3Liproxstatin-1 is an effective inhibitor of oligodendrocyte ferroptosis induced by inhibition of glutathione peroxidase 4显示文摘Our previous studies showed that ferroptosis plays an important role in the acute and subacute stages of spinal cord injury.High intracellular iron levels and low glutathione levels make oligodendrocytes vulnerable to cell death after central nervous system trauma.In this study,we established an oligodendrocyte(OLN-93 cell line)model of ferroptosis induced by RSL-3,an inhibitor of glutathione peroxidase 4(GPX4).RSL-3 significantly increased intracellular concentrations of reactive oxygen species and malondialdehyde.RSL-3 also inhibited the main antiferroptosis pathway,i.e.,SLC7A11/glutathione/glutathione peroxidase 4(xCT/GSH/GPX4),and downregulated acyl-coenzyme A synthetase long chain family member 4.Furthermore,we evaluated the ability of several compounds to rescue oligodendrocytes from ferroptosis.Liproxstatin-1 was more potent than edaravone or deferoxamine.Liproxstatin-1 not only inhibited mitochondrial lipid peroxidation,but also restored the expression of GSH,GPX4 and ferroptosis suppressor protein 1.These findings suggest that GPX4 inhibition induces ferroptosis in oligodendrocytes,and that liproxstatin-1 is a potent inhibitor of ferroptosis.Therefore,liproxstatin-1 may be a promising drug for the treatment of central nervous system diseases.Bao-You Fan Yi-Lin Pang Wen-Xiang Li Chen-Xi Zhao Yan Zhang Xu Wang Guang-Zhi Ning Xiao-Hong Kong Chang Liu Xue Yao Shi-Qing Feng 2021Neural Regeneration Research2021,16,3:19

5Kanglexin,a novel anthraquinone compound,protects against myocardial ischemic injury in mice by suppressing NLRP3 and pyroptosis显示文摘Pyroptosis is a form of inflammatory cell death that could be driven by the nucleotide-binding oligomerization domain-like receptor family pyrin domain-containing 3(NLRP3)inflammasome activation following myocardial infarction(MI).Emerging evidence suggests the therapeutic potential for ameliorating MI-induced myocardial damages by targeting NLRP3 and pyroptosis.In this study,we investigated the myocardial protection effect of a novel anthraquinone compound(4,5-dihydroxy-7-methyl-9,10-anthraquinone-2-ethyl succinate)named Kanglexin(KLX)in vivo and in vitro.Male C57BL/6 mice were pre-treated either with KLX(20,40 mg·kg^?1per day,intragastric gavage)or vehicle for 7 consecutive days prior to ligation of coronary artery to induce permanent MI.KLX administration dose-dependently reduced myocardial infarct size and lactate dehydrogenase release and improved cardiac function as compared to vehicle-treated mice 24 h after MI.We found that MI triggered NLRP3 inflammasome activation leading to conversion of interleukin-1β(IL-1β)and IL-18 into their active mature forms in the heart,which could expand the infarct size and drive cardiac dysfunction.We also showed that MI induced pyroptosis,as evidenced by increased DNA fragmentation,mitochondrial swelling,and cell membrane rupture,as well as increased levels of pyroptosis-related proteins,including gasdermin D,N-terminal GSDMD,and cleaved caspase-1.All these detrimental alterations were prevented by KLX.In hypoxia-or lipopolysaccharide(LPS)-treated neonatal mouse ventricular cardiomyocytes,we showed that KLX(10μM)decreased the elevated levels of terminal deoxynucleotidyl transferase dUTP nick end labeling-and propidium iodide-positive cells,and pyroptosis-related proteins.We conclude that KLX prevents MI-induced cardiac damages and cardiac dysfunction at least partly through attenuating NLRP3 and subsequent cardiomyocyte pyroptosis,and it is worthy of more rigorous investigations for its potential for alleviating ischemic heart disease.Yu Bian Xin Li Ping Pang Xue-ling Hu Shu-ting Yu Yi-ning Liu Xin Li Ning Wang Jin-hui Wang Wei Xiao Wei-jie Du Bao-feng Yang 2020Acta Pharmacologica Sinica2020,41,3:15

