1Liproxstatin-1 is an effective inhibitor of oligodendrocyte ferroptosis induced by inhibition of glutathione peroxidase 4显示文摘Our previous studies showed that ferroptosis plays an important role in the acute and subacute stages of spinal cord injury.High intracellular iron levels and low glutathione levels make oligodendrocytes vulnerable to cell death after central nervous system trauma.In this study,we established an oligodendrocyte(OLN-93 cell line)model of ferroptosis induced by RSL-3,an inhibitor of glutathione peroxidase 4(GPX4).RSL-3 significantly increased intracellular concentrations of reactive oxygen species and malondialdehyde.RSL-3 also inhibited the main antiferroptosis pathway,i.e.,SLC7A11/glutathione/glutathione peroxidase 4(xCT/GSH/GPX4),and downregulated acyl-coenzyme A synthetase long chain family member 4.Furthermore,we evaluated the ability of several compounds to rescue oligodendrocytes from ferroptosis.Liproxstatin-1 was more potent than edaravone or deferoxamine.Liproxstatin-1 not only inhibited mitochondrial lipid peroxidation,but also restored the expression of GSH,GPX4 and ferroptosis suppressor protein 1.These findings suggest that GPX4 inhibition induces ferroptosis in oligodendrocytes,and that liproxstatin-1 is a potent inhibitor of ferroptosis.Therefore,liproxstatin-1 may be a promising drug for the treatment of central nervous system diseases.Bao-You Fan Yi-Lin Pang Wen-Xiang Li Chen-Xi Zhao Yan Zhang Xu Wang Guang-Zhi Ning Xiao-Hong Kong Chang Liu Xue Yao Shi-Qing Feng 2021Neural Regeneration Research2021,16,3:19
2Laparoscopic vs computerized tomography-guided radiofrequency ablation for large hepatic hemangiomas abutting the diaphragm显示文摘AIM: To compare safety and therapeutic efficacy of laparoscopic radiofrequency(RF) ablation vs computed tomography(CT)-guided RF ablation for large hepatic hemangiomas abutting the diaphragm.METHODS: We retrospectively reviewed our sequential experience of treating 51 large hepatic hemangiomas abutting the diaphragm in 51 patients by CT-guided or laparoscopic RF ablation due to either the presence of symptoms and/or the enlargement of hemangioma.Altogether, 24 hemangiomas were ablated via a CTguided percutaneous approach(CT-guided ablation group), and 27 hemangiomas were treated via a laparoscopic approach(laparoscopic ablation group).RESULTS: The mean diameter of the 51 hemangiomas was 9.6 ± 1.8 cm(range, 6.0-12.0 cm). There was nodifference in the diameter of hemangiomas between the two groups(P > 0.05). RF ablation was performed successfully in all patients. There was no difference in ablation times between groups(P > 0.05). There were 23 thoracic complications in 17 patients: 15(62.5%, 15/24) in the CT-guided ablation group and2(7.4%, 2/27) in the laparoscopic ablation group(P< 0.05). According to the Dindo-Clavien classification,two complications(pleural effusion and diaphragmatic rupture grade Ⅲ) were major in two patients. All others were minor(grade Ⅰ). Both major complications occurred in the CT-guided ablation group. The minor complications were treated successfully with conservative measures, and the two major complications underwent treatment by chest tube drainage and thoracoscopic surgery, respectively. 3Effects of planting patterns on yield, quality, and defoliation in machine-harvested cotton显示文摘The aim of this study was to elucidate the effects of different machine-harvested cotton-planting patterns on defoliation,yield,and fiber quality in cotton and to provide support for improving the quality of machine-harvested cotton.In the 2015 and 2016 growing seasons,the Xinluzao 45(XLZ45)and Xinluzao 62(XLZ62)cultivars,which are primarily cultivated in northern Xinjiang,were used as study materials.Conventional wide-narrow row(WNR),wide and ultra-narrow row(UNR),wide-row spacing with high density(HWR),and wide-row spacing with low density(LWR)planting patterns were used to assess the effects of planting patterns on defoliation,yield,and