| 1 | PCN-Fe(Ⅲ)-PTX nanoparticles for MRI guided high efficiency chemo-photodynamic therapy in pancreatic cancer through alleviating tumor hypoxia显示文摘As nanomedicine-based clinical strategies have continued to develop,the possibility of combining chemotherapy and singlet oxygen-dependent photodynamic therapy(PDT)to treat pancreatic cancer(PaC)has emerged as a viable therapeutic modality.The efficacy of such an approach,however,is likely to be constrained by the mechanisms of drug release and tumor oxygen levels.In the present study,we developed an Fe(Ⅲ)-complexed porous coordination network(PCN)which we then used to encapsulate PTX(PCN-Fe(Ⅲ)-PTX)nanoparticles(NPs)in order to treat PaC via a combination of chemotherapy and PDT.The resultant NPs were able to release drug in response to both laser irradiation and pH changes to promote drug accumulation within tumors.Furthermore,through a Fe(Ⅲ)-based Fenton-like reaction these NPs were able to convert H2O2 in the tumor site to O2,thereby regulating local hypoxic conditions and enhancing the efficacy of PDT approaches.Also these NPs were suitable for use as a T1-MRI weighted contrast agent,making them viable for monitoring therapeutic efficacy upon treatment.Our results in both cell line and animal models of PaC suggest that these NPs represent an ideal agent for mediating effective MRI-guided chemotherapy-PDT,giving them great promise for the clinical treatment of PaC. | Tao Zhang Zhenqi Jiang Libin Chen Chunshu Pan Shan Sun Chuang Liu Zihou Li Wenzhi Ren Aiguo Wu Pintong Huang | 2020 | Nano Research2020,13,1: | 4 |

| 20 | An experimental research into endostatin microbubble combined with focused ultrasound for anti-tumor angiogenesis in colon cancer显示文摘Objective:to evaluate the therapeutic effect of targeted endostatin-loaded microbubbles,combined with improved,focused,directional ultrasound radiation for inhibition of subcutaneous translocation in situ colon tumor angiogenesis in colon cancer.Methods:65 BALB/c nude mice with subcutaneous translocation in situ colon tumors were randomly divided into five groups.Group A was the control group,without any treatments.In Group B,the mouse was treated with ultrasonic radiation.In Group C,the mouse was treated with ultrasonic radiation combined with empty SonoVue microbubbles.In Group D,the mouse was treated with ultrasonic radiation combined with empty Targestar-SA microbubbles.In Group E,the mouse was treated with ultrasonic radiation combined with endostatin microbubbles.The tumor size was measured before and 1,14,and 28 days after irradiation.The peak intensity(PI),regional blood volume(RBV)and regional blood flow(RBF)were recorded using contrast-enhanced ultrasound.The tumor tissue was removed for pathological examination;the tumor necrosis area and microvascular density(MVD)were evaluated by immunohistochemistry.Results:Tumors in Groups C,D and E were significantly smaller than in Groups A and B at 28 days after irradiation,with Group E the smallest.PI,RBF and RBV of Groups C,D,and E were significantly decreased 28 days after radiation with Group E the lowest,and significantly lower than Groups A and B(all P<0.05).The tumor tissue necrosis area of Group E was clearly greater while MVD was obviously lower than the other groups(all P<0.01)at 28 days after treatment.Conclusion:The targeted endostatin microbubbles,combined with focused,directional ultrasound radiation can damage tumor microvasculature of subcutaneous colon translocation in situ colon cancer,as well as inhibit the tumor angiogenesis. | Xiufeng Zhang Guangen Yang Ying Zhang Pintong Huang Jianming Qiu Yu Sun Zhong Shen Xiujun Liao Hongsheng Xia Shuxian Shao Dong Wang | 2014 | Gastroenterology Report2014,2,1: | 0 |
