| 1 | Neutrophil extracellular traps mediate the crosstalk between glioma progression and the tumor microenvironment via the HMGB1/RAGE/IL-8 axis显示文摘Objective:Neutrophil extracellular traps(NETs)produced by tumor-infiltrating neutrophils(TINs)are associated with poor prognosis in patients with several types of cancer.However,the mechanisms underlying the involvement of NETs in glioma progression remain largely unknown.This study aimed to elucidate the roles of NETs in biological processes that drive the crosstalk between glioma progression and the tumor microenvironment.Methods:Neutrophil infiltration and NETs formation were investigated in glioma tissue through immunohistochemistry,and their relationships with clinicopathological features and outcomes were statistically evaluated.The effects of NETs on glioma cell progression were studied in a co-culture system.In vivo and in vitro experiments validated the reactive oxygen species activity and cytokine production of TINs,as well as the ERK signaling pathway activation and the metastasis of gliomas.Results:Neutrophil infiltration and NETs formation were induced in high-grade glioma compared with low-grade glioma.NETs induced by TINs were determined to be an oncogenic marker of high-grade gliomas and to be involved in cell proliferation and invasion.NETs overproduction promoted glioma cell proliferation,migration,and invasion.Furthermore,HMGB1 was found to bind to RAGE and activate the NF-κB signaling pathway in vitro.In addition,NETs stimulated the NF-κB signaling pathway,thus promoting IL-8 secretion in glioblastoma.Subsequently,IL-8 recruited neutrophils which in turn mediated NETs formation via the PI3 K/AKT/ROS axis in TINs.Conclusions:Our results suggest that NETs produced by TINs mediate the crosstalk between glioma progression and the tumor microenvironment by regulating the HMGB1/RAGE/IL-8 axis.Targeting NETs formation or IL-8 secretion may be an effective approach to inhibit glioma progression. | Caijun Zha Xiangqi Meng Lulu Li Shan Mi Da Qian Ziwei Li Pengfei Wu Shaoshan Hu Shihong Zhao Jinquan Cai Yanhong Liu | 2020 | Cancer Biology & Medicine2020,17,1: | 19 |
| 2 | A social computing method for energy safety显示文摘Information and communication technologies enable the transformation of traditional energy systems into cyber-physical energy systems(CPESs),but such systems have also become popular targets of cyberattacks.Currently,available methods for evaluating the impacts of cyberattacks suffer from limited resilience,efficacy,and practical value.To mitigate their potentially disastrous consequences,this study suggests a two-stage,discrepancy-based optimization approach that considers both preparatory actions and response measures,integrating concepts from social computing.The proposed Kullback-Leibler divergence-based,distributionally robust optimization(KDR)method has a hierarchical,two-stage objective function that incorporates the operating costs of both system infrastructures(e.g.,energy resources,reserve capacity)and real-time response measures(e.g.,load shedding,demand-side management,electric vehicle charging station management).By incorporating social computing principles,the optimization framework can also capture the social behavior and interactions of energy consumers in response to cyberattacks.The preparatory stage entails day-ahead operational decisions,leveraging insights from social computing to model and predict the behaviors of individuals and communities affected by potential cyberattacks.The mitigation stage generates responses designed to contain the consequences of the attack by directing and optimizing energy use from the demand side,taking into account the social context and preferences of energy consumers,to ensure resilient,economically efficient CPES operations.Our method can determine optimal schemes in both stages,accounting for the social dimensions of the problem.An original disaster mitigation model uses an abstract formulation to develop a risk-neutral model that characterizes cyberattacks through KDR,incorporating social computing techniques to enhance the understanding and response to cyber threats.This approach can mitigate the impacts more effectively than several existing methods,even with limited data availability.To extend this risk-neutral model,we incorporate conditional value at risk as an essential risk measure,capturing the uncertainty and diverse impact scenarios arising from social computing factors.The empirical results affirm that the KDR method,which is enriched with social computing considerations,produces resilient,economically efficient solutions for managing the impacts of cyberattacks on a CPES.By integrating social computing principles into the optimization framework,it becomes possible to better anticipate and address the social and behavioral aspects associated with cyberattacks on CPESs,ultimately improving the overall resilience and effectiveness of the system’s response measures. | Pengfei Zhao Shuangqi Li Zhidong Cao Paul Jen-Hwa Hu Daniel Dajun Zeng Da Xie Yichen Shen Jiangfeng Li Tianyi Luo | 2024 | Journal of Safety Science and Resilience2024,5,1: | 0 |
| 3 | Sulfation of chondroitin and bile acids converges to antagonize Wnt/β-catenin signaling and inhibit APC deficiency-induced gut tumorigenesis显示文摘Sulfation is a crucial and prevalent conjugation reaction involved in cellular processes and mammalian physiology.3’-Phosphoadenosine 5’-phosphosulfate(PAPS)synthase 2(PAPSS2)is the primary enzyme to generate the universal sulfonate donor PAPS.The involvement of PAPSS2-mediated sulfation in adenomatous polyposis coli(APC)mutation-promoted colonic carcinogenesis has not been reported.Here,we showed that the expression of PAPSS2 was decreased in human colon tumors along with cancer stages,and the lower expression of PAPSS2 was correlated with poor prognosis in advanced colon cancer.Gut epithelial-specific heterozygous Apc deficient and Papss2-knockout(Apc^(Δgut-Het)Papss2^(Δgut))mice were created,and the phenotypes were compared to the spontaneous intestinal tumorigenesis of Apc^(Δgut-Het)mice.Apc^(Δgut-Het)Papss2^(Δgutmice) were more sensitive to gut tumorigenesis,which was mechanistically accounted for by the activation of Wnt/β-catenin signaling pathway due to the suppression of chondroitin sulfation and inhibition of the farnesoid X receptor(FXR)-transducin-like enhancer of split 3(TLE3)gene regulatory axis.Chondroitin sulfate supplementation in Apc^(Δgut-Het)Papss2^(Δgutmice) alleviated intestinal tumorigenesis.In summary,we have uncovered the protective role of PAPSS2-mediated chondroitin sulfation and bile acids-FXR-TLE3 activation in the prevention of gut carcinogenesis via the antagonization of Wnt/β-catenin signaling.Chondroitin sulfate may be explored as a therapeutic agent for Papss2 deficiency-associated colonic carcinogenesis. | Pengfei Xu Yue Xi Jong-Won Kim Junjie Zhu Min Zhang Meishu Xu Songrong Ren Da Yang Xiaochao Ma Wen Xie | 2024 | Acta Pharmaceutica Sinica B2024,14,3: | 0 |
