| 3 | Selective killing of K-ras-transformed pancreatic cancer cells by targeting NAD(P)H oxidase显示文摘Introduction:Oncogenic aaivation of the K-ras gene occurs in >90%of pancreatic ductal carcinoma and plays a critical role in the pathogenesis of this malignancy.Increase of reactive oxygen species(ROS) has also been observed in a wide spectrum of cancers.This study aimed to investigate the mechanistic association between K-ras-induced transformation and increased ROS stress and its therapeutic implications in pancreatic cancer.Methods:ROS level,NADPH oxidase(NOX) aaivity and expression,and cell invasion were examined in human pancreatic dua epithelial E6E7 cells transfeaed with K-ras^(G12V) compared with parental E6E7 cells.The cytotoxic effea and antitumor effect of capsaicin,a NOX inhibitor,were also tested in vitro and in vivo.Results:K-ras transfeaion caused activation of the membrane-associated redox enzyme NOX and elevated ROS generation through the phosphatidylinositol 3'-kinase(PI3K) pathway.Importantly,capsaicin preferentially inhibited the enzyme aaivity of NOX and induced severe ROS accumulation in K-ras-transformed cells compared with parental E6E7 cells.Furthermore,capsaicin effeaively inhibited cell proliferation,prevented invasiveness of /(-ras-transformed pancreatic cancer cells,and caused minimum toxicity to parental E6E7 cells.In vivo,capsaicin exhibited antitumor aaivity against pancreatic cancer and showed oxidative damage to the xenograft tumor cells.Conclusions:K-ras oncogenic signaling causes increased ROS stress through NOX,and abnormal ROS stress can selectively kill tumor cells by using NOX inhibitors.Our study provides a basis for developing a novel therapeutic strategy to effectively kill K-ras-transformed cells through a redox-mediated mechanism. | Peng Wang Yi-Chen Sun Wen-Hua Lu Peng Huang Yumin Hu | 2015 | Chinese Journal of Cancer2015,34,4: | 5 |
| 4 | Evolution of microstructures and optical properties of gadolinium oxide with oxygen flow rate and annealing temperature显示文摘In this study, the effects of oxygen flow rate and annealing temperature on Gd_2 O_3 structures and optical properties were systematically analyzed. Gd_2 O_3 films were deposited on both quartz and ZnS substrates by magnetron sputtering and then annealed under vacuum at 700, 800 and 900℃, Restructure and phase transformation from cubic to monoclinic occur at different temperatures depending on the oxygen flow rate. The optical band gap, which is more sensitive to the annealing temperature than oxygen flow rate changes from 5.32 to 5.65 eV. The refractive index is approximately 1.75 at 550 nm and is adjustable by the oxygen flow rate. The transmittance of the ZnS substrate with Gd_2 O_3 film exceeds 80% and reaches82% at the 7.5-9.5 μm range. When ZnS is coated on both sides, the transmittance is increased to approximately 90%. | 6 | Metabolic reprogramming and redox adaptation in sorafenib-resistant leukemia cells:detected by untargeted metabolomics and stable isotope tracing analysis显示文摘Background:Internal tandem duplications(ITD)within the juxtamembrane domain of FMS-like tyrosine kinase 3(FLT3)represent a poor prognostic indicator in acute myeloid leukemia(AML).Therapeutic benefits of tyrosine kinase inhibitors,such as sorafenib,are limited due to the emergence of drug resistance.While investigations have been con-ducted to improve the understanding of the molecular mechanisms underlying the resistance to this FLT3 inhibitor,a profile of cell functioning at the metabolite level and crosstalk between metabolic pathways has yet to be created.This study aimed to elucidate the alteration of metabolomic profile of leukemia cells resistant to the FLT3 inhibitor.Methods:We established two sorafenib-resistant cell lines carrying FLT3/ITD mutations,namely the murine BaF3/ITD-R and the human MV4-11-R cell lines.We performed a global untargeted metabolomics and stable isotope-labe-ling mass spectrometry analysis to identify the metabolic alterations relevant to the therapeutic resistance.Results:The resistant cells displayed fundamentally rewired metabolic profiles,characterized by a higher demand for glucose,accompanied by a reduction in glucose flux into the pentose phosphate pathway(PPP);and by an increase in oxidative stress,accompanied by an enhanced glutathione synthesis.We demonstrated that the highest scoring network of altered metabolites in resistant cells was related to nucleotide degradation.A stable isotope tracing experiment was performed and the results indicated a decrease in the quantity of glucose entering the PPP in resistant cells.Further experiment suggested that the inhibition of major enzymes in the PPP consist of glucose-6-phosphate dehydrogenase deficiency(G6PD)in the oxidative arm and transketolase(TKT)in the non-oxidative arm.In addition,we observed that chronic treatment with sorafenib resulted in an increased oxidative stress in FLT3/ITD-positive leu-kemia cells,which was accompanied by decreased cell proliferation and an enhanced antioxidant response.Conclusions:Our data regarding comparative metabolomics characterized a distinct metabolic and redox adaptation that may contribute to sorafenib resistance in FLT3/ITD-mutated leukemia cells. | Xin You Weiye Jiang Wenhua Lu Hui Zhang Tiantian Yu Jingyu Tian Shijun Wen Guillermo Garcia-Manero Peng Huang Yumin Hu | 2019 | Cancer Communications2019,39,1: | 1 |