7Subcellular Cd accumulation characteristic in root cell wall of rice cultivars with different sensitivities to Cd stress in soil显示文摘The variations of grain cadmiun(Cd) concentrations, translocation factors(TFs) of Cd from roots to shoots/grains of six rice cultivars, characterized with different Cd-sensitivities in polluted soil were studied, the selected rice cultivars were Xiangzao 17(R1), Jiayu 211(R2), Xiangzao 42(R3), Zhuliangyou 312(R4), Zhuliangyou 611(R5), and Jinyou 463(R6), respectively. The Cd subcellular distribution and Cd binding characteristics on subcellular fractions of rice root cell wall(CW) were further investigated. The results showed that the rice grain Cd contents varied significantly, with a maximum variation of 47.0% among the cultivars, the largest grain Cd content was observed with cultivar R1(Cd-sensitivity cultivar) and the smallest with R5(Cd-tolerance cultivar). The translocation factors of Cd from roots to shoots(TF_(shoot)) and roots to grains(TF_(grain)) varied greatly among the cultivars. In general, the TFgrain of the cultivars followed the order of R1>R2>R3>R4> R6-R5. The Cd concentration(mg kg^(–1) FW) in the fraction of root CW, the fraction of cell wall removing pectin(CW-P) and the fraction of cell wall removing pectin and hemicellulose(CW-P-HC) of the cultivars generally followed the order of CW-P>CW>CWP-HC; the ratios of Cd concentration(mg kg–1 FW) in the fraction of CW-P to that of CW were mostly more than 1.10, while the ratios of Cd concentration in the fraction of CW-P-HC to that of CW were mostly less than 0.60, indicating that Cd was mainly stored in the hemicellulose of the root CW. The ratios of Cd of CW-P-HC to CW generally followed the descending order of R1~R2>R3>R4>R5~R6 for the cultivars, which implied that hemicellulose is probably the main subcellular pool for transferring Cd into rice grain, and it restrains the translocation of Cd from shoot to the grain, especially for the Cd-tolerance cultivars(R5 and R6), the compartmentation of more Cd in hemicellulose in root CW is probably one of the main mechanisms for Cd tolerance of rice cultivars.LIU Bin CHEN Li CHEN Shi-bao LI Ning ZHENG Han JIN Ke PANG Huan-cheng MA Yi-bing 2016Journal of Integrative Agriculture2016,15,9:13

9Association Between Serum Uric Acid and Prevalence of Type 2 Diabetes Diagnosed using HbA1c Criteria Among Chinese Adults in Qingdao,China显示文摘Objective To determine whether elevated serum uric acid(UA)levels are associated with type 2 diabetes diagnosed using Hb A1 c levels among Chinese adults.Methods We conducted two population-based cross-sectional studies in Qingdao in China in 2006 and 2009.A total of 6894(39.4% men)subjects aged 35-74 years were included in the data analysis.Newly diagnosed diabetes was defined as Hb A1 c level of ≥6.5%,and prediabetes was classified as Hb A1 c level between 5.7% and 6.4% according to the International Diabetes Federation criteria.Multivariate logistic regression was employed to assess the association between UA and prevalence of type 2 diabetes defined using Glycated hemoglobin A1c(Hb A1 c levels.Results Subjects with prediabetes had higher UA levels than those with normal glucose tolerance,newly diagnosed diabetes,and known diabetes,with corresponding values of 325.1(82.5)μmol/L,310.9(84.2)μmol/L,291.3(81.7)μmol/L,305.2(83.6)μmol/L,respectively(P<0.001 for all comparisons).Binary logistic regression analysis showed that UA was a possible predictor for the prevalence of type 2 diabetes diagnosed using Hb A1 c levels,and the second quartile of UA levels had a higher odds ratio(OR:4.088;95% CI:2.900-5.765)for Hb A1 c than the other quartiles after adjusting for age,body mass index,sex,marital status,education,income,alcohol consumption,smoking,and cardiometabolic parameters.Conclusion Serum UA is significantly associated with type 2 diabetes diagnosed using Hb A1 c levels,independent of other cardiometabolic parameters.XUE Bai TAN Ji Bin NING Feng SUN Jian Ping ZHANG Ke Yi LIU Li WANG Shao Jie ZHANG Dong Feng QIAO Qing PANG Zeng Chang 2015Biomedical and Environmental Sciences2015,28,12:9
