI=1.06~2.90;OR=3.16,95%CI=1.23-8.12;OR=1.94,95%CI=1.21-3.12,respectively).Similarly,the frequencies of the codon 145 Gln/Lys,Lys/Lys,and Gln/Lys+Lys/Lys genotypes were found significantly more frequent in the persistent infection group than in the spontaneous clearance group(OR=1.64,95%CI=1.04-2.57;OR=2.40,95%CI=1.09-5.28;OR=1.76,95%CI=1.152.69,respectively).Stratified analysis indicated that combined genotype Gln/Lys+Lys/Lys of the LMP7 gene was related to an increasing susceptibility to HCV infection(OR=1.91,95%CI=1.02-3.55;OR=2.19,95%CI=1.243.89;OR=1.91,95%CI=1.05-3.48,OR=2.86,95%CI=1.41-5.78,respectively)and the risk of persistent HCV infection(OR=1.94,95%CI=1.12-3.34;OR=2.02,95%CI=1.21-3.38;OR=1.78,95%CI=1.12-2.85,OR=2.23,95%CI=1.09-4.58,respectively)among>30-year-old,males,the injection drug user(IDU)subjects and/or the shorter duration drug users(≤5 y).Conclusion:These results suggest that polymorphism of the LMP7 gene may have an influence on the outcomes of HCV infection,and is one of the factors accounting for the genetic susceptibility to HCV infection among drug users.Qian Cui Yongxiang Zhang Jing Su Chao Shi Na Lei Keqin Ding Jun Li Rongbin Yu L u Wang Ning Wang 2010The Journal of Biomedical Research2010,24,5:6
17A novel wheat NAC transcription factor,Ta NAC_(30), negatively regulates resistance of wheat to stripe rust显示文摘NAC transcription factors are widespread in the plant kingdom and play essential roles in the transcriptional regulation of defense responses. In this study, we isolated a novel NAC transcription factor gene, Ta NAC_(30),from a c DNA library constructed from wheat(Triticum aestivum) plants inoculated with the stripe rust pathogen Puccinia striiformis f. sp. tritici(Pst). Ta NAC_(30) contains a typical NAM domain and localizes to the nucleus. Yeast one-hybrid assays revealed that Ta NAC_(30) exhibits transcriptional activity and that its C-terminus is necessary for the activation of transcription. Expression of Ta NAC_(30) increased when host plants were infected with a virulent race(CYR_(31)) of the rust fungus Pst. Silencing of Ta NAC_(30) byvirus-induced gene silencing inhibited colonization of the virulent Pst isolate CYR_(31). Moreover, detailed histological analyses showed that silencing of Ta NAC_(30) enhanced resistance to Pst by inducing a significant increase in the accumulation of H_2O_2. Finally, we overexpressed Ta NAC_(30) in fission yeast and determined that cell viability was severely reduced in Ta NAC_(30)-transformed cells grown on medium containing H_2O_2. 18Phi-based risk calculators performed better in the prediction of prostate cancer in the Chinese population显示文摘Risk prediction models including the Prostate Health Index(phi)for prostate cancer have been well established and evaluated in the Western population.The aim of this study is to build phi-based risk calculators in a prostate biopsy population and evaluate their performanee in predicting prostate cancer(PCa)and high-grade PCa(Gleason score 27)in the Chinese population.We developed risk calculators based on 635 men who underwent initial prostate biopsy.Then,we validated the performance of prostate-specific antigen(PSA),phi,and the risk calculators in an additional observational cohort of 1045 men.We observed that the phi-based risk calculators(risk calculators 2 and 4)outperformed the PSA-based risk calculator for predicting PCa and high-grade PCa in the training cohort.In the validation study,the area under the receiver operating characteristic curve(AUC)for risk calculators 2 and 4 reached 0.91 and 0.92,respectively,for predicting PCa and high-grade PCa,respectively;the AUC values were better than those for risk calculator 1(PSA-based model with an AUC of 0.81 and 0.82,respectively)(all P<0.001).Such superiority was also observed in the stratified population with PSA ranging from 2.0 ng ml^-1 to 10.0 ng ml^-1.Decision curves confirmed that a considerable proportion of unnecessary biopsies could be avoided while applying phi-based risk calculators.In this study,we showed that,compared to risk calculators without phi,phi-based risk calculators exhibited superior discrimination and calibration for PCa in the Chinese biopsy population.Applying these risk calculators also considerably reduced the number of unnecessary biopsies for PCa.Yi-Shuo Wu Xiao-Jian Fu Rong Na Ding-Wei Ye Jun Qi Xiao-Ling Lin Fang Liu Jian Gong Ning Zhang Guang-Liang Jiang Hao-Wen Jiang Qiang Ding Jianfeng Xu Ying-Hao Sun 2019Asian Journal of Andrology2019,21,6:6
