| 1 | Understanding human diseases with high-throughput quantitative measurement and analysis of molecular signatures显示文摘Microarray and deep sequencing technologies have provided unprecedented opportunities for mapping genome mutations,RNA transcripts,transcription factor binding,and histone modifications at high resolution at the genome-wide level.This has revolutionized the way in which transcriptomes,regulatory networks and epigenetic regulations have been studied and large amounts of heterogeneous data have been generated.Although efforts are being made to integrate these datasets unbiasedly and efficiently,how best to do this still remains a challenge.Here we review major impacts of high-throughput genome-wide data generation,their relevance to human diseases,and various bioinformatics approaches for data integration.Finally,we provide a case study on inflammatory diseases. | YANG Li WEI Gang TANG Kun NARDINI Christine HAN Jing-Dong J. | 2013 | Science China(Life Sciences)2013，56，3： | 2 |
| 2 | Alzheimer’s disease:a tale of two diseases?显示文摘Sporadic late-onset Alzheimer’s disease(SLOAD)and familial early-onset Alzheimer’s disease(FEOAD)associated with dominant mutations in APP,PSEN1 and PSEN2,are thought to represent a spectrum of the same disorder based on near identical behavioral and histopathological features.Hence,FEOAD transgenic mouse models have been used in past decades as a surrogate to study SLOAD pathogenic mechanisms and as the gold standard to validate drugs used in clinical trials.Unfortunately,such research has yielded little output in terms of therapeutics targeting the disease’s development and progression.In this short review,we interrogate the widely accepted view of one,dimorphic disease through the prism of the Bmi1+/–mouse model and the distinct chromatin signatures observed between SLOAD and FEOAD brains. | Eleonora Nardini Ryan Hogan Anthony Flamier Gilbert Bernier | 2021 | Neural Regeneration Research2021，16，10： | 2 |