10Neuroprotective effect of deferoxamine on erastin-induced ferroptosis in primary cortical neurons显示文摘The iron chelator deferoxamine has been shown to inhibit ferroptosis in spinal cord injury.However,it is unclear whether deferoxamine directly protects neurons from ferroptotic cell death.By comparing the survival rate and morphology of primary neurons and SH-SY5Y cells exposed to erastin,it was found that these cell types respond differentially to the duration and concentration of erastin treatment.Therefore,we studied the mechanisms of ferroptosis using primary cortical neurons from E16 mouse embryos.After treatment with 50μM erastin for 48 hours,reactive oxygen species levels increased,and the expression of the cystine/glutamate antiporter system light chain and glutathione peroxidase 4 decreased.Pretreatment with deferoxamine for 12 hours inhibited these changes,reduced cell death,and ameliorated cellular morphology.Pretreatment with the apoptosis inhibitor Z-DEVD-FMK or the necroptosis inhibitor necrostain-1 for 12 hours did not protect against erastin-induced ferroptosis.Only deferoxamine protected the primary cortical neurons from ferroptosis induced by erastin,confirming the specificity of the in vitro ferroptosis model.This study was approved by the Animal Ethics Committee at the Institute of Radiation Medicine of the Chinese Academy of Medical Sciences,China(approval No.DWLL-20180913)on September 13,2018.Yan Zhang Bao-You Fan Yi-Lin Pang Wen-Yuan Shen Xu Wang Chen-Xi Zhao Wen-Xiang Li Chang Liu Xiao-Hong Kong Guang-Zhi Ning Shi-Qing Feng Xue Yao 2020Neural Regeneration Research2020,15,8:9
11Synoptic-scale variation of δ^(18)O in summer monsoon rainfall at Lijiang, China显示文摘Based on the daily δ^(18)O data in June―September 2003 at Lijiang and the daily mean NCEP/ NCAR reanalysis data, synoptic-scale variation of δ^(18)O in summer monsoon rainfall was investigated. The 'precipitation amount effect' is obvious for the daily δ^(18)O variation, whereas the 'temperature effect' is insignificant. Alternate occurrences of active phase and break phase of the southwest monsoon probably influence the synoptic-scale δ^(18)O variation prominently. Moreover, the isotopic composition in precipitation during the late monsoon months is presumably influenced significantly by recycling of monsoon precipitation. Both the above factors disturb the 'amount effect' of isotopic variation in the monsoon region. This study also indicates that the synoptic-scale rainfall δ^(18)O variation at Lijiang in summer is domi-nated by the Indian monsoon depression (low pressure) system at large scale. 12Oscillation analysis of a DFIG-based wind farm interfaced with LCC-HVDC显示文摘With the increasing capacity of wind farm, HVDC technology has become a promising transmission scheme for long distance transportation of large-scale wind power. However oscillation caused by this system will have a great influence on the security and stability of power system operation. In this paper, the oscillation of a doubly-fed induction generator(DFIG)-based wind farm interfaced with line commutated converter(LCC) based HVDC is discussed. Low-frequency oscillation and subsynchronous oscillation(SSO) are studied since these two oscillations are the particularly concerned oscillations in the stability study of power system in recent years. The model of a DFIG-based wind farm interconnected with LCC-HVDC is developed. The impact of drive train model's structure and parameters on the oscillation characteristics is analyzed. Eigenvalue and participation factor analysis are used to identify the three main modes, which include controller mode, electromechanical mode, and shaft mode. The effects of DFIG controller's parameters, wind speed and operating conditions of HVDC on those modes are studied. 