| 8 | The interpolating element-free Galerkin method for elastic large deformation problems显示文摘This paper presents an interpolating element-free Galerkin(IEFG) method for solving the two-dimensional(2D) elastic large deformation problems. By using the improved interpolating moving least-squares method to form shape function, and using the Galerkin weak form of 2D elastic large deformation problems to obtain the discrete equations, we obtain the formulae of the IEFG method for 2D elastic large deformation problems. As the displacement boundary conditions can be applied directly, the IEFG method can acquire higher computational efficiency and accuracy than the traditional element-free Galerkin(EFG)method, which is based on the moving least-squares approximation and can not apply the displacement boundary conditions directly. To analyze the influences of node distribution, scale parameter of influence domain and the loading step on the numerical solutions of the IEFG method, three numerical examples are proposed. | 13 | Generation of Calhm1 knockout mouse and characterization of calhm1 gene expression显示文摘Alzheimer’s disease(AD)is the most common neurode-generative disease among elderly people worldwide.Several genes have been validated to be associated with AD,and calcium homeostasis modulator 1(Calhm1)is the latest suspected one.To investigate the biological and pathological function of Calhm1 systematically,we generated a Calhm1 conventional knockout mouse.However,both the male and female of elderly Calhm1 knockout(KO)mice showed similar ability to their wild type littermates in spatial learning and memory retrieving.Surprisingly,we found that Calhm1 mRNA could not be detected in mouse brains at different ages,although it is expressed in the human brain tissues.We further found that CpG islands(CGIs)of both mouse and human Calhm1 were hypermethylated,whereas CGI of mouse Calhm2 was hypomethylated.In addition,transcriptional active marker H3K4Di occupied on promoters of human Calhm1 and mouse Calhm2 at a considerable level in brain tissues,while the occupancy of H3K4Di on pro-moter of mouse Calhm1 was rare.In sum,we found that mouse Calhm1 was of rare abundance in brain tissues.So it might not be suitable to utilize the knockout murine model to explore biological function of Calhm1 in the pathogenesis of AD. | Junbing Wu Shengyi Peng Rong Wu Yumin Hao Guangju Ji Zengqiang Yuan | 2012 | Protein & Cell2012,3,6: | 1 |

| 17 | Value Evaluation Method for Pumped Storage in the New Power System显示文摘When integrating the generation of large-scale renewable energy,such as wind and solar energy,the supply and demand sides of the new power system will exhibit high uncertainty.Pumped storage power stations can improve flexible resource supply regulation in the power system,which is the key support and important guarantee for building low-carbon,safe,and efficient new power systems.Limited by the current operation mode and electricity price mechanism,the pumped storage power station cost cannot be effectively recovered,and the value cannot be reasonably compensated,resulting in difficult return on investment,single investment subjects,and notable industry development difficulties.According to the operational requirements of the new power system,combined with the various functions of pumped storage power stations,the value of pumped storage power stations in the new power system was analyzed.Based on the equivalent value substitution principle and system operation simulation,a pumped storage value evaluation method for the new power system was proposed.The new power system operation was simulated considering the dispatching model of wind and photovoltaic power abandonment penalties.Under the same dispatching objectives,the output of various power sources and power generation operating costs with and without pumped storage power stations in the system were compared.From economic,safety,social,and environmental benefit perspectives,a quantitative model of the pumped storage power station value was established,covering seven dimensions:asset investment savings,power generation operating cost reduction,flexible adjustment capability improvement,system resilience enhancement,power outage loss reduction,renewable energy consumption,and emission reduction promotion.Based on the new power system operation and planning data for southern China,the value of typical pumped storage power stations was analyzed,and the results showed that with new power system’s construction and development,the value of pumped storage power stations is increasing,and the value structure is closely related to power grid characteristics.This value evaluation method could provide references for pumped storage investment decisions,subsidy policies,and price mechanisms to fully utilize the role of pumped storage power stations and promote high-quality development of new power systems. | Yumin Peng Ying Yang Man Chen Xuelin Wang Yijun Xiong Mu Wang Yikai Li Bushi Zhao | 2023 | Chinese Journal of Electrical Engineering2023,9,3: | 0 |
| 18 | Effect of thickness on infrared optical property of VO2 film deposited by magnetron sputtering显示文摘A series of VO2 films with different thicknesses(from about 25 nm to 250 nm)were prepared on sapphire substrates by radio frequency magnetron sputtering.The deposition times varied from 15 min to 150 min.The metal to insulator transitions(MIT)of the films were studied.The optical transmittance of the films to infrared light(with a wavelength of 4.0μm)at room temperature(30℃)varied significantly with film thickness,ranging from 86.53%to 41.01%.The modulation property also changes with thickness,decreasing from 22.89%to 14.74%.The phase transition temperature remained approximately 70℃during heating,and approximately 53℃during cooling,despite a tenfold increase in the deposition time,and the resulting thickness of the films.Raman spectroscopy of the films indicated that the intensities of the characteristic peaks corresponding to V2O5 increase with the increasing of film thickness.Temperature-dependent Raman spectroscopy indicated that the peaks corresponding to VO2 undergo reversible changes during heating and cooling of the films,while the peaks corresponding to V2O5 remain unchanged throughout.Careful control of the V2O5 content of the films(by varying the duration of the deposition process)allows control over their transmittance and optical modulation properties without changes in the phase transition temperature.This provides a new method of controlling the optical properties of these materials and shows promise for their potential applications in thermochromic windows. | YANG ZhenHuai YANG QiuLing YANG Lei DAI Bing XIA Fei WANG Peng GUO Shuai GAO Gang XU LiangGe ZHANG YuMin ZHU JiaQi | 2020 | Science China(Technological Sciences)2020,63,8: | 0 |