fiber quality.Compared with WNR,the seed cotton yields were significantly decreased by 2.06–5.48%for UNR and by 2.50–6.99%for LWR,respectively.The main cause of reduced yield was a reduction in bolls per unit area.The variation in HWR yield was–1.07–1.07%with reduced bolls per unit area and increased boll weight,thus demonstrating stable production.In terms of fiber quality indicators,the planting patterns only showed significant effects on the micronaire value,with wide-row spacing patterns showing an increase in the micronaire values.The defoliation and boll-opening results showed that the number of leaves and dried leaves in HWR was the lowest among the four planting patterns.Prior to the application of defoliating agent and before machine-harvesting,the numbers of leaves per individual plant in HWR were decreased by 14.45 and 25.00%on average,respectively,compared with WNR,while the number of leaves per unit area was decreased by 27.44 and 36.21%on average,respectively.The rates of boll-opening and defoliation in HWR were the highest.Specifically,the boll-opening rate before defoliation and machine-harvesting in HWR was 44.54 and 5.94%higher on average than in WNR,while the defoliation rate prior to machine-harvesting was 3.45%higher on average than in WNR.The numbers of ineffective defoliated leaves and leaf trash in HWR were the lowest,decreased by 33.40 and 32.43%,respectively,compared with WNR.In conclusion,the HWR planting pattern is associated with a high and stable yield,does not affect fiber quality,promotes early maturation,and can effectively decrease the amount of leaf trash in machine-picked seed cotton,and thus its use is able to improve the quality of machine-harvested cotton.WANG Fang-yong HAN Huan-yong LIN Hai CHEN Bing KONG Xian-hui NING Xin-zhu WANG Xu-wen YU Yu LIU Jing-de 2019Journal of Integrative Agriculture2019,18,9:14
4Temperature dependent expression of cdc2 and cyclin B1 in spermatogenic cells during spermatogenesis显示文摘p34cdc2 and Cyclin B1 are key components of cell cycle controlling machine and are believed to play a fundamental role in gametogenesis. It is also well known that, in scrotal mammals, spermatogenesis depends greatly on the maintenance of comparatively low temperature in the scrotum. To investigate whether the expression of cdc2 and Cyclin B1 in spermatogenic cells during spermatogenesis is actually a temperature dependent event, in situ hybridization, Western blotting and immunohistochemistry analysis were used to study the expression of cdc2 and Cyclin B1 in normal and cryptorchid testis. Results showed that the abdominal temperature had no significant influence on the transcription of cdc2 and Cyclin B1 in the spermatogonia and pachytene/diplotene primary spermatocytes, but it blocked the translation of them. Due to the deficiency of p34cdc2 and Cyclin B1, the spermatogonia and pachytene/diplotene primary spermatocytes were unable to form MPF, hence, they couldn’t undergo karyokinesis. The development of primary spermatocytes was arrested at the G2 to M phase transition. We also found that testosterone could regulate the Cyclin B1 expression in spermatogenic cells. 5Meningeal lymphatic vessels regulate brain tumor drainage and immunity显示文摘Recent studies have shown that meningeal lymphatic vessels(MLVs),which are located both dorsally and basally beneath the skull,provide a route for draining macromolecules and trafficking immune cells from the central nervous system(CNS)into cervical lymph nodes(CLNs),and thus represent a potential therapeutic target for treating neurodegenerative and neuroinflammatory diseases.However,the roles of MLVs in brain tumor drainage and immunity remain unexplored.Here we show that dorsal MLVs undergo extensive remodeling in mice with intracranial gliomas or metastatic melanomas.RNA-seq analysis of MLV endothelial cells revealed changes in the gene sets involved in lymphatic remodeling,fluid drainage,as well as inflammatory