13Putrescine, spermidine, and spermine play distinct roles in rice salt tolerance显示文摘Polyamines(PAs) play diverse roles in plant growth and development,as well as responses to environmental stimuli.In this study,the effects of PAs on rice salt tolerance were investigated.Salt stress resulted in the alteration of endogenous PAs levels in rice roots and leaves,where spermidine(Spd) and spermine(Spm) contents were increased,and putrescine(Put) content was decreased.RT-qPCR analysis revealed that PAs biosynthesis-related genes ADC1,ODC,and Arginase were significantly downregulated by salt treatment;however,SAMDC transcription was significantly upregulated.Exogenous Spm enhanced rice salt tolerance remarkably;however,exogenous Put and Spd undermined rice salt tolerance.Transgenic rice plants overexpressing SAMDC display a higher ratio of Spm/(Put+Spd) and enhanced salt tolerance.Salt stress also increased polyamine oxidase activities in rice,resulting in elevated reactive oxygen species(ROS) production.Our findings revealed that accumulation of Put and Spd substantially reduced salt tolerance in rice,likely by facilitating ROS production;whereas,conversion of Put and Spd to Spm contributes to rice salt tolerance.Md Azizul ISlam PANG Jin-huan MENG Fan-wei LI Ya-wen XU Ning YANG Chao LIU Jun 2020Journal of Integrative Agriculture2020,19,3:5
14Wavelength-switchable vortex beams based on a polarization-dependent microknot resonator显示文摘We experimentally demonstrated a method of generating continuously wavelength-switchable optical vortex beams(OVBs) in an all-fiber laser. A polarization-dependent microknot resonator(MKR) functions as comb filter and accounts for the narrow linewidth(0.018 nm) of multiwavelength channels. The wavelength interval corresponds to the free spectral range of the MKR. We exploit a fused SMF–FMF(single mode fiber–few mode fiber) mode coupler to obtain broadband mode conversion and successfully achieve multiwavelength switchable OVBs. As far as we know, this is the first report about identical multiwavelength vortex beams with topological charges of 1. It has been verified that each channel of the vortex beams preserves the same orbital angular momentum(OAM) properties through their clear spiral interferograms. 15Preparation and characterization of monodisperse spherical particles of X-ray nano-phosphors based on Gd_2O_2S:Tb显示文摘Monodisperse spherical Gd2O2S:Tb nanoparticles have been prepared using an improved homogeneous precipitation method combined with solid-gas sulfuration technology. The effects of Tb3+-doped concentration on luminescent intensity and color purity of samples were investigated, and the optimal Tb3+-doped concentration was determined. 16An aptamer-based shear horizontal surface acoustic wave biosensor with a CVD-grown single- layered graphene film for high-sensitivity detection of a label-free endotoxin显示文摘The thickness of the sensitive layer has an important influence on the sensitivity of a shear horizontal surface acoustic wave(SH-SAW)biosensor with a delay-line structure and lower layer numbers of graphene produce better sensitivity for biological detection.Therefore,a label-free and highly sensitive SH-SAW biosensor with chemical vapor deposition(CVD-)-grown single-layered graphene(SLG)for endotoxin detection was developed in this study.With this methodology,SH-SAW biosensors were fabricated on a 36°Y-90°X quartz substrate with a base frequency of 246.2MHz,and an effective detection cell was fabricated using acrylic material.To increase the surface hydrophilicity,chitosan was applied to the surface of the SLG film.Additionally,the aptamer was immobilized on the surface of the SLG film by cross-linking with glutaraldehyde.Finally,the sensitivity was verified by endotoxin detection with a linear detection ranging from 0 to 100 ng/mL,and the detection limit(LOD)was as low as 3.53 ng/mL.In addition,the stability of this type of SH-SAW biosensor from the air phase to the liquid phase proved to be excellent and the specificity was tested and verified by detecting the endotoxin obtained from Escherichia coli(E.coli),the endotoxin obtained from Pseudomonas aeruginosa(P.aeruginosa),and aflatoxin.Therefore,this type of SH-SAW biosensor with a CVD-grown SLG film may offer a promising approach to endotoxin detection,and it may have great potential in clinical applications.Junwang Ji Yiquan Pang Dongxiao Li Zheng Huang Zuwei Zhang Ning Xue Yi Xu Xiaojing Mu 2020Microsystems & Nanoengineering2020,6,1:3
