| 1 | AXL is a candidate receptor for SARS-CoV-2 that promotes infection of pulmonary and bronchial epithelial cells显示文摘The current coronavirus disease 2019(COVID-19)pandemic presents a global public health challenge.The viral pathogen responsible,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),binds to the host receptor ACE2 through its spike(S)glycoprotein,which mediates membrane fusion and viral entry.Although the role of ACE2 as a receptor for SARS-CoV-2 is clear,studies have shown that ACE2 expression is extremely low in various human tissues,especially in the respiratory tract.Thus,other host receptors and/or co-receptors that promote the entry of SARS-CoV-2 into cells of the respiratory system may exist.In this study,we found that the tyrosine-protein kinase receptor UFO(AXL)specifically interacts with the N-terminal domain of SARS-CoV-2 S.Using both a SARS-CoV-2 virus pseudotype and authentic SARS-CoV-2,we found that overexpression of AXL in HEK293T cells promotes SARS-CoV-2 entry as efficiently as overexpression of ACE2,while knocking out AXL significantly reduces SARS-CoV-2 infection in HI 299 pulmonary cells and in human primary lung epithelial cells.Soluble human recombinant AXL blocks SARS-CoV-2 infection in cells expressing high levels of AXL.The AXL expression level is well correlated with SARS-CoV-2 S level in bronchoalveolar lavage fluid cells from COVID-19 patients.Taken together,our findings suggest that AXL is a novel candidate receptor for SARS-CoV-2 which may play an important role in promoting viral infection of the human respiratory system and indicate that it is a potential target for future clinical intervention strategies. | Shuai Wang Zongyang Qiu Yingnan Hou Xiya Deng Wei Xu Tingting Zheng Peihan Wu Shaofang Xie Weixiang Bian Chong Zhang Zewei Sun Kunpeng Liu Chao Shan Aifu Lin Shibo Jiang Youhua Xie Qiang Zhou Lu Lu Jing Huang Xu Li | 2021 | Cell Research2021,31,2: | 39 |
| 2 | Method of Modulation Recognition Based on Combination Algorithm of K-Means Clustering and Grading Training SVM显示文摘For the existing support vector machine, when recognizing more questions, the shortcomings of high computational complexity and low recognition rate under the low SNR are emerged. The characteristic parameter of the signal is extracted and optimized by using a clustering algorithm, support vector machine is trained by grading algorithm so as to enhance the rate of convergence, improve the performance of recognition under the low SNR and realize modulation recognition of the signal based on the modulation system of the constellation diagram in this paper. Simulation results show that the average recognition rate based on this algorithm is enhanced over 30% compared with methods that adopting clustering algorithm or support vector machine respectively under the low SNR. | 4 | Rapamycin reverses ferroptosis by increasing autophagy in MPTP/MPP+-induced models of Parkinson's disease显示文摘Parkinson’s disease is a neurodegenerative disorder,and fe rroptosis plays a significant role in the pathological mechanism underlying Parkinson’s disease.Rapamycin,an autophagy inducer,has been shown to have neuroprotective effects in Parkinson’s disease.However,the link between rapamycin and ferroptosis in Parkinson’s disease is not entirely clear.In this study,rapamycin was administe red to a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-induced Parkinson’s disease mouse model and a 1-methyl-4-phenylpyridinium-induced Parkinson’s disease PC12 cell model.The results showed that rapamycin improved the behavioral symptoms of Parkinson’s disease model mice,reduced the loss of dopamine neurons in the substantia nigra pars compacta,and reduced the expression of ferroptosis-related indicators(glutathione peroxidase 4,recombinant solute carrier family 7,member 11,glutathione,malondialdehyde,and reactive oxygen species).In the Parkinson’s disease cell model,rapamycin improved cell viability and reduced ferro ptosis.The neuroprotective effect of rapamycin was attenuated by a ferroptosis inducer(methyl(1S,3R)-2-(2-chloroacetyl)-1-(4-methoxycarbonylphenyl)-1,3,4,9-tetrahyyridoindole-3-carboxylate)and an autophagy inhibitor(3-methyladenine).Inhibiting ferro ptosis by activating autophagy may be an important mechanism by which rapamycin exerts its neuroprotective effects.Therefo re,the regulation of ferroptosis and autophagy may provide a therapeutic target for drug treatments in Parkinson’s disease. | Tongyu Liu Peihan Wang Huan Yin Xiangfei Wang Jing Lv Jiang Yuan Jing Zhu Yunfu Wang | 2023 | Neural Regeneration Research2023,18,11: | 1 |
| 5 | Three Novel Avastroviruses Identified in Dead Wild Crows显示文摘Dear Editor,Astroviruses(AstVs)are non-enveloped,single-stranded RNA viruses with a diameter of approximately 35 nm(Madeley and Cosgrove 1975).The genomes of AstVs range in size from 6.8 to 7.9 kb(De Benedictis et al.2011),consisting of a 5'-untranslated region(UTR),three open reading frames(ORFs),a 3'-UTR,and a poly-adenylated(polyA)tail(Pantin-Jackwood et al.2006).In 1975,AstVs were first discovered in faeces of infants(Madeley and Cosgrove 1975).Until now,a variety of animal species,including cows,sheep,pigs,dogs,deer,rats,bats,chickens,ducks,turkeys,sea lions and seals have been reported to be infected with AstVs(Tzipori et al.1981;Cattoli et al.2007;Chu et al.2008,2010;Atkins et al.2009;Toffan et al.2009;Rivera et al.2010;Guix et al.2013;Kang et al.2012;Reuter et al.2012a,2012b;Li et al.2021).Currently,AstVs comprise two genera,Mamastrovirus(MAstV)and Avastrovirus(AAstV).Herein,we describe three novel AAstVs identified from dead crow tissues.To our knowledge,this is the first report for identification of AstVs in crows. | Chunge Zhang Yongchun Yang Tao Hu Hong Zhou Cheng Zhang Jian Cao Juan Li Peihan Wang Gary Wong Xiaodu Wang Houhui Song George FGao Weifeng Shi Yuhai Bi | 2021 | Virologica Sinica2021,36,6: | 1 |