and immunological responses.Disruption of dorsal MLVs alone impaired intratumor fluid drainage and the dissemination of brain tumor cells to deep CLNs(dCLNs).Notably,the dendritic cell(DC)trafficking from intracranial tumor tissues to dCLNs decreased in mice with defective dorsal MLVs,and increased in mice with enhanced dorsal meningeal lymphangiogenesis.Strikingly,disruption of dorsal MLVs alone,without affecting basal MLVs or nasal LVs,significantly reduced the efficacy of combined anti-PD-1/CTLA-4 checkpoint therapy in striatal tumor models.Furthermore,mice bearing tumors overexpressing VEGF-C displayed a better response to anti-PD-1/CTLA-4 combination therapy,and this was abolished by CCL21/CCR7 blockade,suggesting that VEGF-C potentiates checkpoint therapy via the CCL21/CCR7 pathway.Together,the results of our study not only demonstrate the functional aspects of MLVs as classic lymphatic vasculature,but also highlight that they are essential in generating an efficient immune response against brain tumors.Xueting Hu Qiuping Deng Lu Ma Qingqing Li Yidong Chen Yuhan Liao Fan Zhou Chen Zhang Linlin Shao Jun Feng Tubao He Weihai Ning Yan Kong Yingqing Huo Aibin He Bing Liu Jingjing Zhang Ralf Adams Yulong He Fuchou Tang Xiuwu Bian Jincai Luo 2020Cell Research2020,30,3:11

8Neuroprotective effect of deferoxamine on erastin-induced ferroptosis in primary cortical neurons显示文摘The iron chelator deferoxamine has been shown to inhibit ferroptosis in spinal cord injury.However,it is unclear whether deferoxamine directly protects neurons from ferroptotic cell death.By comparing the survival rate and morphology of primary neurons and SH-SY5Y cells exposed to erastin,it was found that these cell types respond differentially to the duration and concentration of erastin treatment.Therefore,we studied the mechanisms of ferroptosis using primary cortical neurons from E16 mouse embryos.After treatment with 50μM erastin for 48 hours,reactive oxygen species levels increased,and the expression of the cystine/glutamate antiporter system light chain and glutathione peroxidase 4 decreased.Pretreatment with deferoxamine for 12 hours inhibited these changes,reduced cell death,and ameliorated cellular morphology.Pretreatment with the apoptosis inhibitor Z-DEVD-FMK or the necroptosis inhibitor necrostain-1 for 12 hours did not protect against erastin-induced ferroptosis.Only deferoxamine protected the primary cortical neurons from ferroptosis induced by erastin,confirming the specificity of the in vitro ferroptosis model.This study was approved by the Animal Ethics Committee at the Institute of Radiation Medicine of the Chinese Academy of Medical Sciences,China(approval No.DWLL-20180913)on September 13,2018.Yan Zhang Bao-You Fan Yi-Lin Pang Wen-Yuan Shen Xu Wang Chen-Xi Zhao Wen-Xiang Li Chang Liu Xiao-Hong Kong Guang-Zhi Ning Shi-Qing Feng Xue Yao 2020Neural Regeneration Research2020,15,8:9
9Long-term postoperative outcomes of bilateral lateral rectus recession vs unilateral recession-resection for intermittent exotropia显示文摘AIM: To discuss the long-term postoperative results of bilateral lateral rectus recession(BLR) and unilateral lateral rectus recession-medial rectus resection(RR) in therapy of intermittent exotropia.METHODS: We retrospectively analyzed 213 cases of intermittent exotropia who underwent surgery between2008 and 2010. The patients were grouped into BLR group and RR group. Motor outcomes were divided into three groups on the basis of the angle of deviation after surgery: overcorrection(esotropia/phoria >5△), orthophoria(esotropia/phoria ≤5△to exotropia/phoria ≤10△), and undercorrection/recurrence(exotropia/phoria >10△).Titmus test was used to evaluate stereoacuity, the stereoacuity <800s of arc meaned the patients had stereopsis. Surgical outcome including motor criteria and sensory status were compared at postoperative 6, 12,24 mo and at 36 mo examination between groups. RESULTS: At 12, 24 mo after surgery, the motor outcomes had no difference(P >0.05) between groups.However, the motor outcomes at 6, 36 mo were signally different in each group, indicating the success rate in RR group at 6mo was higher than that in BLR group(83.02%vs 82.24%, P <0.05) but the result was contrary at the 3y examination(60.75% vs 43.40%, P <0.05). No statistical significance were found in the sensory outcomes between the groups at mean of 3.7y follow-up.CONCLUSION: The motor outcomes in RR group were better than in BLR group at 6mo after surgery, while the3 y outcomes were better in BLR group. 