17Detailed resume of RNA m^(6)A demethylases显示文摘N6-Methyladenosine(m^(6)A) is the most abundant internal modification in eukaryotic mRNA,playing critical role in various bioprocesses. Like other epigenetic modifications, m^(6)A modification can be catalyzed by the methyltransferase complex and erased dynamically to maintain cells homeostasis. Up to now, only two m^(6)A demethylases have been reported, fat mass and obesity-associated protein(FTO)and alkylation protein AlkB homolog 5(ALKBH5), involving in a wide range of mRNA biological progress, including mRNA shearing, export, metabolism and stability. Furthermore, they participate in many significantly biological signaling pathway, and contribute to the progress and development of cancer along with other diseases. 19Liver resection versus liver transplantation for hepatocellular carcinoma within Milan criteria:a meta-analysis of 18,421 patients显示文摘Background:Outcomes after liver resection(LR)and liver transplantation(LT)for hepatocellular carcinoma(HCC)are heterogenous and may vary by region,over time periods and disease burden.We aimed to compare overall survival(OS)and disease-free survival(DFS)between LT versus LR for HCC within the Milan criteria.Methods:Two authors independently searched Medline and Embase databases for studies comparing survival after LT and LR for patients with HCC meeting the Milan criteria.Meta-analyses and meta-regression were conducted using random-effects models.Results:We screened 2,278 studies and included 35 studies with 18,421 patients.LR was associated with poorer OS[hazard ratio(HR)=1.44;95%confidence interval(CI):1.14-1.81;P<0.01]and DFS(HR=2.71;95%CI:2.23-3.28;P<0.01)compared to LT,with similar findings among intention-to-treat(ITT)studies.In uninodular disease,OS in LR was comparable to LT(P=0.13)but DFS remained poorer(HR=2.95;95%CI:2.30-3.79;P<0.01).By region,LR had poorer OS versus LT in North America and Europe(P≤0.01),but not Asia(P=0.25).LR had inferior survival versus LT in studies completed before 2010(P=0.01),but not after 2010(P=0.12).Cohorts that underwent enhanced surveillance had comparable OS after LT and LR(P=0.33),but cohorts undergoing usual surveillance had worse OS after LR(HR=1.95;95%CI:1.24-3.07;P<0.01).Conclusions:Mortality after LR for HCC is nearly 50%higher compared to LT.Survival between LR and LT were similar in uninodular disease.The risk of recurrence after LR is threefold that of LT.Jin Hean Koh Darren Jun Hao Tan Yuki Ong Wen Hui Lim Cheng Han Ng Phoebe Wen Lin Tay Jie Ning Yong Mark D.Muthiah Eunice X.Tan Ning Qi Pang Beom Kyung Kim Nicholas Syn Alfred Kow Brian K.P.Goh Daniel Q.Huang 2022Hepatobiliary Surgery and Nutrition2022,11,1:3
20Few-shot text classification by leveraging bi-directional attention and cross-class knowledge显示文摘Few-shot text classification targets at the situation where a model is developed to classify newly incoming query instances after acquiring knowledge from a few support instances.In this paper,we investigate few-shot text classification under a metric-based meta-learning framework.While the representations of the query and support instances are the key to the classification,existing study handles them independently in the text encoding stage.To better describe the classification features,we propose to exploit their interaction with adapted bi-directional attention mechanism.Moreover,distinct from previous approaches that encode different classes individually,we leverage the underlying cross-class knowledge for classification.To this end,we conceive the learning target by incorporating the large margin loss,which is expected to shorten the intra-class distances while enlarging the inter-class distances.To validate the design,we conduct extensive experiments on three datasets,and the experimental results demonstrate that our solution outperforms its state-of-the-art competitors.Detailed analyses also reveal that the bi-directional attention and the cross-class knowledge both contribute to the overall performance.Ning PANG Xiang ZHAO Wei WANG Weidong XIAO Deke GUO 2021Science China(Information Sciences)2021,64,3:3