10Expression of interleukin-32 in bone marrow of patients with myeloma and its prognostic significance显示文摘BACKGROUND The guiding effect of prognostic stratification in multiple myeloma(MM) for treatment has been increasingly emphasized in recent years. The stratification of risk factors based on the International Staging System(ISS), Durie-Salmon(DS)staging and related indicators is affected by the renal function of patients,resulting in poor performance. This study assesses the relationship between interleukin-32(IL-32) and related risk factors in 67 patients with MM and their clinical outcomes.AIM To investigate the feasibility of IL-32 in evaluating prognosis in patients with MM and the factors influencing prognosis.METHODS This was a pragmatic, prospective observational study of patients with MM at a single center. According to IL-32 level, patients were divided into two groups.The variables under consideration included age, blood β2-microglobulin,albumin, C-reactive protein, serum calcium, serum creatinine, lactate dehydrogenase, M protein type, ISS stage, DS stage, and IL-32 levels and minimal residual disease(MRD) after induction treatment. The main outcomes were progression-free survival(PFS) and overall survival(OS).RESULTS IL-32 was an important factor affecting PFS and OS in patients with MM.Compared with patients with IL-32 levels ≥ 856.4 pg/m L, patients with IL-32 levels < 856.4 pg/m L had longer PFS(P = 0.0387) and OS(P = 0.0379);Univariate analysis showed that IL-32 level and MRD were significantly associated with OS and PFS(P < 0.05). Multivariate analysis showed that IL-32 levels ≥ 856.4 pg/m L and MRD positive were still independent risk factors for OS and PFS(P < 0.05).CONCLUSION IL-32 is valuable for assessing the prognosis of MM patients. 14Host Interferon-Stimulated Gene 20 Inhibits Pseudorabies Virus Proliferation显示文摘Host interferon-stimulated gene 20(ISG20)exerts antiviral effects on viruses by degrading viral RNA or by enhancing IFN signaling.Here,we examined the role of ISG20 during pseudorabies virus(PRV)proliferation.We found that ISG20 modulates PRV replication by enhancing IFN signaling.Further,ISG20 expression was upregulated following PRV infection and poly(I:C)treatment.Ectopic expression of ISG20 inhibited PRV proliferation in PK15 cells,whereas knockdown of ISG20 promoted PRV proliferation.In addition,ISG20 expression upregulated IFN-βexpression and enhanced IFN downstream signaling during PRV infection.Notably,PRV UL24 suppressed the transcription of ISG20,thus antagonizing its antiviral effect.Further domain mapping analysis showed that the N terminus(amino acids 1-90)of UL24 was responsible for the inhibition of ISG20 transcription.Collectively,these findings characterize the role of ISG20 in suppressing PRV replication and increase the understanding of host-PRV interplay.Xiaoyong Chen Dage Sun Sujie Dong Huanjie Zhai Ning Kong Hao Zheng Wu Tong Guoxin Li Tongling Shan Guangzhi Tong 2021Virologica Sinica2021,36,5:4
15Impact of National Centralized Drug Procurement Policy on Antiviral Utilization and Expenditure for Hepatitis B in China显示文摘Background and Aims:The National Centralized Drug Procurement(NCDP)policy was launched in China's Mainland in April 2019,with entecavir(ETV)and tenofovir disoproxil fumarate(TDF)being included in the procurement list.We conducted the current study to investigate the impact of the NCDP policy on the utilization and expenditures of antiviral therapy for chronic hepatitis B(CHB)in China.Methods:Procurement records,including monthly purchase volume,expenditure,and price of nucleos(t)ide analogs(NAs),were derived from the National Healthcare Security Administration from April 2018 to March 2021.The changes in volumes and expenditures of the first-line NAs and bid-winning products were calculated.The effects of price,volume,and structure related to drug expenditure were calculated by the Addis and Magrini(AM)Index System Analysis.Results:The purchase volume of NAs significantly increased from 134.3 to 318.3 million DDDs,whereas the expenditure sharply decreased from 1,623.41 to 490.43 million renminbi(RMB)or 241.94 to 73.09 million US dollars(USD).The proportions of firstline NAs rose from 72.51%(ETV:69.00%,TDF:3.51%)to 94.97%(ETV:77.42%,TDF:17.55%).AM analysis showed that the NCDP policy decreased the expenditure of all NAs(S=0.91)but increased that of the first-line NAs in the bidwinning list(S=1.13).Assuming the population size of CHB patients remains stable and a compliance rate of≥75%,the proportion of CHB patients receiving first-line antiviral therapy would increase from 6.36–8.48%to 11.56–15.41%.Conclusions:The implementation of the NCDP policy significantly increased the utilization of first-line NAs for CHB patients at a lower expenditure.The findings provided evidence for optimizing antiviral therapy strategy and allocating medical resources in China.Xinyu Zhao Min Li Hao Wang Xiaoqian Xu Xiaoning Wu Yameng Sun Canjian Ning Bingqiong Wang Shuyan Chen Hong You Jidong Jia Yuanyuan Kong 2022Journal of Clinical and Translational Hepatology2022,10,3:4

18Microstructure and properties of a beta-solidifying TiAl-based alloy with different refiners显示文摘This study systematically compared the influences of yttrium(Y),boron(B),and carbon(C) on the microstructural refinement and properties of a Ti-43Al-5Nb alloy.The microstructural refinement effect in the TiAl alloy closely depends on the refiner used.The refinement effects of the three elements on colony size and lamellar thickness can be arranged as B > Y > C and Y > C > B,respectively.Moreover,a microstructure with a small grain size and ultra-fine lamellar spacing can be obtained by adding B and Y or B and C.The mechanical properties of TiAl alloy are also influenced by the refiners.TiAl alloys with proper B and Y contents exhibit favorable hot workability,tensile properties,and fracture toughness,whereas the C-containing alloy displays poor tensile properties and low fracture toughness.These results indicate that Y and B are more suitable microstructure refiners than C.This study may serve as a reference for practical alloying design.Ning Cui Xiao-Peng Wang Fan-Tao Kong Yu-Yong Chen Hai-Tao Zhou 2016Rare Metals2016,35,1:3

20Effects of five heavy metals at sub-lethal concentrations on the growth and photosynthesis of Chlorella vulgaris显示文摘The impact of sub-lethal concentrations(0.05,0.5,5 μmol L 1) of the heavy metals copper(Cu),chromium(Cr),zinc(Zn),cadmium(Cd) and lead(Pb) on the growth and photosynthesis of Chlorella vulgaris was studied during 96 h exposure experiments.The results showed that the effects of these five metals on the growth of Chlorella vulgaris were dependent on both concentration and exposure time.It was found that 5 μmol L 1 treatments of Cu,Cr,Zn,Cd and Pb significantly inhibited the growth of Chlorella vulgaris,and the effect became weaker with an increase in exposure duration.Different effects on chlorophyll fluorescence were found for different metals,with Cu and Cr having an inhibiting effect and Zn and Cd having a promoting effect.The effects of heavy metals upon the growth and photosynthesis of Chlorella vulgaris were independent of each other and not causally related.OUYANG HuiLing KONG XiangZhen HE Wei QIN Ning HE QiShuang WANG Yan WANG Rong XU FuLiu 2012Chinese Science Bulletin2012,57,25:2
